Hyperbranched poly(ethylenimine)s (HPEI) were modified with hydrophobic isobutyryl amide groups (HPEI-IBAm0.60) to cause an LCST. These modified polymers were then further substituted with hydrophobic alkyl chains (HPEI-IBAm0.60-R0.40), a mixture of alkyl chains and hydroxyethyl groups (HPEI-IBAm0.60-EtOH0.20-R0.20), and a mixture of alkyl chains and low molecular weight PEG (HPEI-IBAm0.60-PEG0.20-R0.20) to determine the effect of different functional groups on solubility behavior. At pH 7.4, all but three LCSTs were below body temperature (37 °C), while at pH 5, all but three LCSTs were above …

A series of hydroquinone-based poly(arylene ether) copolymers, with truly pendent sulfonated phenyl sulfonyl groups and either benzothiazole or benzoxazole groups was synthesized for use as proton exchange membranes (PEM). Two homopolymers series, benzothiazole and benzoxazole based, were prepared via meta activated nucleophilic aromatic substitution polycondensation reactions. Two copolymer series of N-heterocyclic and sulfonated phenyl sulfonyl groups were synthesized via a similar method. The ratio of 3,5-difluoro-3'-sulfonated diphenylsulfone to 2-[3,5-bis-(4-hydroxyphenoxy)]benzothiazole or 2-[3,5-bis-(4-hydroxyphenoxy)]benzoxaole was varied to afford ion-exchange capacities ranging from 0.60 to 1.39 meq and 0.62 to 1.42 meq, respectively. The ability to predict accurately the energy V0(ρ) of the quasi-free electron along the entire density (ρ) range of a supercritical fluid has applications in determining the ideal thermodynamic conditions for chemical reactions involving charged species. The previously established field ionization method of extracting V0(ρ) from the fluid density dependent shift Δ(ρ) in the ionization energy of a dopant molecule has led to the discovery of a novel effect on V0(ρ) occurring near the critical isotherm of the fluid. A recently development thermal infrared camera system was improved by designing new components to downsize physical aspect and improve the instrument's performance. A new thermal infrared light source was developed and characterized. It is designed from a custom-made Macor backpiece that holds an alumina screw. High resistance wire is coiled around the screw. The coiled wire is covered with an alumina coating. Most light sources have a spectral emissivity similar to that of a blackbody emitter. A colloidal solution of citrate-stabilized gold nanoparticles (AuNPs) with an average size of ca. 2.6 nm has been prepared, characterized and further implemented in electro-sensing applications. This colloidal solution of AuNPs has been prepared via the reduction of NaAuCl4 with sodium tetrahydroborate (NaBH4) using trisodium citrate as a stabilizer. The optical properties of this solution have been studied with UV–Vis spectroscopy. Next, these AuNPs have been immobilized onto a polycrystalline Au (poly-Au) electrode with the assistance of benzenedimethanethiol (BDMT), which served as a binder. A novel route to the exponential trapping-time distribution within a solidlike state in water clusters is described. We propose a simple homogeneous network (SHN) model to investigate dynamics on the potential energy networks of water clusters. In this model, it is shown that the trappingtime distribution in a solidlike state follows the exponential distribution, whereas the trapping-time distribution in local potential minima within the solidlike state is not exponential. To confirm the exponential trapping-time distribution in a solidlike state, we investigate water clusters, (H2O)6 and (H2O)12, by molecular dynamics simulations. Ceria nanorods were demonstrated to be an active, bifunctional catalyst for the cyanosilylation of aldehydes. The catalytic activity of ceria was shown to be positively correlated with a decrease in the coordination numbers of neighbouring oxygen atoms around cerium atoms in the catalyst. Chemisorption and density functional theory studies suggested that the coordinatively unsaturated cerium sites exposed by the surface oxygen vacancy defects functioned as Lewis acid sites and the neighbouring oxygen atoms behaved as Lewis base sites in the catalytic cycle.

We explored the potentials of microarray printing using quill-like microcantilevers onto solid supports that are typically used in microspot printing, including paper, polymeric nitrocellulose and nylon membranes. We compared these membranes with a novel porous poly(2-hydroxyethyl methacrylate-co-ethylene dimethacrylate) support (HEMA) with narrow pore size distribution in the 150 nm range, which demonstrated advantages in pattern definition, spot homogeneity, and consistent spot delivery of different dyes (phloxine B and bromophenol blue) with diameters of several micrometres. Riboswitches are structural elements in the 50 untranslated regions of many bacterial messenger RNAs that regulate gene expression in response to changing metabolite concentrations by inhibition of either transcription or translation initiation. The preQ1 (7-aminomethyl-7-deazaguanine) riboswitch family comprises some of the smallest metabolite sensing RNAs found in nature. Once ligand-bound, the transcriptional Bacillus subtilis and translational Thermoanaerobacter tengcongensis preQ1 riboswitch aptamers are structurally similar RNA pseudoknots; yet, prior structural studies have characterized their ligand-free conformations as largely unfolded and folded, respectively. This disclosure relates to compounds, reagents, and methods useful in the synthesis of aryl fluorides, for example, in the preparation of 18F labeled radiotracers. For example, this disclosure provides universal “locked” aryl substituents that result in StereoElectronic Control of Unidirectional Reductive Elimination (SECURE) from diaryliodonium salts. The reagents and methods provided herein may be used to access a broad range of compounds, including aromatic compounds, heteroaromatic compounds, amino acids, nucleotides, and synthetic compounds.

