We report a biochemically based EPR experiment for undergraduate laboratories that will serve as instruction for learning EPR sample preparation, spectral observation, and spectral analysis. There is a current lack of EPR instructional experiments that present complex spectra in which computational analysis may be required. The biochemical oxidation system of horse radish peroxidase (HRP) activated by H2O2 acts on 2,6-di-t-butyl-4-hydroxyanisole (DTBHA) to form phenoxyl radicals. This experiment produces moderately complex spectra of moderately long-lived radicals using relatively small amounts of readily available reactants. Electron Paramagnetic Resonance (EPR) analysis requires resonators optimized for specific experimental challenges. Volume resonators enclose samples, while surface coil resonators allow imaging of larger subjects. For in vivo studies, L-band frequencies are commonly used because they offer a good balance between signal-to-noise ratio (S/N) and the ability to penetrate tissue. Rapid Scan (RS) EPR at L-band frequencies was chosen for my studies of resonators because of this balance but presents a challenge for sensitivity.

The spectrum of Cu2+ has 4 lines, since I = 3/2. Cu has two major isotopes, 63Cu (69.2%) and 65Cu (30.8%). The magnetic moment of 65Cu is 7% larger than that for 63Cu, so the hyperfine coupling is 7% larger. If the lines are narrow enough, one can observe superposition of spectra due to complexes of the two isotopes.

Local tissue physiology is an important parameter in understanding disease behavior. Rapid Scan (RS) electron paramagnetic resonance (EPR) offers a unique, non-invasive tool for investigation of these so-called microenvironments through EPR Imaging (EPRI). Research into advancement of EPRI falls into many categories. Not least among those are advances in instrumentation and methodology. Presented here are updates to a benchtop EPRI instrument operating at 1 GHz targeted at pre-clinical EPRI applications. Newly developed methods for reducing RS-EPR background through inversion of the magnetic field are also demonstrated. Alpha-Synuclein (αS) – a neuronal, disordered, presynaptic protein – aggregates into amyloid fibrils and accumulates in the substantia nigra pars compacta of Parkinson's Disease (PD) patients. The aggregation and accumulation of αS amyloid fibrils leads to death of dopaminergic neurons; a hallmark of PD. Although it’s not clear why αS aggregates, prior studies have found that intrastriatal injection of fibril alone is sufficient to cause PD pathology in mouse and non-human primates models. These observations implicate αS as a therapeutic target against PD.

Trityl and nitroxide radicals are widely used as probes in chemical and biochemical systems. Understanding of factors that impact the electron spin relaxation for these radicals is important for selection of probes and experimental operating conditions. Enhanced signal sensitivity by the use of Rapid Scan (RS) electron paramagnetic resonance (EPR), a technique that allows for much faster magnetic field scans than traditional field-swept techniques, has facilitated improved data acquisition for many types of samples. For example, irradiated fingernails for radiation dosimetry have been studied using RS-EPR, which resulted in substantial decreases in detection limits. Samarium-mediated reduction mechanisms in organic synthesis have been investigated by RS-EPR providing evidence for a radical intermediate. Rapid scan electron paramagnetic resonance (EPR) was developed in the Eaton laboratory at the University of Denver. Applications of rapid scan to wider spectra, such as for immobilized nitroxides, spin-labeled proteins, irradiated tooth and fingernail samples were demonstrated in this dissertation. The scan width has been increased from 55 G to 160 G. The signal to noise (S/N) improvement for slowly tumbling spin-labeled protein samples that is provided by rapid scan EPR will be highly advantageous for biophysical studies. Variations in the physical deformation of the plasma membrane play a significant role in the sorting and behavior of the proteins that occupy it. Determining the interplay between membrane curvature and protein behavior required the development and thorough characterization of a model plasma membrane with well defined and localized regions of curvature. This model system consists of a fluid lipid bilayer that is supported by a dye-loaded polystyrene nanoparticle patterned glass substrate.
