This thesis investigates the relationship between macroscopic rheological signals and underlying microscopic dynamics in complex fluids, specifically focusing on ”living polymers” within the cetyltrimethylammonium bromide (CTAB) and sodium salicylate (NaSal) surfactant system. The research addresses the inverse parameterization problem, demonstrating how bulk measurements like zero-shear viscosity can mask fundamentally different physical topologies, such as purely cylindrical, reptating micelles versus highly branched networks. Through the successful synthesis of viscoelastically ”degenerate” samples, the study utilizes an array of characterization techniques including frequency sweeps, Large Amplitude Oscillatory Shear (LAOS) to resolve these unique underlying states. Photodynamic therapy (PDT) is a minimally invasive treatment that uses light-activated photosensitizers to selectively destroy diseased tissue through the generation of reactive oxygen species. However, conventional in vitro models often fail to accurately reproduce the pharmacokinetics of photosensitizer accumulation and clearance observed in vivo. This study investigated protoporphyrin IX (PpIX) accumulation following 5-aminolevulinic acid (ALA) exposure under physiologically relevant conditions using two-dimensional (2D) monolayers and three-dimensional (3D) spheroids. By comparing continuous ALA exposure with transient exposure followed by ALA washout, this work provides a more biologically relevant framework for studying PpIX kinetics and optimizing photodynamic therapy.

This thesis develops a documented, reproducible procedure for building diffraction-limited microscope objectives entirely from catalog singlets, for imaging ultracold 6Li atoms in a ring-trap experiment. The objectives must resolve micron-scale features through a 5 mm fused-silica vacuum window, operate at multiple wavelengths, fit inside a 48 mm magnet bore, and contain no conductive or magnetic material, because the surrounding coils switch 0.1 T fields on microsecond timescales. Commercial long-working-distance objectives are universally housed in metal, which the eddy-current constraint rules out, and custom fabrication of a suitable non-conductive matched pair is estimated at nearly $200k.

Measuring Cosmic Expansion From Early To Late Times Using Quasars, Galaxies, & Local Supernovae, Rajeev Vaisakh

This dissertation investigates the evolution of cosmic expansion across a wide redshift ($z$) range by combining measurements from quasars \& galaxies in the distant universe $(z > 0.8)$ with those obtained from stripped-core collapse supernovae observations from the nearby universe $(z < 0.1)$. Using the DESI spectroscopic survey, we analyze large-scale clustering from quasars over $0.8 < z < 2.1$ and emission line galaxies over $0.8 < z < 1.6$, contributing to DESI DR2 BAO and full-shape measurements of cosmic expansion and structure growth. Combined with BBN in flat $\Lambda$CDM, DESI DR2 BAO gives $H_0 = 68.51 \pm 0.58$ km s$^{-1}$ Mpc$^{-1}$, while combinations of DESI BAO with CMB and supernova data show a preference for evolving dark energy, with representative constraints such as $w_0 = -0.752 \pm 0.057$ and $w_a = -0.86^{+0.23}_{-0.20}$ for DESI+CMB+DESY5. Using the ROTSE-III Supernova Survey and the expanding photosphere method (EPM), we measure distances to a sample of six supernovae for which the thermal phase can be clearly identified. This analysis focuses on testing whether reliable EPM distances can be obtained for this population. The resulting distances will eventually be compared with independent estimates from the literature. Establishing consistency with these external measurements represents an important step toward applying this methodology to larger samples, with the eventual goal of using ROTSE supernova distances to provide an independent measurement of the local Hubble constant.

