Synthetic polymers play an essential role in nearly every aspect of our lives. Extending beyond single-use packaging, polymeric material design has progressed to attain tailorable architectures granting exceptional performance across advanced applications, including carbon-fiber reinforced polymer composites for aerospace, conductive materials for soft electronics, and drug carriers for biomedicine. While highly promising, intricately designed polymers needed to achieve excellent performance often have limited processability, complex synthetic methods, and expensive precursors. Furthermore, due to a lack of recyclability, commodity polymer waste streams result in both environmental impacts and a substantial loss of economic value. Poly(ionic liquid)s (PILs) are a fascinating subclass of strong polyelectrolytes formed from polymerizable ionic liquids. As a result of their unique properties and counterion exchangeability, PILs can exhibit conformation structure or material property changes in response to external stimuli such as changes in pH/ionic environment, magnetic fields, and electric potentials. In Chapter 1, a comprehensive review of PILs design as well as their stimuli-responsive behavior is provided. Additional motivation for each dissertation chapter is also discussed. In Chapter 2, magnetically responsive PILs (MPILs) are developed from complexing paramagnetic salts with a random PIL copolymer containing a metal-coordinating co-monomer, acrylamide. A …

Bottlebrush polymers have garnered interest in many disciplines for their unique structure-property characteristics and potential applications. These polymers typically exhibit extended rodlike conformations as a result of sterically crowded side-chains. Further, extensive work has focused on expanding the synthetic toolbox for these systems, resulting in excellent molecular weight control, diverse functional group utility and corresponding stimuli-responsive behaviors. This dissertation examines both synthetic and stimuli-responsive tools in the context of bottlebrush systems.

In this thesis, I report the preparation and characterization of dye-containing PEG-b-PLA block copolymer micelles and the computational design of a novel coiled-coil peptide bundle. The PEG-b-PLA micelles encapsulate hydrophobic molecules into their core and have strong potential as nanocontainers or delivery vesicles. In theory, these internalized molecules can be released upon exposure to mechanical forces that disrupt the micellar structures. This force-responsive nature is one of the inherent properties of micellar systems. However, there is a stark lack of research that utilize this property in applications. Here, we present a method of gravity-drawing polydimethylsiloxane (PDMS) silicone fibers with application as fiber optics and as model foldamers. Beginning as a viscous liquid, PDMS is cured using heat until its measured viscosity reaches 4000 mPa•s. The semi-cured elastomer is then extruded through a tube furnace to produce thin (diameters on the order of hundred micrometers) filaments with scalable lengths. PDMS is biocompatible, gas-permeable, flexible, and hydrophobic. Additionally, the PDMS surface hydrophobicity can be modified via UV exposure, O2 plasma, and corona discharge. Polyester-based amphiphilic block copolymers and their nanoassemblies are of significant interest for a wide range of applications due to the degradability of the polyester block. However, the commonly used polyesters lack functional groups on their backbones, limiting the possibilities to chemically modify these polymers. Described here are new poly(ethylene oxide) (PEO)–poly(β-6-heptenolactone) (PHEL) block copolymers having pendant alkenes at each repeat unit on the PHEL block. First, the self-assembly of these block copolymers in aqueous solution was studied and it was found that they formed solid nanoparticles and vesicles depending on the relative block lengths. Nanoassemblies presenting multivalent displays of biologically active carbohydrates are of significant interest for a wide array of biomedical applications ranging from drug delivery to immunotherapy. In this study, glycodendron–lipid hybrids were developed as a new and tunable class of dendritic amphiphiles. A modular synthesis was used to prepare dendron–lipid hybrids comprising distearylglycerol and 0 through 4th generation polyester dendrons with peripheral protected amines. Following deprotection of the amines, an isothiocyanate derivative of C-linked α-galactose (α-Gal) was conjugated to the dendron peripheries, affording amphiphiles with 1 to 16 α-Gal moieties. The use of block polymers continues to gain attention with their myriad applications in industry for advanced applications in biology, medicine, electronics, and separations. The ability of block polymers to self assemble into ordered states on the nanometer level makes these materials suitable for applications that mandate structural order on this scale. Self-assembly of nanoparticles can potentially provide improvements for many current applications and could influence the design of future technologies. The assembly of nanoparticles, which can be controlled through external forces or through molecular interactions, can affect the properties of a material. Nanoparticles can be chemically modified by attaching molecules such as polymers to their surface. The nature and placement of these tethers affects the arrangement of the nanoparticles leading to materials with novel chemical, electrical, and physical properties. This dissertation is related to the studies of functionalized nanoparticles for self-assembly and as controlled drug delivery system. The whole topic is composed of two parts. In the first part, the research was conducted to design and synthesize a new type of ionic peptide-functionalized copolymer conjugates for self-assembly into nanoparticle fibers and 3D scaffolds with the ability of multi-drug loading and governing the release rate of each drug for tissue engineering. The self-assembly study confirmed that such peptide-functionalized amphiphilic copolymers underwent different self-assembly behavior.
