Cucurbitacins (CUCS) are natural products with highly oxygenated tetracyclic triterpenes produced mostly by Cucurbitaceae family plant. They are known for their therapeutic efficiency with different biological activities, such as anti-inflammatory, hepatoprotective and anti-cancer. Hepatocellular carcinoma (HCC) is the third leading cause of death worldwide. Previous reports have shown the ability of CUCS to inhibit the growth of hepatocellular carcinoma cell lines (HepG-2) significantly. Photochemical Generation Of Carbenes And Ketenes From Phenanthrene-Based Precursors Part I: Dimethylalkylidene Part Ii: Diphenylketene, Tarini S. Hardikar, Tarini Hardikar

Previous studies have shown that the photolysis of phenanthrene-based cyclopropyl precursors can generate carbenes. In this work, a novel phenanthrene precursor was synthesized and photolyzed to generate the dimethylalkylidene carbene, which was subsequently trapped by cyclohexene. The car- bene was not prone to an internal rearrangement as some other carbenes are, and instead favored the intermolecular addition reaction. The precursor also underwent a rearrangement via pericyclic processes. A theoretical analysis of the potential energy surface of the system is presented using coupled cluster and density functional methods. Preliminary analysis for the precursor via time-resolved laser flash photolysis (LFP) was performed. …

Dihydrofurans serve as building blocks for other compounds in organic synthesis. The main goal of this project was to discover an efficient and relatively inexpensive pathway for producing monosubstituted dihydrofurans in high yield from cyclic boronic half acids. Aldehydes were converted to homoallylic alcohols by the addition of allylmagnesium bromide. The alcohols were then transformed into cyclic boronic half acids using ringclosing metathesis with Grubbs 1st Generation Catalyst and alkenyl boronic esters. Finally, monosubstituted dihydrofurans were produced using a palladium (II) catalyst with a base. The receptor of advanced glycation end product (RAGE) is a multiligand receptor of the immunoglobulin superfamily of cell surface molecules, which plays an important role in immune responses. Full-length RAGE includes three extracellular immunoglobulin domains, a transmembrane domain and an intracellular domain. It is a pattern recognition receptor that can bind diverse ligands. NMR spectroscopy and x-ray crystallization studies of the extracellular domains of RAGE indicate that RAGE ligands bind by distinct charge- and hydrophobicity-dependent mechanisms. Synthetic access to systematically substituted tetrafluoro-λ6-sulfanes was challenging and the research in this area had been largely abandoned. The original reports of disubstituted-tetrafluoro-λ6-sulfanes were limited to substitution by diphenyl and dialkyl groups. The dialkyl-tetrafluoro-λ6-sulfanes were reactive while the diphenyl substituted compounds showed decreased reactivity. In this dissertation, the systematic substitution on tetrafluoro-λ6-sulfanes has enabled the studies of the substitution effects on reactivity.

The oxidative stability of ultra-low sulfur diesel (ULSD) has generally been expected to continuously decline over time. However, recent studies have suggested that it may fluctuate.1-2 In this study, the oxidative stability of stored commercially purchased ULSD was monitored with a methanol extraction method.3 In this methodology, proposed by Hardy and Wechter, fuel blends are extracted with methanol before and after thermal stressing.4 The methanol-soluble layer contains the oxygenated and oxidizable components of the fuel (SMORS) and the change in this mass upon stressing thus represents the oxidative stability of …

The iodine-initiated hydration of internal alkynes is a novel, green, and inexpensive synthetic pathway in comparison with the commonly used transition metal catalyzed reactions. Hemoglobin based oxygen carriers (HBOCs) hold promise as an effective emergency treatment of severe traumatic brain injuries (TBI). In the latest generation of HBOCs, polynitroxyl-pegylated hemoglobin (PNPH), cell-free hemoglobin is modified with TEMPO and PEG which reduce the toxicities associated with earlier generations of HBOCs. In our efforts to optimize the economic and therapeutic impacts of PNPH’s we have synthesized polydimethylaminoethyl methacrylate (poly-DMAEMA) under controlled living conditions via reverse addition-fragmentation chain transfer (RAFT) polymerization. In this thesis, we investigate biological molecules on a micron scale in the ultraviolet spectral region through the non-destructive confocal absorption microscopy. The setup involves a combination of confocal microscope with a UV light excitation beam to measure the optical absorption spectra with spatial resolution of 1.4 μm in the lateral and 3.6 μm in the axial direction. Confocal absorption microscopy has the benefits of requiring no labels and only low light intensity for excitation while providing a strong signal from the contrast generated by the attenuation of propagating light due to absorption. Liver X receptors (LXRs) have been increasingly recognized as a potential therapeutic target to treat pathological conditions ranging from vascular and metabolic diseases, neurological degeneration, to cancers that are driven by lipid metabolism. Amidst intensifying efforts to discover ligands that act through LXRs to achieve the sought-after pharmacological outcomes, several lead compounds are already being tested in clinical trials for a variety of disease interventions. Many materials used for storage of museum artifacts have been found to cause damage to the items either through emitting harmful volatile gases or through direct contact. Harmful storage techniques can cause corrosion, discoloration, or deterioration of priceless artifacts. Museum enclosures made of hardwoods have been shown to emit acetic acid vapors that cause damage to the items being stored. In the present study, a review of the effects of acetic acid damage on museum artifacts was performed, and an evolved gas analysis and titration method were used to quantify acetic acid in various hardwoods.

