Point-of-care (POC) biosensors, although rapid and easy-to-use, are orders magnitude less sensitive than laboratory-based tests. Further they are plagued by poor stability of recognition element thus limiting its widespread applicability in resource-limited settings. Therefore, there is a critical need for realizing stable POC biosensors with sensitivity comparable to gold-standard laboratory-based tests. This challenge constitutes the fundamental basis of this dissertation work– to expand access to quality and accurate biodiagnostic tools. It is possible to produce electron beams with non-zero orbital angular momentum. Such beams, known as electron vortex beams, are theoretically able to transfer their orbital angular momenta to matter, causing the matter to rotate. Nanoparticles in an aqueous solution were observed with an electron vortex beam to detect the transfer of orbital angular momentum in a low-friction environment. Observing the transfer of orbital angular momentum to particles in solution is difficult due to the necessity of imaging the particles through a liquid and the random movement of particles in the solution. This report focuses on the optical and electronic properties of graphene and topological insulators and how these Dirac fermion systems interact with energetic irradiation. We first present data exploring the effects of electron-beam and oxygen plasma induced disorder on the electronic properties and Raman spectra of graphene. These initial investigations were important for relating Raman peak intensities and weak localization features to each other and to an average disorder length in graphene, LD. ^ We then integrate gate-effect measurements into the Raman spectroscopy study to fully explore the relationships between carrier density, disorder and Raman spectrum signatures. Field Emission Studies Toward Improving The Performance Of Dc High Voltage Photoelectron Guns, Mahzad Bastaninejad

Field emission is the main mechanism that prevents DC high voltage photoemission electron guns from operating at the very high bias voltages required to produce low emittance beams. Gas conditioning is shown to eliminate field emission from cathode electrodes used inside DC high voltage photoelectron guns. Measurements and simulation results indicate that gas conditioning eliminates field emission from cathode electrodes via two mechanisms: sputtering and implantation, with the benefits of implantation reversed by heating the electrode.
