This collection of studies focuses on a PCO system for the oxidation of a model compound, ethanol, using an adsorption-enhanced silica-Ti02 composite (STC) as the photocatalyst; studies are aimed at addressing the optimization of various parameters including light source, humidity, temperature, and possible poisoning events for use as part of a system for gaseous trace-contaminant control system in closed-environment habitats. Polycyclic aromatic hydrocarbons (PAH) are important environmental pollutants generally formed during incomplete combustion of organic matter containing carbon and hydrogen. Introduced into the human body by adsorption through the skin, ingestion or inhalation, the biotransformation processes of PAH lead to the formation of multiple metabolites. Due to the short elimination lifetime from the body, the quantitative determination of monohydroxy-PAH (OH-PAH) in urine samples provides accurate information on recent exposure to environmental PAH. Urine analysis of OH-PAH with established methodology relies on sample clean-up and pre-concentration followed by chromatographic separation and quantification. PCBs are recalcitrant compounds of no known natural origin that persist in the environment despite their ban by the United States Environmental Protection Agency in 1979 due to negative health effects. Transport of PCBs from elastic sealants into concrete, brick, and granite structures has resulted in the need for a technology capable of removing these PCBs from the materials. The discovery of any new technology is usually accompanied by a need for new or improved materials which make that technology useful in practical applications. In the case of two-photon absorption (2PA) this has truly been the case. Since its first demonstration in 1961, there has been an ever increasing quest to understand the relationships between two-photon absorption and the structure of two-photon absorbing materials. In forensic casework analysis it is necessary to obtain genetic profiles from increasingly smaller amounts of biological material left behind by perpetrators of crime. The ability to obtain profiles from trace biological evidence is demonstrated with so-called ‘touch DNA evidence’ which is perceived to be the result of DNA obtained from shed skin cells transferred from donor to an object or person during physical contact. However, the current method of recovery of trace DNA involves cotton swabs or adhesive tape to sample an area of interest. This dissertation contains the synthesis and characterization of squaraine based new functional materials. In the first part of this thesis work, a water soluble benzothiazolium squaraine dye was synthesized with pyridium pendents, and controlled aggregation properties were achieved. After formation of partially reversible J-aggregation on a polyelectrolyte (poly(acryl acid) sodium salt) template, the nonlinear, two-photon absorption cross section per repeat unit was found to be above 30-fold enhanced compared with nonaggregate and/or low aggregates. Using a similar strategy, sulfonate anions were introduced into the squaraine structure, and the resulting compounds exhibited good water solubilities. A ‘turn on’ fluorescence was discovered …

De novo protein design offers many interesting prospects both as a means to better understand natural protein dynamics and as a potential resource in biomedical and industrial applications. In this work I describe the modification of a simple, well-characterized heme-binding protein by altering side chain residue identities on the hydrophilic surface of the protein to produce variants with a range of net external charges. These charge modifications had a significant impact on nearly every measurable character of the protein. Carbon nano-onions (CNOs) discovered by Ugarte in 1992 are multi-layered fullerenes that are spherical analogs of multi-walled carbon nanotubes with diameters varying from 6 nm to 30 nm. Among the various methods of synthesis, CNOs prepared by graphitization of nanodiamonds (N-CNOs) and underwater electric arc of graphite rods (A-CNOs) are the subject of our research. N-CNOs are considered as more reactive than A-CNOs due to their smaller size, high curvature and surface defects.

Previous studies from our laboratory and others show a significant increase in levels of both nuclear and mitochondrial DNA and RNA oxidation in vulnerable brain regions in the progression of Alzheimer’s disease (AD). Although total DNA oxidation is increased in AD it remains unclear whether oxidative damage is widespread throughout the genome or is concentrated to specific genes. This dissertation serves to study a protein's conformation-function relationship since immobilized proteins often behave differently from their solution-state counterparts. Therefore, this study is important to the application of protein-based biodevices. Another aim of this dissertation is to explore a new approach to realize low voltage electrowetting without the help of oil bath. Utilizing this approach, a protein micro-separation was realized. Additionally, the interfacial properties of ionic liquid (IL) solid-like layer, which played a key role in electrowetting, was studied for further developments of IL-based applications.

Spreading plays an important role in coating, lubrication, printing and etc. During the spreading process, a liquid thin film forms prior to the expansion of a liquid drop. This thin film is called a precursor film. It not only changes the spreading mechanism, but impacts the wettability of a liquid. Carbon nano-onions (CNOs), concentrically multilayered fullerenes, are prepared by several different methods. We are studying the properties of two specific CNOs: A-CNOs and N-CNOs. A-CNOs are synthesized by underwater arc discharge, and N-CNOs are synthesized by high-temperature graphitization of commercial nanodiamond. In this study the synthesis of A-CNOs are optimized by designing an arc discharge aparatus to control the arc plasma. Moreover other synthesis parameters such as arc power, duty cycles, temperature, graphitic and metal impurities are controlled for optimum production of A-CNOs. Also, a very efficient purification method is developed to screen out A-CNOs from carboneseous and metal impurities. …

