K+ channels containing Kv1.1 α subunits, which become prevalent at internodes in demyelinated axons, may underlie their dysfunctional conduction akin to muscle weakness in multiple sclerosis. Small inhibitors were sought with selectivity for the culpable hyper-polarizing K+ currents. Modeling of interactions with the extracellular pore in a Kv1.1-deduced structure identified diaryldi(2-pyrrolyl)methane as a suitable scaffold with optimized alkyl ammonium side chains. The resultant synthesized candidate [2,2′-((5,5′(di-p-topyldiaryldi(2-pyrrolyl)methane)bis(2,2′carbonyl)bis(azanediyl)) diethaneamine·2HCl] (8) selectively blocked Kv1.1 channels (IC50 ≈ 15 μM) recombinantly expressed in mammalian cells, induced a positive shift in the voltage dependency of K+ current activation, and slowed its kinetics. Laser induced breakdown spectroscopy (LIBS) is an emerging technique in the field of food analysis which provides various advantages such as minimal sample preparation, chemical free, rapid detection, provision of spatial information and portability. In this study, LIBS was employed for quantitative analysis of copper content in minced beef samples spiked with beef liver over three independent batches. Copper content was determined with graphite furnace atomic absorption spectroscopy (GFAAS) in order to obtain reference values for modelling. Oral vaccines present an attractive alternative to injectable vaccines for enteric diseases due to ease of delivery and the induction of intestinal immunity at the site of infection. However, susceptibility to gastrointestinal proteolysis, limited transepithelial uptake and a lack of clinically acceptable adjuvants present significant challenges. A study of spatial variability of PM10 elemental components was conducted in Terni city (Central Italy), situated in an intramountain depression characterized by the presence of several particulate matter emission sources. The meteorological conditions of the Terni basin limit the dispersion and enhance the accumulation of atmospheric pollutants. Thanks to the utilization of new smart samplers, used for the first time and working in parallel at 23 sampling sites, spatially resolved data were obtained. In this study, a 150 mJ laser-induced breakdown spectroscopy (LIBS) system was assessed to determine calcium content in infant formula (IF) samples. LIBS is a promising emission spectroscopic technique for elemental analysis, which offers advantages over conventional methods such as real-time analyses, little to no sample preparation and ease of use. Circulating tumor cells (CTCs) from peripheral blood account genetic information for cancer diagnosis and overall disease monitoring. Analysis of “liquid biopsy” holds immense promise as it may lead to new approaches for cancer treatment. The study reports effective and continuous flow microchannel system for isolating CTCs using transferrin conjugated 3D matrix synthesized by crosslinking polyethylene glycol-Fe3O4 nanostructures for rapid and efficient capturing of CTCs. The platform provides option of using multiple microchannel units in series that can influence higher cell-capture efficiency due to increasing cell-substrate contact frequency. Orthogonally protected thioamide-containing dipeptides were efficiently and cleanly prepared from the precursor dipeptides using Curphey’s method (P4S10, hexamethyldisiloxane (HMDO), reflux, DCM) in 67-96% isolated yield. This was in contrast to the use of Lawesson’s or Berzelius’ reagents where significant issues with reaction non-completion, decomposition and purification were observed. Subsequent clean removal of the dipeptides’ t-butyl ester protecting groups gave thioamide dipeptide acids which were suitable for use in solid-phase peptide synthesis (SPPS).

Vibrational spectroscopic techniques can detect small variations in molecular content, linked with disease, showing promise for screening and early diagnosis. Biological fluids, particularly blood serum, are potentially valuable for diagnosis purposes. The so-called Low Molecular Weight Fraction (LMWF) contains the associated peptidome and metabolome and has been identified as potentially the most relevant molecular population for disease-associated biomarker research. The in vitro cytotoxic and intracellular oxidative stress responses to exposure to poly (propylene imine) (PPI) dendritic nanoparticles of increasing generation (number of repeated branching cycles) (G0-G4) were assessed in an immortal non-cancerous human keratinocyte cell-line (HaCaT). Confocal fluorescence microscopy with organelle staining was used to explore the uptake and intracellular trafficking mechanisms. A generation and dose dependent cytotoxic response was observed, increasing according to generation and therefore number of surface amino groups. Orthogonally protected azalanthionines were successfully synthesised by the ring-opening of N-activated aziridine-2-carboxylates with protected diaminopropanoic acids (DAPs). The required DAPs were also prepared by ring-opening of N-activated aziridine-2-carboxylates with para-methoxybenzylamine, but it was found that the choice of aziridine protecting groups dictated both the success of the reaction as well as the regioselectivity of the isolated products. Attempts to extend the methodology to the preparation of the more sterically demanding b-methyl azalanthionines have, so far, been unsuccessful.

Electrochemical copolymerization of pyrrole (Py) and 3,4-ethylenedioxythiophene (EDOT) was performed in an ionic liquid 1-butyl-3-methylimidazolium tetrafluoroborate, (BMIM BF4) employing a ‘micro-cell’ in order to use the materials efficiently. Characterization of the copolymers formed on an ITO substrate was performed for different Py: EDOT ratios (1:2, 1:1, 2:1) in both aqueous and ionic liquid electrolytes. Electrochemical and spectroelectrochemical analysis revealed that the properties of the copolymer depend upon the Py/EDOT monomer ratio used. The copolymer PPy-co-PEDOT (2:1) displayed good stability, a switching time of 20 s and a colouration efficiency of 101.3 C-1cm2. The presence of Py in the copolymer stabilised …

Conventional anodising electrolytes based on sulphuric acid, oxalic acid and phosphoric acid have been used to form nanoporous layers on AA3003-H14 and sealed with silane based sol-gel sealers. It is shown that the sol-gel chemistries have varying levels of pore penetration depending on the synthesis conditions. The extent of sol-gel penetration and pore sealing is analysed by electron microscopy and energy dispersive x-ray spectroscopy. To describe the sealing phenomena observed a sol-gel penetration and sealing rating has been proposed to explain the interactions of the sol-gels with the pores of the anodised layers. The synthesis of zinc oxide through three routes was investigated; carboxylic acid chelation, polymer assisted synthesis in an alcoholic medium and slow thermolysis of precursor. In the case of carboxylic acid chelation, it was determined that the rate at which the precursor is formed has a significant effect on the quality of zinc oxide particles produced.
