Chronic kidney disease (CKD) in children is known to result in poor growth and quality of life, and frequently results in kidney failure. The Chronic Kidney Disease in Children study (CKiD) is a prospective cohort study enrolling children ages 1 to 16 to assess health outcomes in children with CKD including the effects of declining glomerular filtration rate and the resulting consequences of growth failure on morbidity. The Air Force employs ejection seats in its high-performance aircraft. While these systems are intended to ensure aircrew safety, the ejection process subjects the aircrew to potentially injurious forces. System validation includes evaluation of forces against a standard which is linked to the probability of injury. The Muti-Axial Neck Injury Criteria (MANIC) was developed to account for forces in all six degrees of freedom. Female retention rates in the US military have been considerably lower than that of their male counterparts for numerous years. In the Air Force, women represent 14 percent of officer ranks from O-5 level and above. Comparatively, the overall rate of women officers in service is 20 percent. Understanding the negative factors associated with the attrition rate of this group can help the Air Force leverage positive change. Understanding diagnostic tests and examining important features of novel coronavirus (COVID-19) infection are essential steps for controlling the current pandemic of 2020. In this paper, we study the relationship between clinical diagnosis and analytical features of patient blood panels from the US, Mexico, and Brazil. Our analysis confirms that among adults, the risk of severe illness from COVID-19 increases with pre-existing conditions such as diabetes and immunosuppression. Although more than eight months into pandemic, more data have become available to indicate that more young adults were getting infected. A challenge arising in cancer immunotherapy trial design is the presence of non-proportional hazards (NPH) patterns in survival curves. We considered three different NPH patterns caused by delayed treatment effect, cure rate and responder rate of treatment group in this dissertation. These three NPH patterns would violate the proportional hazard model assumption and ignoring any of them in an immunotherapy trial design will result in substantial loss of statistical power.

In this dissertation, improved statistical methods for time-series and lifetime data are developed. First, an improved trend test for time series data is presented. Then, robust parametric estimation methods based on system lifetime data with known system signatures are developed.

In the first part of this dissertation, we consider a test for the monotonic trend in time series data proposed by Brillinger (1989). It has been shown that when there are highly correlated residuals or short record lengths, Brillinger’s test procedure tends to have significance level much higher than the nominal level. Conventional asymptotic methods for survival analysis work well when sample sizes are at least moderately sufficient. When dealing with small sample sizes or rare events, the results from these methods have the potential to be inaccurate or misleading. To handle such data, an exact method is proposed and compared against two other methods: 1) the Cox proportional hazards model and 2) stratified logistic regression for discrete survival analysis data.

Infants with hypoplastic left heart syndrome require an initial Norwood operation, followed some months later by a stage 2 palliation (S2P). The timing of S2P is critical for the operation’s success and the infant’s survival, but the optimal timing, if one exists, is unknown. We attempt to estimate the optimal timing of S2P by analyzing data from the Single Ventricle Reconstruction Trial (SVRT), which randomized patients between two different types of Norwood procedure. For degradation data in reliability analysis, estimation of the first-passage time (FPT) distribution to a threshold provides valuable information on reliability characteristics. Recently, Balakrishnan and Qin (2019; Applied Stochastic Models in Business and Industry, 35:571-590) studied a nonparametric method to approximate the FPT distribution of such degradation processes if the underlying process type is unknown. In this thesis, we propose improved techniques based on saddlepoint approximation, which enhance upon their suggested methods. Numerical examples and Monte Carlo simulation studies are used to illustrate the advantages of the proposed techniques. In this paper, the point at issue is to deliberate point and interval estimations for the parameters of Weibull-Gamma distribution (WGD) using progressively Type-II censored (PROG-II-C) sample under step stress partially accelerated life test (SSPALT) model. The maximum likelihood (ML), Bayes, and four parametric bootstrap methods are used to obtain the point estimations for the distribution parameters and the acceleration factor. The Bayesian adjustment for confounding (BAC) is a Bayesian model averaging method to select and adjust for confounding factors when evaluating the average causal effect of an exposure on a certain outcome. We extend the BAC method to time-to-event outcomes. Specifically, the posterior distribution of the exposure effect on a time-to-event outcome is calculated as a weighted average of posterior distributions from a number of candidate proportional hazards models, weighing each model by its ability to adjust for confounding factors. Bridge decks are a significant factor in the deterioration of bridges, and substantially affect long-term bridge maintenance decisions. In this study, conditional survival (reliability) analysis techniques are applied to bridge decks to evaluate the age at the end of service life using the National Bridge Inventory records. As bridge decks age, the probability of survival and the expected service life would change. In this study, we investigate and propose an efficient way of handling survival clinical data. Simulation studies were conducted to compare performances of various forms of survival model techniques using an R package ``survsim". Models performance was conducted with varying sample sizes as small ($n5000$). Because earthquakes have a large impact on human society, statistical methods for better studying earthquakes are required. One characteristic of earthquakes is the arrival time of seismic waves at a seismic signal sensor. Once we can estimate the earthquake arrival time accurately, the earthquake location can be triangulated, and assistance can be sent to that area correctly. The main goal of this project was to examine the differences in perioperative hyperglycemia treatment received by patients with a diagnosis of diabetes mellitus (DM) and patients without a diagnosis of diabetes (NDM); and how these treatment differences can affect the length of hospital stay. Studies have revealed that, when comparing DM and NDM patients with the same degree of perioperative hyperglycemia, NDM patients suffer worse outcomes. In this paper, the test statistic for testing exponentiality against exponential better than used in Laplace transform order (EBUL) based on the Laplace transform technique is proposed. Pitman’s asymptotic efficiency of our test is calculated and compared with other tests. The percentiles of this test are tabulated. The powers of the test are estimated for famously used distributions in aging problems. In the case of censored data, our test is applied and the percentiles are also calculated and tabulated. Finally, real examples in different areas are utilized as practical applications for the proposed test.

