The integration of photovoltaic (PV) solar energy in zero-energy buildings requires durable and efficient solar windows composed of lightweight and semitransparent thin film solar cells. Inorganic materials with a high optical absorption coefficient, such as Sb2S3 (>105 cm−1 at 450 nm), offer semitransparency, appreciable efficiency, and long-term durability at low cost. Oxide-free throughout the Sb2S3 layer thickness, as confirmed by combined studies of energy dispersive X-ray spectroscopy and synchrotron soft X-ray emission spectroscopy, semitransparent Sb2S3 thin films can be rapidly grown in air by the area-scalable ultrasonic spray pyrolysis method. A new series of hexadentate flexible bis(amidine) ligands (L1–6H2) and the multinuclear coinage metal complexes 1–5 of L4H2 were synthesized and characterized. The synthesis of L1–6H2 followed a 3-step protocol: first, the bis(amide) precursor is formed by reacting ethylene diamine with pivaloyl chloride; second, the bis(amide) is chlorinated with PCl5 to produce the corresponding bis(imidoyl chloride); finally, the bis(imidoyl chloride) is reacted with the primary arylamine (L1–6H2 yields: 62, 54, 56, 61, 63, and 58%). Water soluble CuInS2/ZnS quantum dots (QDs) represent one of the most promising single component photocatalysts for the hydrogen evolution reaction (HER). In this work, we report the effect of cation composition in CuInS2/ZnS QDs on the carrier relaxation and charge separation dynamics as well as their photocatalytic performance for the HER. With decreasing Cu to In ratio (increasing Cu deficiency), we observed slightly faster electron trapping and carrier recombination but significantly improved photocatalytic activity for the HER. Post-menopausal osteoporosis as a consequence of estrogen depletion is a growing concern for women in the United States. As more women take on executive positions and experience sleep deprivation, there is the potential for up regulation of pro-inflammatory cytokines, such as tumor necrosis factor alpha. It follows that the homeostatic imbalance of osteoclastic and osteoblastic activity leads to a greater risk of disease. Bisphosphonates generally, and Zolendronate specifically works by decreasing the number of osteoclasts. Electronic cigarettes (e-cigarettes) are an increasingly popular tobacco product used by ~3% of adults and 21% of 12th grade students (as reported for the prior 30-days in surveys) in the United States in 2018. As of 2019, this number went up to ~25% for 12th grade students. Due to how quickly e-cigarettes gained popularity and the rapid design changes that have ensued, there has not been enough time to accurately assess the effects of e-cigarettes, particularly for long-term use. Identity has been theorized to aid in student persistence toward science, engineering, technology, and math (STEM) degrees. However, before we can explore the impacts of learning environments on identity, and subsequently persistence, robust measures that can aid in understanding how identity is fostered need to be available. This research study aims to create measures of identity by expanding an existing physics identity framework and contextualizing it to science and chemistry identities. Development of the measures was carried out through two distinct phases that provided evidence for reliability and multiple aspects of validity.

