Effects Of Bidisperse Gold Nanoparticles On Near-Infrared Surface-Enhanced Raman Spectroscopy (Sers) In Quick-Freezing-Induced Gold Nanoparticle Aggregates (Qfiaas), Elissa M. Dojka

Gold nanoparticle (AuNP) aggregates formed via rapid freezing in liquid nitrogen exhibit strong near-infrared (NIR) plasmonic coupling. These quick-freezing-induced AuNP aggregates (QFIAAs) form moderately sized, colloidally stable structures that remain suspended for over three months, supporting a variety of analytical applications. Mechanistically, QFIAA formation arises from the confinement of nanoparticles and ions within advancing ice grain boundaries during freeze-concentration. This mechanical crowding promotes extreme physical proximity while simultaneously elevating local ionic strength, which screens electrostatic barriers to drive the assembly of dense, tightly coupled plasmonic networks.

The main focus of this dissertation was to apply enhanced Raman spectroscopy techniques to a variety of applications, ranging from drug discovery, mRNA vaccine stability, to forensic analysis. There are five chapters in this dissertation. The first chapter discusses the basics of enhanced Raman spectroscopy techniques and their relevance. The second chapter is focused on the use of surface-enhanced Raman spectroscopy (SERS) for drug discovery. The third chapter introduces the use of deep ultraviolet resonance Raman (DUVRR) spectroscopy for the detection of mRNA degradation in a mRNA vaccine model. In this dissertation are presented three projects that contribute to the body of research on SERS in the forensic, heritage, and semiconductor fields. The first project, Charge-Transfer mapping on GaN/Ag, a silver-decorated nanopillar semiconductor substrate fabricated from the GaN is used with the Raman probe Rhodamine 6 G (R6G) to map the effect of the nanofeatures on the CT resonance. The second project, in collaboration with Marco Leona from the Metropolitan Museum of Art, explores the use of AgNIFs to identify colorants used on textile fiber samples from four 19th century works of Japanese art. Monitoring chemical reactions in aqueous solution is a challenge because most instrumental techniques either are not suited for the rapid timescales, are not sensitive enough to detect products at low concentrations, or do not have sufficient structure-to-spectrum relationships. Raman spectroscopy is a promising method to monitor reactions, as it is fast, dependent on chemical structure, and has little interference from water. Near-IR (NIR) surface-enhanced Raman scattering (SERS) is an emerging bioimaging technique that can be a safe alternative with a resolution better than current bioimaging techniques, such as MRI, CT, and PET. These can be hazardous with long-term exposure due to powerful magnetic fields or ionizing radiation. The success of NIR SERS bioimaging lies partially in the selection of highly sensitive NIR Raman reporters, which is essential to achieve highly resolved bioimaging of the body system. Surface enhanced Raman spectroscopy (SERS), although popular in research, has not yet been adopted in industrial applications. This is largely due to the challenge of manufacturing cost-effective, reproducible SERS substrates in volume that can be read by a compact reader for practical deployment in the field. Here we demonstrate and characterize a new dual-nanostructured SERS substrate approach that could be amenable to large-scale production. Raman spectroscopy is an effective tool for detecting trace amounts of material by fingerprint-like vibrational spectra. At times, the weak intensity of Raman scattering can make it difficult to distinguish trace materials. This shortcoming is addressed by surface‐enhanced Raman spectroscopy (SERS), which produces strong signal enhancements when target compounds are near metal nanoparticles. For the first part of this thesis, the identification of fentanyl and carfentanil, main culprits in the opioid epidemic, was done using normal Raman and the SERS spectroscopy. A SERS device was made using a dual-nanostructured surface comprised of silver nanoparticle and silver nanowires. The ability of each nanostructure to produce a uniform surface was characterized and the surface-enhanced Raman scattering (SERS) response of the resulting surfaces were examined using the reporter molecule 4-aminothiolphenol (ATP) and a 638 nm excitation laser.

A synthetic method was developed to produce silver nanowires with lengths of ~20 μm and diameters of ~100 nm with a narrow size distribution. The method utilized a simple, one-pot synthesis that is amenable to large-scale production. A selective precipitation method was used to the isolate the …

In situ SERS and ATR-SEIRAS were applied to investigate the electrochemical and structural properties of iron protoporphyrin IX (FePP) self-assembled monolayers (SAMs) on Pt electrodes in 0. 1 mol· L -1 HClO4. The potential dependent SERS spectra of the FePP SAM on a roughened Pt electrode with the enhancement factor of ca. 40 were acquired with the excitation line of 514 nm. Analyses of SERS data over the potential region from 0. 5 to -0. 3 V enabled to obtain potential dependent adsorption isotherms,from which the formal redox potential could be estimated to be ca. -0. 2 V. …

Experimental and theoretical results are presented in Chapters 2, 3, and 4 on the effects of dielectric medium, particle size, and interparticle distance, respectively, on plasmon coupling in two-dimensional arrays of silver nanoparticles. The arrays were fabricated via the self-assembly of single crystal particles from c.a. 46 nm to 287 nm on PVP-modified glass substrates. Spin-coated poly(methylmethacrylate) layers were used to immobilize the particles and prevent their surface aggregation. Varying the thickness of the PMMA layer made it possible to change the average dielectric medium between the particles. This work studied the potential difference and galvanic currents on particulate contamination of the abrasive on the patterned surface and on corrosion behaviors of TiN and W. The polarization curves of TiN and W were, respectively, obtained in 0.01mol/L KNO3 solutions in the absence and presence of three typical oxidants (H2O2, KIO3, Fe(NO3)3) by DC polarization technique. Two-dimensional arrays of Cu and Au nanowires have been fabricated by template synthesis method, using the anodic aluminum oxidation films as the template. The morphology of the nanowire array was monitored by atomic force microscope (AFM) in the dissolution processes of the alumina film. Based on the probe molecule strategy, the spectral feature of SCN- adsorbed at the Cu and Au nanowires with different diameters has been investigated by surface enhanced Raman spectroscopy (SERS).
