Exploring Ancestral Enzymes Through Sequence Reconstruction And Stability Prediction, Jeyun Park, Masakatsu Watanabe

Proteins evolve through sequence changes that shape structure, stability,

and function. Ancestral sequence reconstruction (ASR) infers ancestral

proteins from modern homologs, but many studies focus on sequence

inference without evaluating structural or energetic feasibility.

β-lactamases provide an ideal model due to their evolutionary diversity and

clinical relevance in antibiotic resistance.

Here, we present an integrated ASR workflow combining phylogenetic

inference (IQ-TREE), ancestral reconstruction, and structural and stability

validation using AlphaFold and FoldX. By reanalyzing a curated β-

lactamase dataset and comparing with a recent study (Risso et al., 2013),

The spread of antibiotic-resistant bacteria poses a global health threat, and it is predominantly driven by enzymes that neutralize antibiotics or alter their drug targets. This study uses Ancestral Sequence Reconstruction (ASR) to explore the evolutionary history of a key bacterial enzyme that plays a role in antibiotic resistance. Protein sequences were aligned, and a phylogenetic tree constructed utilizing MEGA 11. Ancestral sequences are predicted to identify the conserved regions that are essential for enzyme function. Understanding these conserved locations provides insights into potential drug targets for developing new drugs against resistant strains of bacteria.

Unlocking The Secret To Weight Loss: Discovering The Most Effective Green Tea Products, Seoyeon Kim

To find the most effective green tea product that can replace

weight loss drugs, we compared the amount of catechin in six

different green tea products. The result was green tea latte

powder contained a significantly small amount of catechin

compared to other products, and the dried pearl green tea leaves

had the highest amount of catechin. Also, the unexpected result

was that green tea supplements had less amount of catechin

compared to dried leaves or even tea bags that are commonly

sold in markets.