The focus of this dissertation is to utilize chemical functionalization as a probe to investigate the reactivity of N-doped multiwalled carbon nanotubes (N-MWCNTs). The surface of N-MWCNTs, being a set of potentially reactive graphene edges, provides a large number of reactive sites for chemical modification, so considerable changes in chemical and physical properties can be envisaged. We observed that both reduction (dissolving metal reduction/alkylation) and oxidation (H2SO4/HNO3 and H2SO4/KMnO4 mixtures) of N-MWCNTs lead to formation of interesting spiral channels and spiraled carbon nanoribbons. The pursuit of chemical specificity and efficiency of chemical reaction in molecular crystals has attracted considerable attention over the last decade. The successful control of molecular alignment prior to such transformations continues to provide opportunities for near quantitative yields of new materials. This thesis reports the predictable self-assembly of aryl sulfonamide materials constructed from cinnamic acid and phenylpentadienoic acid fragments capable of forming supramolecular dimers that undergo unique photocyclization reactions. Use of racemic and enantiopure single-component building-blocks provides access to enantiocontrolled [2+2] photodimerized products.

Designing new materials with predictable chemical and physical properties has long been a goal of synthetic chemists. The functional properties of molecular compounds are intimately tied to the underlying chemical framework. In addition to atom selection and chemical connectivity, the spatial organization of the molecules often contributes to the function of materials. Because crystals are constructed of periodic arrays of components (i.e. atoms or molecules) with structures easily assessed via X-ray diffraction, crystalline materials offer ideal candidates to explore the relationship of component selection and crystal packing to material property.

Electrochemical diazonium grafting offers versatile functionalization of chemically inert graphite under mild condition, which is particularly suitable for CNT composite modification. Tetrafluorinated carboxylphenyl diazonium grafting provides the most controllable functionalization chemistry allowing near monolayer levels of functionality on carbon nanotubes. The functional density was successfully quantified by anion selective dye-assay and X-ray photoelectron spectroscopy (XPS) of thiol-Au self-assembled monolayers (SAM) as a calibration reference. This technique enables monolayer functionality at the tips of carbon nanotube membranes for biomimetic pumps and valves as well as thin conductive layers for CNT-based high area electrochemical support electrodes.

Fast pyrolysis, the rapid thermal decomposition of organic material in the absence of oxygen, is a process that can be used to convert biomass into liquid fuels and chemicals. When performed at the micro-scale, pyrolysis is useful for characterizing biomass structure, as well as determining the pyrolysis products that can be generated from specific biomass feedstocks. Stratospheric chemistry and denitrification are simulated for the Arctic winter 2009/2010 with the Lagrangian Chemistry and Transport Model ATLAS. A number of sensitivity runs is used to explore the impact of uncertainties in chlorine activation and denitrification on the model results. In particular, the efficiency of chlorine activation on different types of liquid aerosol versus activation on nitric acid trihydrate clouds is examined. Additionally, the impact of changes in reaction rate coefficients, in the particle number density of polar stratospheric clouds, in supersaturation, temperature or the extent of denitrification is investigated. "Aerogels are 3D assemblies of nanoparticles with high open porosity and high surface area, and are pursued for their low density, low thermal conductivity, low dielectric constant and high acoustic attenuation. The foundation for those exceptional properties is their complex hierarchical solid framework (agglomerates of porous, fractal secondary nanoparticles). On the down side, however, aerogels are also fragile materials. The mechanical strength of silica aerogels has been improved by crosslinking the framework with organic polymers. "Aerogels are nanoporous, low-density bulk objects, consisting of three-dimensional assemblies of nanoparticle. Structured similarly, polymeric aerogels are emerging as a mechanically strong alternative to traditional silica aerogels, which are fragile. Amongst polymeric aerogels, those based on polybenzoxazine (PBO - a type of phenolic resin), are extremely robust and comprise an economic alternative to resorcinol-formaldehyde aerogels, also a class of phenolic resins, as the main source of carbon aerogels. The drawback of the PBO chemistry has been the long (days) processing time at high-temperatures (>130 ºC). New methods for studying enzymes and enzyme inhibition using capillary electrophoresis (CE) were developed and applied. Because CE is a separation technique, spectral interference is minimized by separation of substrates, products, inhibitors, and sample matrix components. Two types of CE enzyme assays were explored in this dissertation research. The first assay was based on optically gated vacancy capillary electrophoresis (OGVCE) with laser-induced fluorescence detection (LIF). This approach involves periodic photobleaching of fluorescent substrates and products. The initial goal of the study was to develop an assay for adenosine deaminase (ADA). In Skp1 of Dictyostelium discoideum (Dd), Pro143 is located at the N-terminus of an α-helix with four consecutive Glu residues immediately following Pro. Preceeding Pro143 is a segment of random coil. The proline residue undergoes post-translational modifications: hydroxylation and glycosylation. A cytoplasmic prolyl hydroxylase (P4H1) delivers a hydroxyl group to Pro143 and N-acetylglucosamine transferase 1 (Gnt1) transfers GlcNAc from UDP to Hyp143 of Skp1. The installation of the first GlcNAc residue in Skp1 in Dictyostelium is important for the organism to differentiate into a fruiting body to disperse spores.