Waterborne viral contamination can pose substantial public-health risks, making early detection essential for protecting water quality and responding quickly to treatment failures or contamination events. This dissertation presents a single-shot phase and amplitude retrieval technique based on spatial light modulator (SLM)-based digital holography. The proposed method enables accurate reconstruction of complex optical fields from a single interferogram, reducing sensitivity to environmental fluctuations and improving experimental stability compared to conventional multi-shot approaches. In addition, a programmable arbitrary mode converter is developed using SLM-based wavefront shaping. By introducing controlled astigmatism and engineering the differential Gouy phase between orthogonal components, the system enables flexible transformation between structured light modes, including Hermite–Gaussian and Laguerre–Gaussian beams. Fiber laser amplifier (FLA) models are often implemented without rigorous mathematical justification or thorough numerical validation. Without a proper theoretical basis for assumptions and approximations, or a technical analysis of model performance, there is significant uncertainty about the limitations of any given reduced model and its suitability for an application. Off-axis digital holographic microscopy (DHM) is a powerful tool for 3D, non-invasive live-cell tracking without moving parts. However, traditional setups face an inherent dilemma: split-path interferometers offer high spatial resolution but poor temporal stability, while more stable common-mode configurations are historically limited to lower numerical aperture (NA) regimes. Birefringence offers a promising way to observe stress concentration in materials such as glass and plastic, and thereby to identify weaknesses. Polarimetric imaging can be used to measure the birefringence of material, so long as the material is transparent to the light being used for the imaging. In this research, 2D Terahertz imaging was investigated as a means of measuring the birefringence of plastics that are opaque to visible light but transparent to THz radiation, for the eventual purpose of analyzing the residual stress present. This dissertation presents two experimental investigations at the intersection of quantum sensing and precision optical instrumentation. The primary project demonstrates optically detected nuclear magnetic resonance (NMR) of 13C nuclear spins in diamond, using state-selective Landau-Zener transitions under microwave frequency sweeping to bidirectionally transfer spin polarization between nitrogen-vacancy (NV) electron spins and remote 13C nuclear spins. This enables optical polarization and readout of large ensembles of polarized nuclear spins at low magnetic fields and room temperature, with spin dephasing times limited by longitudinal relaxation of nearby NV electron spins. Optical frequency combs consist of equidistant optical frequencies and have numerous applications ranging from optical metrology to medical diagnostics. Initially, frequency combs were based on bulky mode-locked lasers, but advancements in integrated photonics enabled the generation of frequency combs in chip-scale resonators (microcombs) using Kerr nonlinearity. These miniaturized systems present various challenges, including increased propagation losses, enhanced thermal effects, and the extension of microcombs to visible wavelengths. This thesis presents the design and simulation of an 8-tube single-ring anti-resonant hollow-core fiber for sensing applications, with particular emphasis on methane gas detection at the fundamental absorption wavelength of 3.3 µm. Conventional solid-core silica optical fibers exhibit strong multi-phonon material absorption beyond 2.5 µm, rendering them fundamentally unsuitable for efficient light guidance and direct gas sensing at mid-infrared wavelengths. Einstein’s formula to calculate the deflection angle of light through space as it interacts with gravity was introduced in his 1916 publication on general relativity. This was not a new idea, but his equation was, and it was correct. Just 3 years after this publication, it was empirically validated by Sir Arthur Eddington and Sir Frank Dyson. Since that experiment in 1919, at least seven others have been performed that also gave definitive answers in support of Einstein’s deflection constant of 1.751 arcseconds. Multiphoton lithography (MPL) is a method of laser-based 3D-printing for fabricating micron-scale structures point-by-point in a photopolymerizable medium. MPL’s high-resolution capabilities have made it a powerful method for the fabrication of many devices, such as microelectromechanical systems (MEMS) and tissue scaffolds. The throughput of MPL processes can be increased by using spatially shaped laser beams that expose large areas or volumes simultaneously. Mid-wave infrared (MWIR) optical systems require compact, broadband, manufacturable components whose performance can be predicted and measured reliably. Silicon metalenses are attractive because silicon combines high refractive index, MWIR transparency, and semiconductor-compatible fabrication. This dissertation develops a scale-bridging design–fabrication-modeling–metrology framework for MWIR silicon metalenses.