Nuclear magnetic resonance (NMR) is a highly versatile analytical technique for studying molecular configuration, conformation, and dynamics, especially of biomacromolecules such as proteins. However, due to the intrinsic properties of NMR experiments, results from the NMR instruments require a refencing step before the down-the-line analysis. Poor chemical shift referencing, especially for 13C in protein Nuclear Magnetic Resonance (NMR) experiments, fundamentally limits and even prevents effective study of biomacromolecules via NMR. There is no available method that can rereference carbon chemical shifts from protein NMR without secondary experimental information such as structure or resonance assignment.

Quantifying human biological age is an important and difficult challenge. Different biomarkers and numerous approaches have been studied for biological age prediction, each with its advantages and limitations. In this work, we first introduce a new anthropometric measure (called Surface-based Body Shape Index, SBSI) that accounts for both body shape and body size, and evaluate its performance as a predictor of all-cause mortality. Delayed entry arises frequently in follow-up studies for survival outcomes, where additional study subjects enter during the study period. We propose a quantile regression model to analyze survival data subject to delayed entry and right-censoring. Such a model offers flexibility in assessing covariate effects on survival outcome and the regression coefficients are interpretable as direct effects on the event time. Under the conditional independent censoring assumption, we proposed a weighted martingale-based estimating equation, and formulated the solution finding as a $\ell_1$-type convex optimization problem, which was solved through a linear programming algorithm. Alzheimer's Disease (AD) affects nearly 5.4 million Americans as of 2016 and is the most common form of dementia. The disease is characterized by the presence of neurofibrillary tangles and amyloid plaques [1]. The amount of plaques are measured by Braak stage, post-mortem. It is known that AD is positively associated with hypercholesterolemia [16]. As statins are the most widely used cholesterol-lowering drug, there may be associations between statin use and AD. We hypothesize that those who use statins, specifically lipophilic statins, are more likely to have a low Braak stage in post-mortem analysis.

The popular Kaplan-Meir estimator has traditionally been used to great effect as a survival function estimator. However, the Kaplan-Meir estimator is dependent upon a maximum likelihood parameter estimator which may not be the best estimator in all cases. We modify the Kaplan-Meir estimator, based on a Bayes parameter estimation, in hopes of providing a more accurate survival estimator for small sample sizes. In family studies, we are interested in estimating the penetrance function of the event of interest in the presence of competing risks. Failure to account for competing risks may lead to bias in the estimation of the penetrance function. In this thesis, three statistical challenges are addressed: clustering, missing data, and competing risks. We proposed the cause-specific model with shared frailty and ascertainment correction to account for clustering and competing risks along with ascertainment of families into study. A widely used tool in survival analysis is the Cox proportional hazards regression model.
