Efficacy Analysis In Clinical Trials: A Comprehensive Review Of Statistical And Machine Learning Approaches, Dhrubajyoti Ghosh, Samhita Pal
Faculty Articles
Efficacy testing is a cornerstone of clinical trials, ensuring that medical interventions achieve their intended therapeutic effects. Over the decades, a wide range of statistical methodologies have been developed to address the complexities of clinical trial data, including parametric, nonparametric, Bayesian, and machine learning approaches. Parametric methods, such as t-tests, ANOVA, and LMMs, have traditionally been the foundation of efficacy testing due to their efficiency under well-defined assumptions. Nonparametric techniques, including the Friedman test, Brunner-Munzel test, and modern extensions like nparLD, have emerged as robust alternatives, particularly for skewed, ordinal, or non-normal data. Bayesian methodologies have enabled the incorporation of …

An Association Test For Ordinal Outcomes In Clustered Data With Informative Cluster Size, Hasika K. Wickrama Senevirathne, Sandipan Dutta
Mathematics & Statistics Faculty Publications
In cluster-correlated data, the number of observations in a cluster can be associated with the outcome from that cluster. This phenomenon is known as informative cluster size which can occur in cluster-randomized clinical trial data. Several studies have found that ignoring the issue of informative cluster size can produce biased results in the analysis of clustered data. Most of the existing methods for addressing informative cluster size are suited to continuous outcomes. However, ordinal outcomes and covariates are often encountered in clustered data obtained from large clinical studies. The existing methods for ordinal association testing in clustered data can produce …
Multiple Changepoint Detection For Non-Gaussian Time Series, Robert Lund, Thomas J. Fisher, Norou Diawara, Michael Wehner
Multiple Changepoint Detection For Non-Gaussian Time Series, Robert Lund, Thomas J. Fisher, Norou Diawara, Michael Wehner
Mathematics & Statistics Faculty Publications
This article combines methods from existing techniques to identify multiple changepoints in non‐Gaussian autocorrelated time series. A transformation is used to convert a Gaussian series into a non‐Gaussian series, enabling penalized likelihood methods to handle non‐Gaussian scenarios. When the marginal distribution of the data is continuous, the methods essentially reduce to the change of variables formula for probability densities. When the marginal distribution is count‐oriented, Hermite expansions and particle filtering techniques are used to quantify the scenario. Simulations demonstrating the efficacy of the methods are given and two data sets are analyzed: 1) the proportion of home runs hit by …
Bridging Machine Learning And Islamic Scholarship: A Study In Hadith Translation And Similarity Analysis, Asiyah R. Speight
Bridging Machine Learning And Islamic Scholarship: A Study In Hadith Translation And Similarity Analysis, Asiyah R. Speight
Student Scholar Symposium Abstracts and Posters
Translation of Islamic religious texts poses unique challenges requiring both linguistic and theological expertise. This study explores the application of neural machine translation (NMT) models to Arabic-English hadith translation while analyzing semantic similarity patterns across different human translations. Using the complete Sahih Bukhari corpus (7,550 hadiths) as the primary dataset, we adopt a dual approach combining transfer learning and comprehensive neural network analysis to demonstrate the critical impact of corpus size on model performance.
First, we fine-tune a pre-trained MarianMT Arabic-English translation model on the full Sahih Bukhari corpus, comparing models trained on 40 hadiths versus 7,550 hadiths. Performance is …
Changepoint Detection As Model Selection: A General Framework, Michael A. Grantham
Changepoint Detection As Model Selection: A General Framework, Michael A. Grantham
Dissertations and Doctoral Documents, University of Nebraska-Lincoln, 2023–
This dissertation presents a general framework for changepoint detection based on ℓ0 model selection. The core method, Iteratively Reweighted Fused Lasso (IRFL), improves upon the generalized lasso by adaptively reweighting penalties to enhance support recovery and minimize criteria such as the Bayesian Information Criterion (BIC). The approach allows for flexible modeling of seasonal patterns, linear and quadratic trends, and autoregressive dependence in the presence of changepoints.
Simulation studies demonstrate that IRFL achieves accurate changepoint detection across a wide range of challenging scenarios, including those involving nuisance factors such as trends, seasonal patterns, and serially correlated errors. The framework is …
Latent Classification Of Time-Dependent Transition Rates In Longitudinal Binary Outcome Data, Joonha Chang, Wenyaw Chan
Latent Classification Of Time-Dependent Transition Rates In Longitudinal Binary Outcome Data, Joonha Chang, Wenyaw Chan
School of Public Health Faculty Publications
Continuous-time Markov chain (CTMC) models and latent classification methods are commonly used to analyze longitudinal categorical outcomes in medical research. While CTMC models are popular for their simplicity and effectiveness, their assumption of constant transition rates presents limitations in capturing dynamic behaviors. To address this, non-homogeneous continuous-time Markov chains (NH-CTMCs) have been developed, incorporating time-varying transition rates to enhance model flexibility. In this study, we leverage closed-form transition probabilities for a fully ergodic two-state NH-CTMC model and propose a latent class clustering approach to identify heterogeneous transition rate patterns within the population. We emphasize the potential advantages of these models …

