  • β-lactams are essential structural components of many antibiotics and widely studied due to their biological importance and synthetic versatility.
  • The Ugi reaction is a multicomponent reaction, involving a primary amine, carboxylic acid, isocyanide, and an aldehyde or a ketone, that rapidly generates α-acetamido carboxamide derivatives (Ugi adducts).
  • The synthesis and characterization of four oxindole-based β-lactam derivatives:
    • 1-Benzyl-N-(tert-butyl)-2-oxo-3-(2-oxoazetidin-1-yl)indoline-3-carboxamide (4a).
    • N-(tert-butyl)-2-oxo-3-(2-oxoazetidin-1-yl)-1-phenylindol-3-carboxamide (4ab).
    • N-(tert-Butyl)-2-oxo-3-(2-oxoazetidin-1-yl)indoline-3-carboxamide (4d).
    • N-(tert-butyl)-1-methyl-2-oxo-3-(2-oxoazetidin-1-yl)indol-3-carboxamide (4e).

The continued rise of antibiotic-resistant bacteria, such as MRSA (methicillin-resistant Staphylococcus aureus), has made the discovery of novel antibiotics critical. While bacteriostatic and bactericidal antibiotics inevitably exert evolutionary pressure on bacteria to develop resistance, small molecules that target mechanisms of bacterial virulence present a promising alternative for treatment. Sortase A (Srt A), a cysteine protease of S. aureus, promotes bacterial virulence by covalently attaching proteins such as pilin to the bacterial surface, enabling bacterial adhesion to mammalian cells. Alkenes serve as one of the most important feedstocks for organic synthesis, having two vicinal sites for bond formation. In alkenes, both vicinal sites can be functionalized with two reagents in a process commonly known as alkene difunctionalization, which results in the formation of two new bonds. A number of alkenes difunctionalization reactions, such as diamination, dioxygenation, carboamination and carbooxygenation, are known. However, difunctionalization of alkenes with two carbon-based entities, termed alkene dicarbofunctionalization, is relatively less common. An improved synthesis of the antiviral drug adefovir is presented. Problems associated with current routes to adefovir include capricious yields and a reliance on problematic reagents and solvents, such as magnesium tert-butoxide and DMF, to achieve high conversions to the target. A systematic study within our laboratory led to the identification of an iodide reagent which affords higher yields than previous approaches and allows for reactions to be conducted up to 10 g in scale under milder conditions. The selective oxidation of organic compounds represents a leading technology for chemical industries. They are used in chemical synthesis in the pharmaceutical and petrochemicals industries, and possible the decontamination of harmful substances. However, oxidations reaction are among the most challenging processes to control. Many stoichiometric oxidants with heavy metals are expensive, or toxic maybe both, and therefore unfeasible to be utilized. HER2 overexpression in breast cancer tumors predicts lower overall survival. Because of the aggressive nature of HER2 tumors and the association with metastatic disease, the HER2 receptor holds great promise as a therapeutic target in metastatic breast cancer. We are developing small molecule inhibitors that bind to the ATP binding site of the tyrosine kinase domain in order to inhibit tyrosine auto-phosphorylation. This process controls biological pathways that mediate the cell growth. In normal cells this process is highly controlled. Highly reactive transition metal-oxo intermediates are important active oxidant involved in numerous enzymes such as cytochrome P450 monooxygenases as well as in many useful metal-catalyzed oxidations. Many transition metal catalysts are designed for biomimetic studies of the predominant oxidation catalysts in Nature, the cytochrome P450 enzymes. In this work, a series of metalloporphyrin and metallocorrole complexes have been successfully synthesized and spectroscopically characterized by UV-vis, GCMS and 1H-NMR. The utilization of these complexes as catalysts for selective oxidation of sulfides and photocatalytic aerobic oxidations of activated hydrocarbons were investigated.
