The following thesis shall primarily explore the current research regarding the implementation of modifiable microscopic and nanoscopic polymer particles as novel pharmaceutical delivery systems. Polymethacrylate-based copolymers, such as EudragitTM, are sensitive to alterations in pH levels, becoming increasingly more soluble in response to an increase in pH. The selective solubility and modifiable nature of these polymers allows for greater flexibility of treatment options for patients (Patra et al., 2017). In this study, a hyperbranched polyester co-polymer was designed using a proprietary monomer and diethylene glycol or triethylene glycol as monomers. The synthesis was carried out using standard melt polymerization technique and catalyzed by p-Tolulenesulfonic acid. The progress of the reaction was monitored with respect to time and negative pressure, with samples being subjected to standard characterization protocols. The resulting polymers were purified using the solvent precipitation method and characterized using various chromatographic and spectroscopic methods including GPC, MALDI-TOF, and NMR. The major part of this dissertation discusses the engineering of the refractive index of materials using solution-processable polymer nanocomposites and their applications in building optical components and devices. Three particular polymer nanocomposites have been introduced to achieve materials with tunable refractive indices and enhanced optical properties, which can be used to manipulate the behavior of light or electromagnetic radiations. In the first system, polyhedral oligomeric silsesquioxane (POSS)/polymer nanocomposites are developed. Thin films with tunable, low refractive indicies were fabricated from the composites. Advances in the synthetic strategies of engineered nanomaterials, multifunctional molecules and polymers have opened pathways for the development of functional nanomaterials having unique optoelectronic, mechanical, and biological properties. By designing the chemistry of surface ligands, the organic interface of nanoparticles, one can further the versatility and utilization of engineered nanomaterials, opening pathways for breakthroughs in sensing, catalysis, and delivery using nanomaterials. Encapsulation of materials can be performed through the stabilization of fluid-fluid interfaces and the formation of emulsion droplets, which is commonly achieved with surfactants, including small molecules and polymers, as well as particles that are, typically, micron-scale in diameter. The worked contained in this dissertation centered on droplets that are stabilized by nanoparticles, including metallic nanoparticles and semiconductor quantum dots, which bring the conductive and fluorescent properties inherent to such nanoparticles into the droplet construction. Double emulsion droplets, both oil-in-water-in-oil (o/w/o) and water-in-oil-in-water (w/o/w) were formed using nanoparticles as the only surfactant in solution. Conjugated semi-conducting polymers have become well known for their potential applications in hybrid electronic devices like solar cells, LEDs, and organic displays. These hybrid devices also contain inorganic nanoparticles, which complement the polymer when they are combined into the same layer. Control over the conformation and crystallinity of the polymer is critical for device performance, yet not much is known about the effect that these nanoparticles have on the polymer. Here, zinc oxide nanowire was surface modified with mono-substituted-carboxylic acid tetraphenylporphyrin and dodecanethiol, and introduced to poly(3-hexyl thiophene) in solution. This dissertation is related to the studies of functionalized nanoparticles for self-assembly and as controlled drug delivery system. The whole topic is composed of two parts. In the first part, the research was conducted to design and synthesize a new type of ionic peptide-functionalized copolymer conjugates for self-assembly into nanoparticle fibers and 3D scaffolds with the ability of multi-drug loading and governing the release rate of each drug for tissue engineering. The self-assembly study confirmed that such peptide-functionalized amphiphilic copolymers underwent different self-assembly behavior.
