Photodynamic therapy (PDT) is a minimally invasive treatment that uses light-activated photosensitizers to selectively destroy diseased tissue through the generation of reactive oxygen species. However, conventional in vitro models often fail to accurately reproduce the pharmacokinetics of photosensitizer accumulation and clearance observed in vivo. This study investigated protoporphyrin IX (PpIX) accumulation following 5-aminolevulinic acid (ALA) exposure under physiologically relevant conditions using two-dimensional (2D) monolayers and three-dimensional (3D) spheroids. By comparing continuous ALA exposure with transient exposure followed by ALA washout, this work provides a more biologically relevant framework for studying PpIX kinetics and optimizing photodynamic therapy.

The overarching motivation of this work is the development of seedless, non-intrusive, laser-based diagnostics for supersonic airflow in wind tunnels. To advance this goal, this dissertation fo-cuses on three-photon excitation in a research-grade argon beam within a tabletop vacuum system, which offers a controlled environment for testing and refining the approach. The chosen excitation scheme drives argon atoms to the 3d[5/2]₃ state using a pulsed Ti:Sapphire laser system, enabling time-of-flight (ToF) measurements on atoms that subsequently undergo a multi-step decay to the metastable 4s[3/2]₂ state. This dissertation presents novel algorithms to improve the mapping capabilities of a 360-degree underwater Pulsed Laser Line Scanner LiDAR (PLLS-360°). Due to its 360° field-of-view (FOV), the PLLS-360° is a compact full-waveform omnidirectional imager suitable for seafloor mapping, underwater asset inspection, object detection, ice-sheet mapping, and construction progress monitoring. The proposed methodology includes an improved waveform fitting technique for saturated waveform recovery, detection array response correction, radiometric corrections, and fusion of LiDAR and sonar bathymetric datasets. High-power fiber lasers are steadily increasing in maximum power and rapidly approaching theoretical maximum power output from a single fiber aperture. The low thermal conductivity of silica fibers is an inherent limitation to the dissipation of heat from the fiber core, and the buildup of heat within the laser gain media is the principal driver of transverse mode instability within fiber systems. Silicon photonic integrated circuits enable the capability of producing on-chip processing of photonic signals, including both emission and detection. Emission materials, however, have been limited for monolithic integration due to high processing temperatures, indirect bandgap materials, and material integration complications. The (Si)GeSn material system has recently attracted attention due to the bandgap transition from the Si and Ge, naturally indirect bandgap, to direct bandgap with sufficient Sn incorporation. This research documents steps towards building a novel spectroscopic technique, namely, cavity-enhanced double-resonance (CEDR) spectroscopy, for investigating methane (CH4) and other molecules with significance in astrochemistry. These efforts focus on tackling the complexities of their ro-vibrational energy levels and the inefficiencies in analyzing vibrational spectra, particularly for molecules with high symmetry and strong intramolecular interactions. In the CEDR spectroscopy, the first photon, generated by a continuous-wave optical parametric oscillator (CW-OPO) locked to a Doppler-free saturation absorption line, excites the molecule from its ground level to a selected excited ro-vibrational (rotational-vibrational) level, e.g., that of the asymmetric CH-stretch mode. …

Master Oscillator Power Amplifiers (MOPA) are laser systems that utilize a seed and pump amplification system to boost the output power of high-quality lower power seeding signals. MOPAs can generate high gain while avoiding many of the nonlinearities that negatively affect resonance-based lasers that are known to feature higher internal intensities. Additionally, MOPAs provide an easy alternative to the construction of novel laser technologies for higher output power as they can be easily combined with existing laser sources to amplify their output power.

