Efficacy testing is a cornerstone of clinical trials, ensuring that medical interventions achieve their intended therapeutic effects. Over the decades, a wide range of statistical methodologies have been developed to address the complexities of clinical trial data, including parametric, nonparametric, Bayesian, and machine learning approaches. Parametric methods, such as t-tests, ANOVA, and LMMs, have traditionally been the foundation of efficacy testing due to their efficiency under well-defined assumptions. Nonparametric techniques, including the Friedman test, Brunner-Munzel test, and modern extensions like nparLD, have emerged as robust alternatives, particularly for skewed, ordinal, or non-normal data. Meta-analysis of longitudinal data, where some of the studies report results with means and standard errors while others use medians and ranges, is a complex analytical problem with several challenges which must be overcome. Existing methods for estimating means from medians and either ranges, interquartile ranges, or both have not previously been evaluated in a longitudinal setting. In this work, a simulation study was used to estimate mean bias, between studies variance bias, and coverage of confidence intervals in a longitudinal setting. In this paper, we present an empirical evaluation of the randomness of the ciphertext blocks generated by the Advanced Encryption Standard (AES) cipher in Counter (CTR) mode and in Cipher Block Chaining (CBC) mode. Vulnerabilities have been found in the AES cipher that may lead to a reduction in the randomness of the generated ciphertext blocks that can result in a practical attack on the cipher. We evaluate the randomness of the AES ciphertext using the standard key length and NIST randomness tests. Background: Longitudinal measurement is commonly employed in health research and provides numerous benefits for understanding disease and trait progression over time. More broadly, it allows for proper treatment of correlated responses within clusters. We evaluated 3 methods for analyzing genome-by-epigenome interactions with longitudinal outcomes from family data.

Results: Linear mixed-effect models, generalized estimating equations, and quadratic inference functions were used to test a pharmacoepigenetic effect in 200 simulated posttreatment replicates. Adjustment for baseline outcome provided greater power and more accurate control of Type I error rates than computation of a pre-to-post change score.

With the influx of high-dimensional data there is an immediate need for statistical methods that are able to handle situations when the number of predictors greatly exceeds the number of samples. One such area of growth is in examining how environmental exposures to toxins impact the body long term. The cytokinesis-block micronucleus assay can measure the genotoxic effect of exposure as a count outcome. To investigate potential biomarkers, high-throughput assays that assess gene expression and methylation have been developed. Longitudinal studies of older adults usually need to account for deaths and missing data. The study databases often include multiple health-related variables, whose trends over time are hard to compare because they were measured on different scales. Here we present a unified approach to these three problems that was developed and used in the Cardiovascular Health Study. Data were first transformed to a new scale that had integer/ratio properties, and on which “dead” logically takes the value zero. Missing data were then imputed on this new scale, using each person’s own data over time. Imputation could thus be informed by …

Background: The aging process can be described as the change in health-related variables over time. Unfortunately, simple graphs of available data may be misleading if some people die, since they may confuse patterns of mortality with patterns of change in health. Methods have been proposed to incorporate death into self-rated health (excellent to poor) and the SF-36 profile scores, but not for other variables.