Children spend most of their time at home in their early years, yet efforts to promote human capital at home in many low- and middle-income settings remain limited. We conduct a randomized controlled trial to evaluate an intervention which encourages parents and caregivers to foster human capital accumulation among their children between ages 3 and 5, with a focus on math and phonics skills. Children gain 0.52 and 0.51 standard deviations relative to the control group on math and phonics tests, respectively (p<0.001). A year later effects persist, but math gains dissipate to 0.15 (p=0.06) and phonics to 0.13 (p=0.12). Effects appear to be mediated largely through instructional support by parents and not other parent investment mechanisms, such as more positive parent-child interactions or additional time spent on education at home beyond the intervention. Our results show that parents can be effective conduits of educational instruction even in low-resource settings.
Gender-Specific Mental Health Outcomes In Central America: A Natural Experiment, Thea Nagasuru
Gender-Specific Mental Health Outcomes In Central America: A Natural Experiment, Thea Nagasuru
Computer Science Summer Fellows
While COVID lockdown measures have had varying effects on the mental health of different demographics, several bodies of research have noted their disparate effect on women. Why is women's mental health more negatively impacted by lockdown measures, and how much more are they impacted than men? How can we predict and mitigate these negative effects on women? This paper aims to contribute to answering those questions by comparing COVID stringency measures and their effect on the gap in depression rates between men and women in two neighboring countries: Nicaragua and Honduras.

This paper describes the creation of a fast, deterministic, 3D fractal cloud renderer for the AFIT Sensor and Scene Emulation Tool (ASSET). The renderer generates 3D clouds by ray marching through a volume and sampling the level-set of a fractal function. The fractal function is distorted by a displacement map, which is generated using horizontal wind data from a Global Forecast System (GFS) weather file. The vertical windspeed and relative humidity are used to mask the creation of clouds to match realistic large-scale weather patterns over the Earth. Small-scale detail is provided by the fractal functions which are tuned to …

Prevalence Of Sars-Cov-2 Antibodies In Liberty University Student Population, Emily Bonus
Prevalence Of Sars-Cov-2 Antibodies In Liberty University Student Population, Emily Bonus
Senior Honors Theses
In 2020, the virus SARS-CoV-2 gained attention as it spread around the world. Its antibodies are poorly understood, and little research focuses on those with few COVID-19 complications yet large numbers of close contacts: university students. This longitudinal study recorded SARS-CoV-2 antibody presence in 107 undergraduate Liberty University students twice during early 2021. After extensive data cleaning and the application of various statistical tests and ANOVAs, the data seems to show that in the case of COVID-19 infections, SARS-CoV-2 IgM antibodies are immediately produced, and then IgG antibodies follow later. However, the COVID-19 vaccine causes the production of both IgM …