In this experiment, an intensity autocorrelator is set up in order to measure the pulse duration of a passively mode-locked Titanium-Sapphire laser with a power of 267mW producing femtosecond pulses. Then this measurement, as well as a measurement of the laser’s spectrum, is used to test the Uncertainty Principle. Intensity autocorrelation is a well-established technique for measuring pulse duration, and is among the more intuitive techniques for this purpose, which is why it was selected for this experiment. Laser filamentation is a fascinating phenomenon that occurs when an intense laser beam travels through transparent materials, in particular air. At sufficiently high power (TW in the near IR, GW in the UV), instead of spreading out like a regular laser beam, something remarkable happens: the laser beam becomes tightly focused, creating a thin and intense column of light called a laser filament. In this dissertation, I investigate the possibility of lasing and nonlinear phenomena in completely solid-state transverse Anderson localizing optical fibers (TALOFs). I examine three areas within this range of topics. The research in nonlinear phenomena focuses on four-wave mixing (FWM). FWM is of high interest in TALOFs due to the fact that guided localized modes of the fiber each have different propagation constants, and thus unique possible FWM pairs can be generated from the same input pump beam. The focus of this dissertation is the development and investigation of nested cavity mode-locked lasers and their resultant tailored frequency combs. A nested cavity is made up of two cavities, known as parents. One parent is a larger, active, 100MHz Ti:Saph oscillator and the other is a smaller, passive, 7GHz Fabry-Perot Etalon (FPE). Two-photon fluorescence microscopy is a powerful tool to study the molecular and cellular interactions. This technology is a non-invasive approach with the advantage of three-dimensional imaging up to submicron resolution. Two-photon excitation process is the result of simultaneous absorption of two photons that has special features of reduced photodamaged and elongated penetration depth on samples. In this work, blinking statistics of silver-coated Cobalt ferrite (CoFe2O4) single nanocrystal is studied using two-photon fluorescence microscopy. Every gas molecule has a unique absorption spectrum that can be captured using optical spectroscopy to identify an unknown sample's composition. Frequency combs systems can provide an extremely broad mid-infrared spectrum that is very useful for molecular detection. A degenerate optical parametric oscillator (OPO) was built to generate the down-converted and shifted frequency comb spectrum. This system utilizes an ultra-short pulse 1.56µm pump laser and a never before used orientation patterned gallium-phosphide crystal. This research was part of an effort to experimentally validate computational models under development for radiation-induced atmospheric effects. Cavity Ringdown Spectroscopy (CRDS) was used to measure the concentration of chemical products generated as a result of radiation interactions in a controlled atmosphere. Experiments were conducted in a vacuum chamber interfaced with a gas introduction system that controlled the initial atmospheric composition. A quadrupole mass spectrometer and tunable dye laser were integrated to confirm initial atmospheric composition, and provide wavelength flexibility for detecting a variety of chemical products generated by radiation interactions. This thesis focuses on a technique of delivering spatially focused and temporally compressed picosecond laser pulses through multimode fibers. This study was inspired by recent success in focusing light through optically diffusive media of which multimode fibers were a special case in terms of causing scrambled phase distribution in the transmitted light. We report the results of experimental and numerical-simulation studies of shielded radiography using narrowband MeVenergy X-rays from a compact all-laser-driven inverse-Compton-scattering X-ray light source. This recently developed Xray light source is based on a laser-wakefield accelerator with ultra-high-field gradient (GeV/cm). We demonstrate experimentally high-quality radiographic imaging (image contrast of 0.4 and signal-to-noise ratio of 2:1) of a target composed of 8-mm thick depleted uranium shielded by 80-mm thick steel, using a 6-MeV X-ray beam with a spread of 45% (FWHM) and 107 photons in a single shot. The number density of cold atoms confined in a magneto-optical trap (MOT) is critically dependent on the intensity of the lasers used to cool the sample. To generate large optical powers while retaining the practicality of homemade external cavity diode lasers (ECDLs), a tapered amplifier (TA) system was designed and constructed to amplify the output of an existing 780 nm, continuous-wave ECDL. The goal of this project is to experimentally investigate the optical Talbot effect and the electron Talbot effect. The Talbot effect is a near-field diffraction effect which occurs when plane waves are incident upon a grating. The Talbot effect creates full grating revivals at integral Talbot lengths and revivals with greater spatial periodicity at fractional Talbot lengths. We use a green helium neon laser and Ronchi rulings to take CCD camera images of the fractional Talbot revivals directly. This paper will discuss both theoretical and experimental attempts to improve the spatial beam quality of diode laser stacks using an external optical system. An overview and derivation of the mathematics of both the optical system and diode lasers will be discussed. Laser induced damage is of interest in studying the transmission of large amounts of optical energy through step-index, large core multimode fibers. Optical fibers often have to be routed around objects when laser light is being transmitted between two locations which require the fiber to bend into a curve. The Linac Coherent Light Source (LCLS) at SLAC depends on a photocathode electron gun to provide the linear accelerator with the raw material – electrons – used for making X-ray laser pulses. The photocathode used in the LCLS Injector is a clean copper plate in high vacuum. When the cathode is struck with high energy UV light, electrons are liberated from its surface and then accelerated down the linac with radio-frequency electric fields. These fast-moving bunches of electrons are directed through an undulator magnet to radiate X-ray light.
