Per- and polyfluoroalkyl substances (PFAS) are anthropogenic compounds developed for various applications; some are connected to adverse health impacts including immunosuppression and higher susceptibility to some cancers. Current PFAS remediation treatments from aqueous sources include granular activated carbon (GAC) adsorption, membrane separation, and anion- exchange resin (AER) removal. Each has specific disadvantages, hence the need for a new and efficient technology. Herein, acrylamide-based hydrogel composites were synthesized with powdered activated carbon (PAC) and characterized to determine their affinity for PFAS. Additive manufacturing, more commonly known as 3D-printing, is bringing about a grand technological transformation in material design and fabrication. In contrast to traditional fabrication techniques, 3D-printing features rapid prototyping, complex and customized 3D structural designs, and multi-material integration. Among current 3D-printing systems, direct-ink-writing (DIW) exhibits unique functionalities due to its mild operation conditions and the ability to integrate various organic and inorganic entities. Although supramolecular binding motifs are increasingly utilized in 3D-printing designs, the optimization of 3D-printable inks remains largely empirical. Gellan gum (GG) has gained considerable attention in the food, chemical, and pharmaceutical industries due to its functional characteristics. It has versatile properties, such as water solubility, easy bio-fabrication, good film/hydrogel-formation, biodegradability, and biocompatibility. These properties render GG a promising material in biomedical applications, specifically in the development of wound dressing materials. In this review, the use of GG biopolymer as a wound dressing material was discussed. Drug delivery systems (DDS) have highly evolved in the last decades with the development of hydrogels and nanoparticles. However, high systemic uptake, side effects, low bioavailability, and encapsulation efficiency continue to be a major hurdle faced by such DDSs.
Nanoparticles and hydrogels can be specifically designed for targeted DDSs to mitigate some of the problems. This dissertation aimed to design two DDSs for ocular drug delivery and one for cancer treatment. The first project sought to develop chitosan nanoparticles (Cs-NP) using PEGDA as a copolymer to encapsulate gentamicin (GtS) for ocular drug delivery. Carbohydrate based low molecular weight gelators have been an intense subject of study over the past decade. The self-assembling systems built from natural products have high significance as biocompatible materials and renewable resources. The versatile structures available from naturally existing monosaccharides have enriched the molecular libraries that can be used for the construction of gelators. The bottom-up strategy in designing low molecular weight gelators (LMWGs) for a variety of applications has been adopted by many researchers. Here, we present a systematic approach to design robust hydrogel compositions. The goal was to achieve excellent properties to open up new opportunities in especially two challenging applications not previously well-served by hydrogels.
The first part of this research will discuss the systematic screening of polymers for their hydrothermal stability under brine, acidic, neutral, and basic pH conditions followed by novel designs of hydrogel compositions. Certain hydrogel compositions withstand temperatures of up to 150 °C for more than 24 months in high salinity brines.
