In this project, water near factories in Rubbertown, Louisville, KY were tested to see if a significant enough amount of waste is deposited in the Ohio River that the concentrations of Pb2+ and Cu2+ exceed the maximum contaminant levels the World Health Organization (WHO) and the United States Environmental Protection Agency (EPA) have set in place. Waste from industrial factories may contain contaminants that have negative effects on aquatic ecosystems and even human health, so two potentially harmful metal ions were tested. My dissertation describes how to alter the speciation and concentration of molecules at the electrode-electrolyte interface, and how those alterations affect reactivity and selectivity. This dissertation presents two distinct approaches to control the mass transport of neutral molecules such as carbon dioxide (CO2) at the electrode-electrolyte interface by using an external magnetic field. The first finding demonstrates that magnetic fields indirectly affect the mass transport of target analytes such as CO2 molecules by selectively interacting with charged species, such as hydroxide ions (OH-). Metabolomics and lipidomics are the comprehensive studies of metabolites and lipids in cells, biological fluids, tissues, or organisms. These distinct chemical fingerprints are left behind by cellular processes. Epitranscriptomics is the field that focuses on examining chemical modifications on RNA molecules at the transcriptome level. Liquid chromatography-mass spectrometry (LC-MS) has been widely used in those omics studies. This dissertation investigates the electrochemical deposition (ECD) and electrophoretic deposition (EPD) processes involving citrate (cit)-stabilized Au nanoparticles (NPs) and calcium alginate (Ca-Alg-Au NP) hydrogel films. The first part of the dissertation investigates the influence of NP size, surface area (SA) coverage, and ligand stabilizers on the electrochemical growth of Au on Au NPs, aiming to enhance our understanding of the growth process during the nucleation and growth of metal NPs. The growth kinetics depend on Au NP size (4 nm > 15 nm > 50 nm) for a constant SA coverage, as determined by the overpotentials for ECD. The growth rate increases …

Ruddlesden Popper (RP) oxides are perovskite-derived functional materials with the general formula An-1Aʹ2BnO3n+1, where A/Aʹ is often a lanthanide or alkaline earth metal, and B is usually a transition metal. These materials contain perovskite-like connectivity, where BO6 units share apexes to form layers that are stacked above each other. The number of layers in each stack is represented by n in the above formula. The spaces between stacks are often occupied by lanthanide or alkaline-earth metals (Aʹ-site metals), which also reside in intra-stack spaces (A-site) between the octahedra. The peroxidation of unsaturated fatty acids is a widely recognized metabolic process that creates a complex mixture of volatile organic compounds including aldehydes. Elevated levels of reactive oxygen species in cancer cells promote random lipid peroxidation, which leads to an increase in a variety of aldehydes. Many of these volatile aldehydes are exhaled and are of interest as potential markers of disease. Chapter 1 presents a review of reported aldehydes in the exhaled breath of lung cancer patients. alpha,beta-Unsaturated aldehydes, detected primarily when derivatized during exhaled breath preconcentration, are underreported in the reviewed articles. The major aim of my dissertation was to investigate the etiology of dry eye disease which affects about 7 million people in the United States, causing symptoms that can lead to visual disturbance. Correlation between dry eye and an abnormal lipid layer of the tear film has been found. Tear film lipids originate mostly from the meibomian glands. Cholesteryl ester (CE) and Wax ester (WE) lipids make up most of the human meibum lipidome and the CE/WE ratio has been shown to decrease in patients with meibomian gland dysfunction. Metabolomics and lipidomics studies are important to understand living organisms at the molecular level. In this dissertation, I conducted various untargeted and targeted metabolomics and lipidomics studies using mass spectrometry based analytical platforms. Firstly, I investigated the androgen depletion effects on fecal metabolites in a mouse model of systemic lupus erythematosus and metabolite biomarkers for colorectal cancer using two-dimensional gas and liquid chromatography-mass spectrometry. Aminooxy (-ONH2) groups are most commonly known for their chemoselective reaction with carbonyl compounds (aldehydes/ketones) under mild reaction conditions. Aminooxy-based click chemistry is a versatile means of ligation as evidenced by broad application in material science, biology, biochemistry, analytical chemistry, and nanoscience. The first involves studies aimed at developing anodic stripping voltammetry (ASV) as an analytical tool to characterize metal nanoparticles (NPs), with a focus on the size and aggregate structure of single metal NPs and the composition and atomic arrangement of bimetallic NPs. This dissertation describes how anodic stripping voltammetry (ASV) was used to study size-controlled electrophoretic deposition (EPD) and the aggregation-dependent oxidation properties of citrate-stabilized Au nanoparticles (NPs). EPD of citrate-coated Au NPs occurs in the presence of hydroquinone (HQ) onto indium-tin-oxide-coated glass electrodes (glass/ITO) at potentials positive of the HQ oxidation potential. HQ oxidation produces protons at the electrode surface, which serve to neutralize the citrate molecules that electrostatically stabilize the Au NPs. Neutralization leads to the loss of stabilization and deposition onto the electrode. EPD in the presence of ferrocyanide, a non-proton-producing molecule, at oxidation potentials resulted in no deposition …

The vitreous humor (VH) is located in between the lens and the retina. It is composed of 98% water, hyaluronan (HA), collagen, proteins, phospholipids (PLs), and other metabolites. With aging, the VH undergoes liquefaction, a process that causes the gel-like structure of the VH to turn into a liquid and can lead to serious ocular diseases, including retinal detachment, vitreal detachment, and macular hole formation. The liquefaction process is expedited in diabetic VHs. This study mainly focuses on the fabrication of single one dimensional (1D) NP chains across the 5-micron gap of Au two electrode devices (TED). This has been achieved by electrochemical deposition of Ag onto the two electrodes, soaking the TED in 0.1 M Cetyltrimethyl ammonium bromide for 2-3 hours, and then close monitoring of the current as a function of time while a voltage across the two Ag coated Au electrodes in air. This dissertation focuses on the electrochemical formation of one-dimensional (1D) Ag nanostructures (in particular nanowires and nanochains) directly on electrode surfaces or gap of two electrodes. Ag nanowires (NWs) and nanorods (NRs) grow directly on glass/indium tin oxide (ITO) surfaces from electrode-attached Au nanoparticle seeds in the presence of cetyltrimethylammonium bromide (CTAB) at appropriate potentials with a 5-15 % yield and the average length ranging from 100 nm to 3 µm for growth times of 30 to 120 min. This dissertation describes the factors controlling metal nanoparticle (NP) oxidation, using Au oxidation in Br- and Ag oxidation in acid as model systems. We found that the oxidation potential depends on size, surface charge, and electrode material. Size has a dramatic effect, shifting about 200 mV from > 200 nm diameter down to 4 nm diameter. The shift is more dramatic below 4 nm. 2.3 nm average diameter Au NPs oxidized in the 300-500 mV range and 1.5 nm average diameter Au NPs displayed several sharp peaks from 200 mV to as low as -200 mV.