Parallel spectroscopic and computational studies of iron(III) cysteine dioxygenase (CDO) and synthetic models are presented. The synthetic complexes utilize the ligand tris(4,5-diphenyl-1-methylimidazol-2-yl)phosphine (Ph2TIP), which mimics the facial three-histidine triad of CDO and other thiol dioxygenases. In addition to the previously reported [FeII(CysOEt)(Ph2TIP)]BPh4 (1; CysOEt is the ethyl ester of anionic l-cysteine), the formation and crystallographic characterization of [FeII(2-MTS)(Ph2TIP)]BPh4 (2) is reported, where the methyl 2-thiosalicylate anion (2-MTS) resembles the substrate of 3-mercaptopropionate dioxygenase (MDO). Barth syndrome (BTHS) is an X-linked recessive multisystem disorder caused by mutations in the TAZ gene (TAZ, G 4.5, OMIM 300394) that encodes for the acyltransferase tafazzin. This protein is highly expressed in the heart and plays a significant role in cardiolipin biosynthesis. Heart disease is the major clinical manifestation of BTHS with a high incidence in early life. The formation of carbonaceous by-products (e.g. soot) during the operation of an internal combustion engine is unavoidable and the aggregation of this soot leads to deleterious effects including abrasive wear of the engine, increased oil viscosities, and sludge deposition. Dispersants, which are composed of a hydrophobic tail and a polar headgroup, are used as oil additives to aid in the suspension and stabilization of the soot particles. Polyisobutylene succinimide (PIBSI) is the most well-studied class of dispersants and is characterized by a linear architecture and polyamine headgroup that interacts with soot by acid-base and dipole-dipole interactions. Essential oils (EOs) are plant-derived products that have been long exploited for their antimicrobial activities in medicine, agriculture, and food preservation. EOs represent a promising alternative to conventional antibiotics due to the broad-range antimicrobial activity, low toxicity to human commensal bacteria, and the capacity to kill microorganisms without promoting resistance. The mannose-6-phosphate/insulin-like growth factor II (M6P/IGF2) receptor is a transmembrane protein known to sequester growth factors from the extracellular matrix. This behavior suggests a mechanism of tumor suppression. Structurally, the receptor’s extracellular region is segmented into 15 homologous repeats, which are divided further into 5 triplet domains, labelled 1-3, 4-6, 7-9, 10-12, and 13-15. What is notable about the triplets is their propensity to form dimers with triplets on a second M6P/IGF2 receptor. Transition-metal catalyzed direct functionalization of inert C-H and C=C bonds through nitrene- and oxo-transfer reactions are powerful and attractive synthetic strategies for constructing C-N and C-O bonds (e.g. found in amines, aziridines, alcohols, and carbonyl compounds) directly from hydrocarbon feedstocks, and have significant synthetic potential in chemical processes leading to natural products, pharmaceuticals and biologically active molecules. Chapters 2 and 3 of this work mainly focus on nitrene- and oxo-insertion chemistry catalyzed by transition metal complexes. Coupling supercritical fluid extraction (SFE) on-line with supercritical fluid chromatography (SFC) – tandem mass spectrometry (MS/MS) provides a platform for extraction, separation, and detection in a single analysis. Utilization of this technique simplifies sample preparation while limiting sample loss and/or contamination and significantly decreases total analysis time. Mass spectrometry (MS)-based chemical cross-linking technique is a potential tool for understanding the protein structure and protein-protein interactions. However, low abundances of cross-linked products and the data complexity are the major challenges in this field. New strategies are required to pinpoint cross-linked peptides with high confidence. Our lab designed and synthesized a novel cross-linker with two differential MS-cleavable bonds, namely DUCCT. Highly uniform and vertically aligned p-type CuInSe2 (CISe) nanotube arrays were fabricated through a unique protocol, incorporating confined electrodeposition on lithographically patterned nanoelectrodes. This protocol can be readily adapted to fabricate nanotube arrays of other photoabsorber and functional materials with precisely controllable design parameters. Ternary CISe nanotube arrays were electrodeposited congruently from a single electrolytic bath and the resulting nanotube arrays were studied through powder X-ray diffraction as well as elemental analysis which revealed compositional purity. KTM is a 16 amino acid peptide with the sequence WCCSYPGCYWSSSKWC. Here, we present the nuclear magnetic resonance (NMR) structure and bioactivity of this rationally designed α-conotoxin (α-CTx) that demonstrates potent inhibition of rat α3β2-nicotinic acetylcholine receptors (rα3β2-nAChRs). Two bioassays were used to test the efficacy of KTM. First, a qualitative PC12 cell-based assay confirmed that KTM acts as a nAChR antagonist. Second, bioactivity evaluation by two-electrode voltage clamp electrophysiology was used to measure the inhibition of rα3β2-nAChRs by KTM (IC50 = 0.19 ± 0.02 nM), and inhibition of the same nAChR isoform by α-CTx MII (IC50 = …

