This project serves to provide additional information on the prevalence of non-self DNA on the exposed skin locations specifically the inner upper arm, the nape area of the neck and the neck area just below the ear, where current literature is lacking. Skin tape lifts from these locations were collected from 15 male participants and 13 female participants once a week over a period of three weeks. Only single source profiles were obtained from six volunteers. Female blow flies will lay eggs (colonize) on remains often within minutes of death and entomological methods are based on their behavior and development to extrapolate postmortem intervals (PMI). This project suggests improvements to the treatment methods utilized for the sample preparation and evaluation of minerals in soil samples encountered in forensic science casework and research. The evaluation of pre-treated and post-treated soil samples were performed using Fourier-Transform Infrared Spectroscopy with Attenuated Total Reflection (FTIR-ATR) and Scanning Electron Microscopy-Energy Dispersive Spectroscopy (SEM-EDS). Because soil samples contain a significant amount of organic matter and water which interfere with examination of these minerals while using these instruments, pretreatment methods are necessary to isolate the mineral fraction for examination and analysis.

The water-induced emission of mercury from soil has been studied, but a complete understanding of the mechanisms driving mercury reduction and emission in soil has not been reached. The current understanding is an immediate expulsion of soil gaseous mercury upon water treatment and an upward transport of dissolved soil ionic mercury as the water evaporates. An opioid abuse crisis in recent years has contributed to many overdoses and overdose-related deaths in the United States. In 2017, fentanyl and fentanyl analogs contributed to more than half of NYC’s overdose deaths in 2017. Carfentanyl – an extremely dangerous fentanyl analogue 100 times more potent than fentanyl – is very difficult to detect in drug samples because it is typically present in concentrations lower than analytical instruments can detect at standard settings. The shedding propensity of a person can assist data interpretation in casework when assessing the possibility of passive transfer for DNA analysis. Past studies on shedding propensity evaluated palmar skin (washed and unwashed) deposits. This study compared different skin locations with respect to shedding propensity, and explored the potential of tape-lifts as a skin surface collection method. Contact traces are an important part of DNA casework, but the probative value of any identified associations depends on the possibility of passive transfer. There is known individual variation in DNA left behind during contact, this DNA shedding propensity has an effect on whose DNA is detected. This study evaluated this variability using a cell staining approach. Volunteers were asked to deposit a fingerprint on a clean glass slide, then wash their hands and deposit a second fingerprint after a 30-minute wait without touching anything. Decomposed human remains are complex forensic puzzles, escalating in difficulty as the remains’ age obscures evidence, like trauma. Research has shown that scanning electron microscopes with energy dispersive X-ray spectrometers (SEM-EDX) are capable of detecting and identifying gunshot residue (GSR) particles on bones. However, SEM-EDX work is time consuming, expensive, and not accessible to every forensic department. In this research, bovine leg bones were burned, both with and without meat intact, in order to determine if it was possible to extract and amplify DNA to obtain profiles. The meat was burned over an open flame and the DNA was extracted using the Qiagen DNeasy Blood & Tissue Mini Kit, quantified with the NanoDrop, and amplified using the Bovine Genotypes Panel 3.1 Kit, and separated using the 3500 Genetic Analyzer. Cocaine overdose deaths are on the rise in the United States. This increase may be due to the co-consumption of cocaine with other drugs. When cocaine is ingested in conjunction with alcohol, a specific metabolite called cocaethylene forms. Information regarding cocaethylene, specifically its interpretive value in post-mortem cases, has not been widely studied. Data from the Office of the Chief Medical Examiner in New York City revealed that there were 195 cases in 2017 positive for cocaethylene. These cases were most likely to involve black (42.1%) males (79.5%) in Manhattan (28.2%) aged 50 - 59 (34.4%). Most of the cases …

Methamphetamine (N-methylamphetamine) is a central nervous system stimulant (CNS) and sympathomimetic drug with a high addiction potential. In the United States, there has been a significant increase in the presence of methamphetamine in recent years, specifically in the Northeastern region of the country. The purpose of the current study was to determine the clinical utility of the Ohio State University Traumatic Brain Injury Identification Method (OSU-TBI-ID) in a criminal justice sample, by evaluating the criterion-related validity of this instrument. It was hypothesized that this tool could differentiate between incarcerated individuals with or without a history of Traumatic Brain Injury (TBI) on measures evaluating important TBI-related sequalae. Shooter distance is most often estimated using the Modified Griess test, which can be used to visualize gunshot residue (GSR) patterns around bullet holes. In this experiment, a method of calculating ballistic coefficients of .22LR (long rifle) caliber ammunition was developed using Doppler radar to track the pre-impact and post-impact velocities of projectiles passing through gypsum board. The method demonstrated has distinct advantages in accuracy and flexibility over conventional chronograph projectile tracking that forensic firearm experts should consider when determining the optimal method to obtain a ballistic coefficient (BC). Effective DNA extraction methods are important for forensic applications. The main goal of this experiment was to determine if a newly developed trypsin based protein/DNA co-extraction method applied to contact traces would yield comparable results to a commercial Proteinase K method (QIAamp DNA Investigator Kit) and a Chelex extraction method with Tween 20 used in forensic laboratories. This was tested on 20 sets of sebaceous fingerprints on glass slides.

A previously developed, non-destructive, homemade vacuum method for collecting biological material from handwritten documents had promising DNA results for white copy paper, preserved indented writing, and latent prints. Prior to casework implementation, additional validation experiments are warranted and here the method was tested for different paper substrates. This work describes testing of notebook paper, bank deposit slips, magazine pages, and manila envelopes. This study will address this issue by developing a simple method for processing bone samples prior to DNA isolation. A liquid-based technique was applied to macerate bone fragments by incubating bone fragments in an enzyme solution, removing potential contaminants, followed by a bone tissue disruption. The TissueLyser II was used to disrupt animal bones and plant tissue without dry ice or liquid nitrogen for species identification. Bone fragments of fresh swine (Sus scrofa) ribs were used.
