RNA therapeutics are a rising drug category with potential use for a range of conditions encompassing infectious diseases to therapies for cancer, diseases, and genetic disorders. This dissertation uncovers the ultrafast molecular mechanisms underlying the decomposition of nitrate ester explosives—nitroglycerin (NG), ethylene glycol dinitrate (EGDN), and amyl nitrate—through the integration of femtosecond time-resolved mass spectrometry and advanced computational chemistry. Although these compounds play fundamental roles in both military and civilian contexts, the precise molecular dynamics governing their pronounced reactivity and impact sensitivity have remained inadequately characterized. Our research establishes that low O–NO2 bond dissociation energies are the primary driver of ultrafast fragmentation and high reactivity in all three esters. Shock initiation of energetic materials produces cations and anions that can contribute to initial endothermic chemical reactions which lead to detonation. Hence, knowledge of the initial dissociation pathways of ionized energetic molecules is important for advancing the understanding of initiation mechanisms and meeting longstanding goals of the Department of Defense. Antibiotic-resistant bacteria pose a significant and escalating threat to hospitals around the world, necessitating the development of rapid and sensitive detection methods. Single-entity electrochemistry has emerged as a promising approach for detecting and identifying such bacteria, and monitoring the efficacy of antibiotics in real-time. Herein, we employ the translational diffusion equation for circular cylinders to predict the collision frequency of rod-shaped bacteria, informing our experimental setup. Our work demonstrates that lab-fabricated Pt ultramicroelectrodes can sensitively detect bacteria at femtomolar concentrations under migration-controlled conditions. Energetic materials and the unimolecular dissociation dynamics attributed to their initiation and subsequent bimolecular detonation have been a subject of interest for defense agencies such as the Department of Defense (DoD) for decades. The timescale in which these initial dynamics exist ranges from several picoseconds to dozens of femtoseconds, necessitating equally fast time-resolved experiments to capture and characterize these events. Our lab specializes in one such laser-based technique: femtosecond time-resolved mass spectrometry (FTRMS). Single Entity Electrochemistry (SEE) is an emerging electrochemical technique that has been used to characterize discrete entities by measuring the change in current or potential during individual stochastic events (collision or adsorption) of an entity with an ultramicroelectrode (UME) of similar dimensions. The shape and magnitude of the SEE signal depend on the underlying mechanism of interaction with the UME surface. There is a critical need for quantitative models that correlate the SEE signal with properties of the entity-UME system, including effects of acquisition instrumentation, to prevent misinterpretation of data.

While DNA breaks can be single-stranded or double-stranded, the double-stranded DNA (dsDNA) breaks are more dangerous. If such damage is not repaired, it can lead to genetic instability and serious health issues including cancers. According to the Centers of Disease Control and Prevention, antibiotics decrease in effectiveness as bacteria gain resistance for previously treatable illnesses. Currently, antibiotic susceptibility is typically carried out via the Kirby-Bauer method. Even with automation, this process requires two incubation periods so a less time-consuming technique is desirable. Current-time recordings of toluene microdroplets emulsified in water and containing 20 mM Ferrocene (Fc), show multiple electrochemical peaks from oxidation of Fc on disk microelectrodes (5μm-diameter). The average droplet diameter (~0.7 μm) determined from area integration of the peaks was close to Dynamic Light Scattering measurements (~1 μm). Random walk simulations were performed deriving equations to simulate droplet electrolysis using the diffusion and thermal velocity expressions established by Einstein. The simulations show that multiple droplet-electrode collisions, lasting ~0.11 μs each, occur before a droplet wanders away. Updating the Fc-concentration at every collision shows that a droplet only oxidizes ~0.58 % …

Single entity electrochemistry (SEE) is an emerging electroanalytical technique with the ability to push the limit of detection of electrochemical sensors to the single particle level. It measures the change in current over time, making SEE rapid, simple, and cost-effective. The type of current response observed in the scan can provide information about the physiochemical processes underlying the signal. Blood is often a type of evidence found at crime scenes. There has been a long history of criminal cases involving blood evidence samples that were allegedly planted at the scene by a third party. Ethylenediaminetetraacetic acid, or EDTA, is an anticoagulant that is not naturally occurring in blood samples; it chelates to ions in blood to prevent clotting. Characterization was done using scanning electron microscopy (SEM), X-ray photoelectron spectroscopy (XPS), and cyclic voltammetry (CV). In this work, the open circuit potential (OCP) of ferricyanide, cysteine, uric acid, and ascorbic acid was measured in phosphate buffer solution (PBS) and biofouling solution containing bovine serum albumin (BSA) at pH 7.4. Multicomponent synthetic chemical gradients containing amine (NH2) and carboxylic acid (COOH) groups have been prepared on the silanol (Si-OH) containing a base-layer substrate by the controlled-rate infusion (CRI) method. The spatial variation of weakly acidic COOH and basic NH2 functional groups on weakly acidic Si-OH surface was created in such a way that the two antagonist groups at high concentrations were in close proximity (aligned gradient) and were separated (opposed gradient). Before an IED is sent to a laboratory for analysis, it needs to be rendered safe if it did not already initiate. Render safe procedures (RSPs) include utilizing a percussion-actuated non-electric (PAN) disrupter or a mineral water bottle disrupter. Each disrupter utilizes explosives to render the device safe by breaking open the container or disrupting the fuzing system. However, the same explosives used in the RSP are also used by criminals in IED construction. Choosing a stationary phase is the first step in developing a liquid chromatography (LC) separation method, where the selectivity is governed by the differential interactions of the analytes with the stationary and mobile phases. The importance of porous materials has risen substantially in the last few decades due to their ability to reduce the size and cost of bioanalytical devices and fuel cells. First, this work aims to describe the fabrication of nanoporous gold (NPG) electrodes that are resistant to electrode passivation due to fibrinogen biofouling in redox solutions. The objective of the research was to investigate proton-coupled electron transfer (PCET) reactions involving antioxidants to gain insight into the detailed mechanisms of glutathione (GSH), Trolox, and α-tocopherol (α-TOH). PCET reactions are complex redox reactions that transfer electrons and protons sequentially or in concert. These reactions are ubiquitous in natural or artificial processes that produce electrochemical energy that is extractable as electricity or as chemical fuels of high energy content. Researchers are actively exploring several avenues to reduce TSNA levels in tobacco products like moist snuff tobacco. The method development process in liquid chromatography (LC) involves optimization of a variety of method parameters including stationary phase chemistry, column temperature, initial and final mobile phase compositions, and gradient time when gradient mobile phases are used. Here, a general simulation program to predict the results (i.e., retention time, peak width and peak shape) of LC separations, with the ability to study various complex chromatographic conditions is described. Gradient surfaces exhibit a variation in functionality along the length of the surface. One method for preparing gradients is controlled-rate infusion (CRI). In Part 1 of this work, CRI was used to prepare gradients for the purpose of separating transition and heavy metals. Initial work on this project was focused on controlling the retention of the metal ions by varying the number of amine groups, aminoalkoxysilane concentration, and the infusion time. “Kratom” is the common name for the botanical mitragyna speciosa. It is a tree native to Southeast Asia in which leaves contain the psychoactive alkaloids mitragynine and 7-hydroxymitragynine. Kratom is often ingested as teas, chewed, or smoked. It acts as a stimulant in small doses and as an opioid in large doses. Overdoses can result in vomiting, seizures, and death. Recently the Drug Enforcement Agency (DEA) placed Kratom on Schedule 1, but, due to public outcry, it was almost immediately removed.
Eleven kratom based products were obtained from various tobacco shops, “headshops” and via the internet including: Choice brand …