Many recent theoretical and experimental techniques have been developed to probe the structurefunction relationships of complex biomolecules. The roles of RNAs are dependent upon various intricate structural motifs and interactions, including hairpins, pseudoknots, long range territory contacts, bulges and internal loops, that are not easily captured by these methods. A pheochromocytoma of the rat adrenal medulla derived (a.k.a. PC12) cell-based assay for dopamine measurement by luminescence detection was customized for the qualitative evaluation of agonists and antagonists of nicotinic acetylcholine receptors (nAChRs). The assay mechanism begins with ligand binding to transmembrane nAChRs, altering ion flow into the cell and inducing dopamine release from the cell. Following release, dopamine is oxidized by monoamine oxidase generating hydrogen peroxide that catalyzes a chemiluminescence reaction involving luminol and horseradish peroxidase, thus producing a detectable response. We present a computational study of amorphous boron carbide (a-BxC) models using Molecular Dynamics (MD) studied with Stillinger-Weber (SW) and ReaxFF potential. The atomic structure factor (S(Q)), radial distribution function (RDF) and bond lengths comparison with other experimental and ab initio models shows that a random arrangement of icosahedra (B12, B11C) interconnected by chains (CCC, CBC) are present in a-BxC. Veratrum californicum, commonly referred to as corn lily or Californian false hellebore, grows in high mountain meadows and produces bioactive steroidal alkaloids, including cyclopamine. Cyclopamine is a potent inhibitor of the Hedgehog (Hh) signaling pathway. Our lab has optimized methods to extract cyclopamine and related steroidal alkaloids from V. californicum, and implemented in-house Hh signaling bioactivity evaluation of these steroidal alkaloids using a Shh-Light II cell assay. A survey of extraction methods and solvents resulted in the identification of conditions most favorable for alkaloid extraction yield and antagonist activity in the Shh-Light II cell assay. The dissertation describes asymmetric synthesis towards C29-C34 moiety of fragment A of the Antascomicin B and Thermal azole based Claisen rearrangements. In chapter 1, we describes asymmetric synthesis towards C29-C34 moiety of fragment A of the Antascomicin B. The non-immunosuppressant Rapamycin, Ascomycin, and Tacrolimus (FK506), strongly binds with FKBP12, the ligand FKBP12 complexes responsible for immunosuppressive activity. Antascomicin B structurally related to Rapamycin, Ascomycin, and Tacrolimus (FK506), binds strongly to FKBP12, yet does not shown immunosuppressive activity. Siderophores are molecules capable of binding ferric ions with high affinity (Ka = 1052). This thesis outlines our attempts to synthesize an artificial siderophore, which incorporates a signaling group to help monitor the uptake of ferric ions. The artificial siderophore is a tripodal molecular receptor synthesized in a five-step procedure with a proposed final sixth step. Rhodamine 6G azide (95.0% yield) was formed from rhodamine 6G and the synthesis of 2-azidoethanamine was modified from the literature. The tripodal arm portion of the sensor was successfully synthesized by protecting the catechol of 2,3-dihydroxybenzaldehyde to form 3-formyl-1,2-phenylene dimethanesulfonate (98.8% yield). The first involves studies aimed at developing anodic stripping voltammetry (ASV) as an analytical tool to characterize metal nanoparticles (NPs), with a focus on the size and aggregate structure of single metal NPs and the composition and atomic arrangement of bimetallic NPs. The second main theme involves studies that use ASV and electrochemical surface area-to-volume (SA/V) measurements to study the unique reactivity and transformations of single metal and bimetallic NPs. This project investigates the synthesis and physical properties of polyethylene glycol monomethyl ether (MePEGn where n=3 or 7) based polymer electrolytes. The two polymer electrolytes explored in the experiments were designed from two different MePEG chains that varied in their overall length and degree of polymerization. The first MePEG backbone contained three polymerized ethoxy groups and the second contained seven. Both MePEGn polymers were modified by substituting an imidazolium group in place of the alcohol functional group at the end of the PEG chain. This dissertation describes an electrochemical approach for measuring the surface area-to-volume ratio (SA/V) of electrode-attached metal nanoparticles (NPs), which was used to analyze their size, aggregation state, and porosity. This dissertation further describes the effect of the metal NP electrode assembly method on the SA/V, which is related to metal NP catalytic activity and stability. Cyclic voltammetry (CV) in acid electrolyte followed by anodic stripping voltammetry (ASV) in KBr electrolyte allows the electrochemical measurement of the SA/V of electrode-attached Au nanospheres (NSs). In CV, the forward scan produces a thin surface Au2O3 layer on the Au NSs. …
