Naphthenic acids (NAs) are water-soluble components of petroleum. The characterization and quantification of NAs by analytical methods have proved quite challenging, whilst the toxic effects of these water-soluble compounds on a variety of organisms adversely affecting reproduction and steroid production is becoming apparent. In this study, we report a fluorescence-based competitive binding method for rapid sensing of the presence of NAs using cellulosic peptide array strips as sensors. The first chapter of this thesis presents an introduction to sol-gel methodology whose usefulness as a synthetic route will be demonstrated with two applications in chromatography. The first application involves the fabrication of a capillary micro-extraction (CME) device by coating a phenyl functionalized extracting phase on the inner surface of a fused silica capillary for analyte pre-concentration. The device was coupled on-line to a RP-HPLC system and practicality was demonstrated using allergens as target analytes. Calpain mediated cleavage of CDK5 natural precursor p35 causes a stable complex formation of CDK5/p25, which leads to hyperphosphorylation of tau. Thus inhibition of this complex is a viable target for numerous acute and chronic neurodegenerative diseases involving tau protein, including Alzheimer’s disease. Since CDK5 has the highest sequence homology with its mitotic counterpart CDK2, our primary goal was to design selective CDK5/p25 inhibitors targeting neurodegeneration. The title compound, C19H17N3O3S (I), was prepared by a [3 + 2]cyclo­addition azide condensation reaction using sodium azide and l-proline as a Lewis base catalyst. N-Methyl­ation of compound (I) using CH3I gave compound (II), C20H19N3O3S. The benzo­thio­phene ring systems in (I) and (II) are almost planar, with r.m.s deviations from the mean plane = 0.0205 (14) in (I) and 0.016 (2) Å in (II). A novel optically transparent glass fiber reinforced polymer matrix composite has been developed by infusing a clear epoxy resin system of matching refractive index into a conventional E-glass fabric preform. Transparent composites are manufactured using a low cost, environmentally friendly vacuum infusion process. Physical and mechanical tests have been conducted. Transparent composites manufactured using the modified vacuum infusion process had a fiber volume fraction of 40%. Tensile strength and tensile modulus of these composites were 374.9 MPa and 31.74 GPa respectively. To understand the chemistry of the interfacial region of reverse micelles (RM), we studied RM system made with the cationic surfactant cetyltrimethylammonium bromide (CTAB), alkanol cosurfactants dissolved in cyclohexane with water core. Spectroscopic methods, specifically UV-Vis absorption of Coumarin 343 (C343) as a probe molecule, were used to determine basic properties of RM systems. However, the probe location was difficult to determine because the spectrum (absorbance), when dissolved in RM solution, didn’t match the spectra in any of the pure components. Reverse micelles (RM) are nanopools of water surrounded by surfactant molecules in a non-polar solvent. Our studies investigated properties of RM probe molecules in the interfacial boundary between water and non-polar solvents. Reichardt’s dye, a probe molecule, was used to uncover properties of RM. UV/vis spectroscopy of Reichardt’s dye reveals information about polarity and the dye’s approximate residential location and also helps confirm molecular orbital energy diagram. We discovered an interesting phenomenon with Reichardt’s dye in RM. Surprisingly, the UV/vis spectrum of Reicahrdt’s dye in CTAB RM with octanol as a cosurfactant exhibits decreasing absorbance and a blue shift. Reichardt’s …

The actin cytoskeleton and its network of associated proteins constitute a physical barrier that viruses must circumvent to gain entry into cells for productive infection. The mechanisms by which the physical signals of infection are sensed by the host to activate an innate immune response are not well understood. The antiviral endoribonuclease RNase L is ubiquitously expressed in a latent form and activated upon binding 2-5A, a unique oligoadenylate produced during viral infections. We provide evidence that RNase L in its inactive form interacts with the actin-binding protein Filamin A to modulate the actin cytoskeleton and inhibit virus entry. Cells …

Galactic open and globular clusters (OCs, GCs) appear to inhabit separate regions of the age-mass plane. However, the transition between them is not easily defined because there is some overlap between high-mass, old OCs and low-mass, young GCs. We are exploring the possibility of a clear-cut separation between OCs and GCs using an abundance feature that has been found so far only in GCs: (anti)correlations between light elements. Among the coupled abundance trends, the Na-O anticorrelation is the most widely studied. These anticorrelations are the signature of self-enrichment, i.e., of a formation mechanism that implies multiple generations of stars. Here …

The glmS riboswitch undergoes self-cleavage upon binding its metabolic product GlcN6P, thereby providing a negative feedback mechanism limiting translation of the glmS protein when GlcN6P is abundant. As a first step toward the development of novel antimicrobials, we have synthesized a series of GlcN6P analogues bearing phosphatase-inert surrogates in place of the natural phosphate ester functionality. The self-cleavage assay identified two such compounds that display significant riboswitch actuator activity; namely those bearing a 6-phosphonomethyl group or a 6-O-malonyl ether. These two analogues exhibit a 22-fold and a 27-fold higher catalytic efficiency, respectively, than does glucosamine. Docking experiments were …

We present a hybrid nano-lithographic approach to minimizes the effects of line edge roughness and shot noise in nano-hole patterning by reflowing photoresist polymers around the nanoparticles deposited using self-assembly and simple etch chemistries. The method extends the transistor contact holes patterning limits to below 20 nm.