The Impact Of Service Dogs On Objective And Perceived Sleep Quality For Veterans With Ptsd, Madhuri Vempati, Elise A. Miller, Sarah C. Leighton, Leanne O. Nieforth, Marguerite O’Haire
Discovery Undergraduate Interdisciplinary Research Internship
One in four post-9/11 veterans (Fulton et al., 2015) have been diagnosed with posttraumatic stress disorder (PTSD), facing sleep disruptions as one of their most common symptoms. Service dogs have become an increasingly popular complementary intervention and anecdotes suggest they may impact sleep for veterans with PTSD. There is a need for empirical investigation into these claims through measurement and analysis of sleep quality.
The purpose of this study was to longitudinally investigate the impact of service dogs on sleep quality through both objective and subjective measures.
Participants in the treatment group (n=92) received a service dog after baseline, while …

A Novel Correction For The Adjusted Box-Pierce Test, Sidy Danioko, Jianwei Zheng, Kyle Anderson, Alexander Barrett, Cyril S. Rakovski
Mathematics, Physics, and Computer Science Faculty Articles and Research
The classical Box-Pierce and Ljung-Box tests for auto-correlation of residuals possess severe deviations from nominal type I error rates. Previous studies have attempted to address this issue by either revising existing tests or designing new techniques. The Adjusted Box-Pierce achieves the best results with respect to attaining type I error rates closer to nominal values. This research paper proposes a further correction to the adjusted Box-Pierce test that possesses near perfect type I error rates. The approach is based on an inflation of the rejection region for all sample sizes and lags calculated via a linear model applied to simulated …

Deep Depression Prediction On Longitudinal Data Via Joint Anomaly Ranking And Classification, Guansong Pang, Ngoc Thien Anh Pham, Emma Baker, Rebecca Bentley, Anton Van Den Hengel
Research Collection School Of Computing and Information Systems
A wide variety of methods have been developed for identifying depression, but they focus primarily on measuring the degree to which individuals are suffering from depression currently. In this work we explore the possibility of predicting future depression using machine learning applied to longitudinal socio-demographic data. In doing so we show that data such as housing status, and the details of the family environment, can provide cues for predicting future psychiatric disorders. To this end, we introduce a novel deep multi-task recurrent neural network to learn time-dependent depression cues. The depression prediction task is jointly optimized with two auxiliary anomaly …

Using Hac Estimators For Intervention Analysis, Ashok K. Singh, Rohan J. Dalpatadu
Using Hac Estimators For Intervention Analysis, Ashok K. Singh, Rohan J. Dalpatadu
Hospitality Faculty Research
The purpose of this article is to present an alternative method for intervention analysis of time series data that is simpler to use than the traditional method of fitting an explanatory Autoregressive Integrated Moving Average (ARIMA) model. Time series regression analysis is commonly used to test the effect of an event on a time series. An econometric modeling method, which uses a heteroskedasticity and autocorrelation consistent (HAC) estimator of the covariance matrix instead of fitting an ARIMA model, is proposed as an alternative. The method of parametric bootstrap is used to compare the two approaches for intervention analysis. The results …

Diabetes And Its Effect On Abdominal Aortic Aneurysm Growth Rate In Hispanic Patients, Monica Betancourt-Garcia, Kristina Vatcheva, Amrit Thakur, Prateek Gupta, Eugene Postevka, Ricardo Martinez, R. Armour Forse
School of Mathematical & Statistical Sciences Faculty Publications
Background
The growth rate of abdominal aortic aneurysms (AAA) can vary depending on age, baseline diameter, blood pressure, race, and history of smoking. Paradoxically, previous studies show evidence of a protective effect of diabetes on the rate of AAA expansion despite its well-established role in the morbidity and mortality of cardiovascular disease. This study aims to investigate the impact diabetes plays on AAA growth within a Hispanic population.
Methods
Data were collected from patients who were predominantly Mexican-American at a single hospital site. Baseline and follow-up measures for AAA diameter were obtained from serial imaging studies. Demographics, medical history, the …
Feature Selection For Longitudinal Data By Using Sign Averages To Summarize Gene Expression Values Over Time, Suyan Tian, Chi Wang
Feature Selection For Longitudinal Data By Using Sign Averages To Summarize Gene Expression Values Over Time, Suyan Tian, Chi Wang
Biostatistics Faculty Publications
With the rapid evolution of high-throughput technologies, time series/longitudinal high-throughput experiments have become possible and affordable. However, the development of statistical methods dealing with gene expression profiles across time points has not kept up with the explosion of such data. The feature selection process is of critical importance for longitudinal microarray data. In this study, we proposed aggregating a gene’s expression values across time into a single value using the sign average method, thereby degrading a longitudinal feature selection process into a classic one. Regularized logistic regression models with pseudogenes (i.e., the sign average of genes across time as predictors) …

