This study covers a modified semi-quantitative approach for liquid chromatography mass spectrometry (LC-MS/MS) signature peptide detection for body fluid identification. Peptide concentrations were measured based on synthetic peptide standards. Samples were processed with varying trypsin digestion and purification protocols, including a three-hour trypsin digestion and Microcon membrane filtration. The Microcon filtration method generates polymerase chain reaction (PCR) compatible DNA and peptide fractions that can be analyzed without any further purification. Preliminary validation tests covered stains on different substrates, semen/ saliva mixtures, minimum sample volume, and repeatability. Conventional STR typing, commonly used in forensics for human identification, poses a problem in criminal cases and paternity disputes involving monozygotic (MZ) twins because they share identical DNA sequences. To date, no routine method is available in forensics to differentiate between individuals of MZ pairs. Recently, epigenetic methods measuring differential DNA methylation patterns have been applied to MZ twin differentiation. Environmental factors, such as heavy metal exposures, have been suggested to have an impact not only on neurodegenerative disease, such as Parkinson’s disease, but also on psychostimulants abuse and their toxicity. In this study, two questions were addressed: 1) effects of mercuric chloride on parkinsonian toxicant 1-methyl-4-phenylpyridinium (MPP+) induced cytotoxicity and 2) effects of mercuric chloride on cell surface dopamine transporter. Pheochromocytorma cells (PC12) were treated with various concentrations of mercuric chloride (0.02~2.0 ppm) for 4 hours with and without 0.1 mM MPP+. The main focus of this project was to investigate the protein and DNA components in both sebaceous and eccrine fingerprints. This study investigated the relative content of DNA and proteins in eccrine fingerprints to sebaceous fingerprints. All volunteers were instructed to wash and dry their hands prior to depositing parallel thumbprints. Twenty volunteers were instructed to touch their face to produce sebaceous prints, and 5 volunteers were instructed to wear gloves over a heat source to produce sweaty or eccrine prints. Adult temporary tattoos have become more popular, yet there is very little research on them, especially regarding their use in forensics. This poses a problem because forensic investigators may not be able to distinguish a temporary tattoo from a real tattoo, possibly hindering their investigation. In this study, various types of adult temporary tattoos were examined and analyzed in attempts to identify and distinguish them, both from each other and from permanent tattoos. The present study investigated the effects of varying witness testimony on mock jurors’ perceptions of a case where a woman utilizes self-defense as a reason for killing her husband during a domestic dispute. Using the equation of an ellipse in spherical coordinates, an EXCEL™ (Microsoft Inc., Seattle, WA) spreadsheet was developed that generated tables of data that provide a foundation for a new and more efficient method in fiber analysis. Forensic analysis of DNA from non-human bones can be important in investigating a variety of forensic cases. However, decomposed bone is difficult to process for isolating DNA. In this study, a previously established enzymatic method was utilized to process bone samples that simulate decomposed specimens. Zolpidem is a sedative hypnotic whose adverse effects make it a common drug found in drug facilitated crimes (DFC). In order to provide indelible examples for training and help avoid identification errors, 3D topography surface maps and statistical methods of pattern recognition are applied to toolmarks on bullets containing known subclass characteristics.