This report used high-performance affinity microcolumns to examine the changes in binding by sulfonylurea drugs to in vivo glycated HSA that had been isolated from individual patients with diabetes. An immunoextraction approach was developed to isolate HSA and glycated HSA from clinical samples, using only 20 μL of plasma or serum and 6–12 nmol of protein to prepare each affinity microcolumn. It was found that the affinity microcolumns could be used in either frontal analysis or zonal elution studies, which typically required only 4–8 min per run. We have studied the structure and stability of (H3O+)(H2O)8 clusters using a combination of molecular dynamics sampling and high-level ab initio calculations. 20 distinct oxygen frameworks are found within 2 kcal/mol of the electronic or standard Gibbs free energy minimum. The impact of quantum zero-point vibrational corrections on the relative stability of these isomers is quite significant. The box-like isomers are favored in terms of electronic energy, but with the inclusion of zero-point vibrational corrections and entropic effects tree-like isomers are favored at higher temperatures. Climatic effects of newly-formed atmospheric secondary aerosol particles are to a large extent determined by their condensational growth rates. However, all the vapours condensing on atmospheric nanoparticles and growing them to climatically relevant sizes are not identified yet and the effects of particle phase processes on particle growth rates are poorly known. Interest in determining the mechanism by which pro biotic bacteria in the gastrointestinal tract produce short chain fatty acids (SCFA) has increased over the past few years. This study uses gas chromatography and mass spectrometry to characterize lactate production in aerobic test tube cultures, aerobic bioreactor cultures, and anaerobic bioreactor cultures. To collect anaerobic bioreactor samples, a novel in vitro anaerobic model was developed. The pH and colony forming units of each bacterial sample was also measured in order to understand the correlation between lactate production and bacterial growth. De novo protein design offers many interesting prospects both as a means to better understand natural protein dynamics and as a potential resource in biomedical and industrial applications. In this work I describe the modification of a simple, well-characterized heme-binding protein by altering side chain residue identities on the hydrophilic surface of the protein to produce variants with a range of net external charges. These charge modifications had a significant impact on nearly every measurable character of the protein. Elastin-like polypeptides (ELPs) are polypentapeptides that undergo hydrophobic collapse and aggregation above a specific transition temperature, Tt. ELP diblocks sharing a common “core” block (I60) but varying “outer” blocks (A80, P40) were designed, where Tt,I < Tt,A < Tt,P. The formation of ~55 nm diameter mixed micelles from these ELP diblocks was verified using dynamic light scattering (DLS), multiangle light scattering (MALS) and fluorescence resonance energy transfer (FRET). Alzheimer disease (AD) is an age-related neurodegenerative disease characterized by the presence of three pathological hallmarks: synapse loss, extracellular senile plaques (SP) and intracellular neurofibrillary tangles (NFTs). The major component of SP is amyloid β-peptide (Aβ), which has been shown to induce oxidative stress. The AD brain shows increased levels of lipid peroxidation products, including 4-hydroxy-2-nonenal (HNE). HNE can react covalently with Cys, His, or Lys residues on proteins, altering structure and function of the latter. Conventional anodising electrolytes based on sulphuric acid, oxalic acid and phosphoric acid have been used to form nanoporous layers on AA3003-H14 and sealed with silane based sol-gel sealers. It is shown that the sol-gel chemistries have varying levels of pore penetration depending on the synthesis conditions. The extent of sol-gel penetration and pore sealing is analysed by electron microscopy and energy dispersive x-ray spectroscopy. To describe the sealing phenomena observed a sol-gel penetration and sealing rating has been proposed to explain the interactions of the sol-gels with the pores of the anodised layers. A novel ambient pressure microwave-assisted technique is developed in which silver and indium modified ZnS is synthesised. The as prepared ZnS is characterised by X-ray diffraction, UV-Vis spectroscopy, X-ray photoelectron spectroscopy and luminescence spectroscopy. This procedure produced crystalline materials with particle sizes below 10 nm. The synthesis technique leads to defects in the crystal which induce mid energy levels in the band gap and lead to indoor light photocatalytic activity. Targeting the anticancer agents selectively to cancer cells is desirable to improve the efficacy and to reduce the side effects of anticancer therapy. Previously reported passive tumor targeting by PEGylated liposomes (stealth liposomes) have resulted in their higher tumor accumulation. However their interaction with cancer cells has been minimal due to the steric hindrance of the PEG coating. This dissertation reports two approaches to enhance the interaction of stealth liposomes with cancer cells. Microbial fuel cells (MFC) utilize bacteria to generate an electrical current that can be used in the decomposition of sludge and human urine. In a MFC there is an anode (for oxidation of organic compounds), cathode (reduction of oxygen or carbon dioxide), and a proton exchange membrane (PEM, allows protons to migrate); reduction-oxidation reactions between the anode and cathode produce a measurable current. Bacteria that are found in sludge can be used to produce electrons in a voltaic cell, but optimizing conditions for harnessing the energy is crucial to making a MFC efficient. This dissertation describes a new vinyl-acrylic copolymer that displays great potential for applications in lithium ion batteries by enabling processes that are novel, faster, safer, and less costly than existing manufacturing methods. Overall, the works presented are based on tailored chemical synthesis directly applied to lithium ion battery manufacturing. Current manufacturing methods still have many flaws such as toxic processes and other time consuming if not costly steps. Understanding the chemistry of materials and processes related to battery manufacturing allows the design of techniques and methods that can ultimately improve the performance of existing batteries while reducing the cost. The H355A, H355K, H80A, and H80K mutant enzymes of the argE-encoded N-acetyl-L-ornithine deacetylase (ArgE) from Escherichia coli were prepared, however, only the H355A enzyme was found to be soluble. Kinetic analysis of the Co(II)-loaded H355A exhibited activity levels that were 380-fold less than Co(II)-loaded WT ArgE. Electronic absorption spectra of Co(II)-loaded H355A-ArgE indicate that the bound Co(II) ion resides in a distorted, five-coordinate environment and Isothermal Titration Calorimetry (ITC) data for Zn(II) binding to the H355A enzyme provided a dissociation constant (Kd) of 39 μM. A surface adlayer is generated on organic single crystals (tetracene and rubrene) using the site specific Diels–Alder reaction and a series of vapor phase dienophiles. X-ray photoelectron spectroscopy (XPS) confirms adsorption on the surfaces of tetracene and rubrene and mass spectrometry demonstrates the reaction’s applicability to a range of dienophiles.