A Logitudinal Feature Selection Method Identifies Relevant Genes To Distinguish Complicated Injury And Uncomplicated Injury Over Time, Suyan Tian, Chi Wang, Howard H. Chang
Biostatistics Faculty Publications
Background: Feature selection and gene set analysis are of increasing interest in the field of bioinformatics. While these two approaches have been developed for different purposes, we describe how some gene set analysis methods can be utilized to conduct feature selection.
Methods: We adopted a gene set analysis method, the significance analysis of microarray gene set reduction (SAMGSR) algorithm, to carry out feature selection for longitudinal gene expression data.
Results: Using a real-world application and simulated data, it is demonstrated that the proposed SAMGSR extension outperforms other relevant methods. In this study, we illustrate that a gene’s expression profiles over …

Role Of Misclassification Estimates In Estimating Disease Prevalence And A Non-Linear Approach To Study Synchrony Using Heart Rate Variability In Chickens, Dola Pathak
Department of Statistics: Dissertations, Theses, and Student Research
Infectious disease assays can be imperfect. When estimating disease prevalence, these imperfections are accounted for by incorporating assay sensitivity and specificity into point and variance estimates. Unfortunately, these accuracy measures are often treated as fixed constants, rather than acknowledging that they are estimates from an assay validation process. The purpose of this study is to show the detrimental effect of not taking into account this sampling variability when samples are obtained through group testing (aka, pooled testing). We show that confidence interval coverage can dramatically decline as the sample size increases for the main sample of interest. As a remedy …

Monitoring And Evaluating The Influences Of Class V Injection Wells On Urban Karst Hydrology, James Adam Shelley
Masters Theses & Specialist Projects
The response of a karst aquifer to storm events is often faster and more severe than that of a non-karst aquifer. This distinction is often problematic for planners and municipalities, because karst flooding does not typically occur along perennial water courses; thus, traditional flood management strategies are usually ineffective. The City of Bowling Green (CoBG), Kentucky is a representative example of an area plagued by karst flooding. The CoBG, is an urban karst area (UKA), that uses Class V Injection Wells to lessen the severity of flooding. The overall effectiveness, siting, and flooding impact of Injection Wells in UKA’s is …

Longitudinal Data Methods For Evaluating Genome-By-Epigenome Interactions In Families, Justin C. Strickland, I-Chen Chen, Chanung Wang, David W. Fardo
Psychology Faculty Publications
Background: Longitudinal measurement is commonly employed in health research and provides numerous benefits for understanding disease and trait progression over time. More broadly, it allows for proper treatment of correlated responses within clusters. We evaluated 3 methods for analyzing genome-by-epigenome interactions with longitudinal outcomes from family data.
Results: Linear mixed-effect models, generalized estimating equations, and quadratic inference functions were used to test a pharmacoepigenetic effect in 200 simulated posttreatment replicates. Adjustment for baseline outcome provided greater power and more accurate control of Type I error rates than computation of a pre-to-post change score.
Conclusions: Comparison of all modeling approaches indicated …

Based on the six-wave data collected from Grade 7 to Grade 12 students (N = 3,328 at Wave 1), this pioneer study examined the development of problem behaviors (risk and delinquent behavior and problem behavioral intention) and the predictors (moral competence and spirituality) among adolescents in Hong Kong. Individual growth curve models revealed that while risk and delinquent behavior accelerated and then slowed down in the high school years, adolescent problem behavioral intention slightly accelerated over time. After controlling the background socio-demographic factors, moral competence and spirituality were negatively associated with risk and delinquent behavior as well as problem …