A shared full-aperture framework connects meta-atom libraries, physical layout generation, angular-spectrum propagation, point-spread-function calculation, and focal-plane energy metrics. Building on this foundation, the Dispersive Sweatt Model (DSM) incorporates meta-atom dispersion into conventional ray-tracing software, enabling broadband co-optimization of metasurfaces and refractive elements. Non-invasive imaging schemes are highly sought after methodologies in biology and metrology, where maintaining the physical state of a sample is important. Quantitative phase imaging (QPI) techniques non-invasively reconstruct the relative phase delay imparted onto an optical field due to transmission through a sample. Phase delay is proportional to the thickness and refractive index of the sample, but is lost in the intensity measurement process. Optical fiber systems based on solid-core silica waveguides underpin modern telecommunications, high-power laser delivery, precision sensing, and coherent optical systems. However, nonlinear effects, material absorption, and thermal limitations within silica increasingly constrain further scaling in both optical power and transmission performance. Antiresonant hollow-core fibers provide a promising alternative by guiding light predominantly in air, substantially reducing nonlinear interactions, latency, and optical damage while enabling transmission regimes inaccessible to conventional solid-core fibers. Physical reality is rarely deterministic; which, when probed by electromagnetic fields that also fluctuate, leads to observables that are most efficiently modelled as statistical processes. At equilibrium, this is usually achieved by invoking the Gaussian statistics of underlying physical processes, however, in practice, one often encounters out-of-equilibrium conditions where Gaussian descriptions may not suffice. This dissertation investigates the ultrafast dynamics of reactive intermediates, fragment trapping, and radical-mediated bond formation on oxide surfaces using time-of-flight mass spectrometry in conjunction with femtosecond pump-probe spectroscopy. Fluorescence microscopy is an indispensable tool in the biological sciences, enabling researchers to investigate intricate subcellular structures, particularly for volumetric studies. However, conventional optical microscopy for volumetric imaging remains fundamentally constrained by imaging speed and throughput. To bypass traditional serial z-scanning, we introduce an axially scan-free method using a phase layer cake to modulate the system's point spread function. This approach projects volumetric information onto a 2D plane in a single shot, offering high flexibility in tuning axial depth alongside simultaneous multicolor imaging with high spatial resolution and sensitivity. While many methods of time resolution are already established using classical resources, they fail to combine high precision outcomes with large-scale implementations. Additionally, numerous quantum networking architectures using satellite-assisted methods have demonstrated quantum communication on scales exceeding ground-based methods. The overarching motivation of this work is the development of seedless, non-intrusive, laser-based diagnostics for supersonic airflow in wind tunnels. To advance this goal, this dissertation fo-cuses on three-photon excitation in a research-grade argon beam within a tabletop vacuum system, which offers a controlled environment for testing and refining the approach. The chosen excitation scheme drives argon atoms to the 3d[5/2]₃ state using a pulsed Ti:Sapphire laser system, enabling time-of-flight (ToF) measurements on atoms that subsequently undergo a multi-step decay to the metastable 4s[3/2]₂ state. Oceanographic lidar systems remotely characterize the vertical structure of the upper ocean by recording the light backscattered from a laser pulse as it propagates through the water column. Historically these systems have been constrained by limited detection ranges, often capturing only a portion of the illuminated water column, typically fading to noise around the 10% isolume. A primary constraint has been the limited dynamic range of digitizers, which inhibits the ability to resolve both intense near-field and faint far-field backscatter signals within a single acquisition. The hypersonic regime poses numerous challenges that researchers face in the development of hypersonic flight vehicles. Due to their excellent thermomechanical properties, ultra-high-temperature ceramics (UHTCs) have risen as a promising solution to act as a protective barrier between the harsh environment and surface materials of these flight bodies. Background: Photodynamic Therapy (PDT) utilizes specific wavelengths of light to activate photosensitizing chemical compounds, known as photosensitizers, which induce the generation of cytotoxic reactive oxygen species (ROS) for the targeted destruction of cancer cells. Among various photosensitizers for PDT, Protoporphyrin IX (PpIX) is widely employed in oncology and dermatology due to its natural in situ generation via the metabolic conversion of 5- aminolevulinic acid (ALA), a non-phototoxic prodrug. Systemic administration of ALA after 3-6 hr drug delay leads to peak PpIX accumulation in tissues, facilitating therapeutic and diagnostic applications.