Currently, perovskite solar cells (PSCs) are the most rapidly developing type of solar cells with certified efficiencies rising from 3.8% in 2009 to 22.1% in 2016. Alanine scanning mutagenesis was employed to determine the structural epitopes of preptin; a newly discovered polypeptide found on the proinsulin-like growth factor II. The alanine scanning mutagenesis was accomplished by the use of, cloning, affinity chromatography, gel electrophoresis, gene sequencing, and mass spectra. Flavonoids are highly luminescent compounds that are used in diagnosing and treating certain diseases.They are composed of two aromatic rings (ring A and ring B) and a six-membered heterocyclic pyran ring (ring C) that contains a carbonyl and hydroxyl group, which are responsible for unique photophysical characteristics associated with flavonoids. The target flavonoids (compound 1, or 1, and compound 2 or 2) were synthesized, purified and characterized by NMR spectroscopy. Parkinson’s disease (PD) is a neurodegenerative disorder characterized by fibrillar cytoplasmic aggregates of α-synuclein (i.e., Lewy bodies) and the associated loss of dopaminergic cells in the substantia nigra. Mutations in genes such as α -synuclein (SNCA) account for only 10% of PD occurrences. Exposure to environmental toxicants including pesticides and metals (e.g., paraquat (PQ) and manganese (Mn)) is also recognized as an important PD risk factor. Thus, aging, genetic alterations, and environmental factors all contribute to the etiology of PD. Atmospheric aerosols play an important role in climate by scattering and absorbing radiation and by serving as cloud condensation nuclei. An aerosol’s optical or nucleation properties are driven by its chemical composition. Chemical aging of aerosols by atmospheric oxidants, such as ozone, alters the physiochemical properties of aerosol to become more hygroscopic, light absorbing, and viscous during transport. However the mechanism of these transformations is poorly understood. With the dwindling availability of petroleum, focus has shifted to renewable energy sources such as lignocellulosic biomass. Lignocellulosic biomass is composed of three main constituents, lignin, cellulose and hemicellulose. Due to the low value of cellulosic ethanol, utilization of the lignin component is necessary for the realization of an economically sustainable biorefinery model. Once depolymerized, lignin has the potential to replace petroleum-derived molecules used as bulk and specialty aromatic chemicals. Lolines are specialized metabolites produced by endophytic fungi, such as Neotyphodium and Epichloë species, that are in symbiotic relationships with cool-season grasses. Lolines are vital for the survival of the grasses because their insecticidal and antifeedant properties protect the plant from insect herbivory. Although lolines have various bioactivities, they do not have any concomitant antimammalian activities.

Lolines have complex structures that are unique among naturally occurring pyrrolizidine alkaloids. Polycyclic polyprenylated acylphloroglucinols (PPAPs) are plant- (Guttiferae) derived natural products. They have fascinating bicyclo[3.3.1]nonane-2,4,9-trione or [3.2.1]nonane-2,4,8-trione cores decorated with prenyl or geranyl groups. More than 200 PPAPs have been isolated, but only a few of them have been synthesized, although most of the synthesized PPAPs are of type A and have an exo substituent at C (7). The introduction of fluorine uniquely affects the biological properties of organic molecules such as making them more bioavailable, lipophilic and metabolically stable, and possibly increase the strength of a compound’s interactions with a target protein. Approximately 30% of agrochemicals and 20% of pharmaceuticals contain fluorine, including commonly prescribed drugs such as Lipitor, Lexapro and Prozac. The focus of this dissertation is the study of alkene hydroaylation reactions. In the first chapter, a review of the relevant literature concerning zinc- and iron-catalyzed hydroarylation of unsaturated bonds is presented. In chapter two, the development of a zinc-catalyzed hydroarylation of alkenes is presented, which is quickly followed by a detailed investigation into the probable mechanism. Chapter three discusses the development of an iron/silver-catalyzed arene prenylation reaction, followed by the development of an iron/zinc-catalyzed arene prenylation reaction. Organic semiconductors have gained a lot of interest due to their ease of processing, low-cost and inherent mechanical flexibility. Although most of the research has been on their electronic and optical properties, knowledge of the thermal properties is important in the design of electronic devices as well. Our group has used ac-calorimetric techniques to measure both in-plane and transverse thermal conductivities of a variety of organic semiconductors including small-molecule crystals and polymer blends. The use of porous sorbents for physisorptive capture of CO2 from gas mixtures has been deemed attractive due to the low energy penalty associated with recycling of such materials. Porous organic polymers (POPs) have emerged as promising candidates with potential in the treatment of pre- and post- fuel combustion processes to separate CO2 from gas mixtures. Concurrently, significant advances have been made in establishing calculation methods that evaluate the practicality of porous sorbents for targeted gas separation applications. However, these methods rely on single gas adsorption isotherms without accounting for the dynamic gas mixtures encountered in real-life applications. …