Observations of carbon-rich circumstellar envelopes were made using the Phoenix spectrograph on the Gemini South telescope to determine the abundance of small carbon chain molecules. Vibration-rotation lines of the ν3 antisymmetric stretch of C3 near 2040 cm-1 (4.902 μm) have been used to determine the column density for four carbon-rich circumstellar envelopes: CRL 865, CRL 1922, CRL 2023 and IRC +10216. We additionally calculate the column density of C5 for IRC +10216, and provide an upper limit for five more objects. An upper limit estimate for the C7 column density is also provided for IRC+10216. The removal of arsenic(III) and arsenic(V) from an aqueous solution through adsorption on to Fe3O4, MnFe2O4, 50% Mn substituted Fe3O4, 75% Mn substituted Fe3O4, and Mn3O4 nanomaterials was investigated. Characterization of the nanomaterials using XRD showed only pure phases for Mn3O4, MnFe2O4, and Fe3O4. The 50% and 75% substituted nanomaterials were found to be mixtures of Mn3O4 and Fe3O4. From batch studies the optimum binding pH of arsenic(III) and arsenic(V) to the nanomaterials was determined to be pH 3. Lipophilic cationic technetium-99m-complexes are widely used for myocardial perfusion imaging (MPI). However, inherent uncertainties in the supply chain of molybdenum-99, the parent isotope required for manufacturing 99Mo/99mTc generators, intensifies the need for discovery of novel MPI agents incorporating alternative radionuclides. Recently, germanium/gallium (Ge/Ga) generators capable of producing high quality 68Ga, an isotope with excellent emission characteristics for clinical PET imaging, have emerged. Herein, we report a novel 68Ga-complex identified through mechanism-based cell screening that holds promise as a generator-produced radiopharmaceutical for PET MPI.

The nano-cubic Pt modified by tin (Sn) was synthesized and used to investigate the role of this adatom played in the ethanol oxidation. The onset potential of ethanol oxidation was significantly shifted negatively which can be forward about 300 mV when the coverage of Sn (θSn)was 0.9. The electrtochemical in situ FTIR result demonstrated that the amount of CO2 increased first, and then decreased with θSn increased, and reached the maximun when θSn was 0.38. The DNA-multi-walled carbon nanotubes (MWCNTs)/glassy carbon (GC) electrode was prepared by modifying the DNA functionalized MWCNTs composite on a GC electrode. A novel non-enzymatic H2O2 sensing electrode was fabricated by electrodepositing Pt nanoparticles on the DNA-MWCNTs/GC electrode. The modified electrodes were characterized by scanning electron microscope (SEM). The response properties of the sensing electrode to H2O2 were investigated by cyclic voltammetry and chronoamperometry. Nanostructured CoPt alloy thin film was deposited on glassy carbon substrate (denoted as CoPt/GC) under cyclic voltammetric conditions. SEM studies demonstrated that the CoPt/GC was composed of nonuniform nanoparticles with an average size of 139 nm. The composition of the CoPt/GC was analyzed by energy dispersive X-ray spectroscopy (EDS), which depicts a Co:Pt ratio of ca. 3:5. In situ FTIR spectroscopy employing CO adsorption as a probe reaction revealed that the CoPt/GC exhibits abnormal infrared effects (AIREs). Developing catalytic materials for oxygen reduction reactions (ORR) with high performance and low cost has been one of the major challenges for large-scale applications of fuel cells. In this review, we summarize the recent progress in the heteroatoms doped carbon materials as a catalyst for the catalysis of ORR, with an emphasis on the universal origin of their catalytic mechanisms. The future prospects of the metal-free catalyst were to find precisely controlled doping methods, to explore the role of heteroatoms in the catalysis, and to generate more active catalysts with suitable nanostructure.

Graphene oxide (GO) was prepared by modified Hummers method. Using the GO solution as "ink", Au nanoparticles (AuNPs) and GO were transferred to the surface of ITO substrate modified with (3-aminopropyl) triethoxysilane (APTES/ITO) in a sequence. The transferred AuNPs and GO could form a uniform and dense composite pattern which was characterized by FE-SEM and AFM. Moreover, using the APTES/ITO substrate surface potential as zero, the sequences of the surface potential were APTES>GO>Au.

Solid oxide fuel cell (SOFC) directly operating on hydrocarbon without external reforming has the potential of greatly speeding up the application of SOFCs for transportation. In this paper, a three-layer structure anode was fabricated by tape casting and screen printing method. The addition of Cu-LSCM-CeO2 to the supported anode surface presented better performance running on H2 and ethanol. The maximum power densities were 511 and 390 mW?cm-2, respectively, running on H2 and ethanol at 750 °C. No significant degradation was observed on the anode. Bimetallic PtRu nanotubes array electrodes (NTAEs) with varied Pt to Ru atomic ratios have been prepared by electrochemical codeposition in 3-aminopropyltri-methoxysilane modified porous anodic alumina (PAA) membranes. The structure and morphology of the catalysts were characterized using X-ray diffraction (XRD) and scanning electron microscopy (SEM), respectively. Electrochemical results showed that the NTAEs with varied atomic ratios could be achieved by controlling the precursor concentration for deposition. Biomimic membrane with nano-channels, which is made up of porin MspA and 1,2-Dimyristoyl-sn-glycero-3-phosphocholine (DMPC) is constructed on glassy carbon substrate, and glucose oxidase (GOD) is modified on it. The direct electrochemical reaction and electrocatalytic behavior to oxygen and glucose of GOD on the GOD/MspA-DMPC/GC electreode are expounded by the cyclic voltammetric method. The interactions between four Imidazole ionic liquids (EMIMBF4, BMIMBF4, BMIMHSO4, BMIMAc) and p-methoxy toluene were investigated by ultraviolet spectroscopy. The red shifts of absorption peak were obviously observed, and the fine structures of the spectroscopy were disappeared on the long wave district. p-MT producing stable excited state was benefit from the interaction with solvent molecules.