Stable, aqueous, red-to-near infrared emission is critical for the use of silicon nanoparticles (Si NPs) in biological fluorescence assays, but such Si NPs have been difficult to attain. We report a synthesis and surface modification strategy that protects Si NPs and preserves red photoluminescence (PL) in water for more than 6 mo. The Si NPs were synthesized via high temperature reaction, liberated from an oxide matrix, and functionalized via hydrosilylation to yield hydrophobic particles. The hydrophobic Si NPs were phase transferred to water using the surfactant cetyltrimethylammonium bromide (CTAB) with retention of red PL. CTAB apparently serves a double role …

Ground-based aerosol measurements made in June 2010 within Sacramento urban area (site T0) and at a 40-km downwind location (site T1) in the forested Sierra Nevada foothills area are used to investigate the evolution of multispectral optical properties as the urban aerosols aged and interacted with biogenic emissions. Along with black carbon and non-refractory aerosol mass and composition observations, spectral absorption (ᵝabs), scattering (ᵝsca), and extinction (ᵝext) coefficients for wavelengths ranging from 355 to 1064nm were measured at both sites using photoacoustic (PA) instruments with integrating nephelometers and using cavity ring-down (CRD) instruments. Introduction of S-ethyl groups in all four ortho positions of azobenzene prevents reduction of the azo group by intracellular glutathione, while enhancing the absorptivity to ∼10000 M−1 cm−1 in the blue and green regions of the visible spectrum. cis-to-trans isomerization occurs thermally on the minutes timescale. Further, this substitution pattern permits switching with red light, a color that is more penetrating through biological tissues than other parts of the visible spectrum.

This collection of studies focuses on a PCO system for the oxidation of a model compound, ethanol, using an adsorption-enhanced silica-Ti02 composite (STC) as the photocatalyst; studies are aimed at addressing the optimization of various parameters including light source, humidity, temperature, and possible poisoning events for use as part of a system for gaseous trace-contaminant control system in closed-environment habitats. The first goal focused on distinguishing the effect of photon flux (i.e., photons per unit time reaching a surface) from that ofphoton energy (i.e., wavelength) of a photon source on the PCO of ethanol. For centuries chemists have sought to improve humankind’s quality of life and address many of society’s most pressing needs through the development of chemical processes and synthesis of new compounds, often with phenomenal results. Unfortunately, there also are many examples where these chemicals have had unintended, detrimental consequences that are not apparent until years or decades after their initial use. There are numerous halogenated molecules in this category that are globally dispersed, resistant to natural degradation processes, bioaccumulative, and toxic to living organisms.
