An extensive set of carbonyl sulfide (OCS) observations were made as part of the NASA Intercontinental Chemical Transport Experiment-North America (INTEX-NA) study, flown from 1 July to 14 August 2004 mostly over the eastern United States and Canada. These data show that summertime OCS mixing ratios at low altitude were dominated by surface drawdown and were highly correlated with CO2. Although local plumes were observed on some low-altitude flight legs, anthropogenic OCS sources were small compared to this sink. Start codon selection is a key step in translation initiation as it sets the reading frame for decoding. Two eukaryotic initiation factors, eIF1 and eIF1A, are key actors in this process. Recent work has elucidated many details of the mechanisms these factors use to control start site selection. eIF1 prevents the irreversible GTP hydrolysis that commits the ribosome to initiation at a particular codon. eIF1A both promotes and inhibits commitment through the competing influences of its two unstructured termini. Both factors perform their tasks through a variety of interactions with other components of the initiation machinery, in many cases mediated …

β-lactamases hydrolyze penicillin, cephalosporin, and carbapenemβ-lactam antibiotics. The class D β-lactamase OXA-1 has a catalytic serine (position 67) thought to be deprotonated and thereby activated by an unusual Nζ-carboxylated lysine (position 70). We have made several mutations of OXA-1 at both K70 and S67 to help elucidate the role of these two critical residues in the catalytic mechanism. We have used the fluorescent substrate BOCILLIN FL to demonstrate that the K70 mutants can acylate but are severely impaired for deacylation of various substrates. Pigments developed by the Mayan civilization around 8th century, represent some of the most versatile pigments known to date. Several derivatives of these pigments are popular subjects of current research interest. This is due to the characteristic stability which is provided by a bonding mechanism between the dye and the clay. One such pigment "Maya Blue", a mixture of Indigo and Palygorskite, provides a dramatic background for murals and ceramics throughout Mesoamerica. Maya Blue has been the focus of numerous studies and is believed to be a mixture of palygorskite clay and indigo dye.1,2 Several derivatives of this pigment have been developed with intriguing properties. For instance, the dye thioindigo reacts with the palygorskite clay to exhibit a broad range of colors from red to blue under UV-Vis excitation. Magnetic nanoparticles are of great interest for a wide range of applications. This work has focused on three primary forms of iron based nanoparticles and combinations thereof: α-iron, iron oxide, and iron carbide or cementite. The synthesis of several core-shell particles including cementite-iron oxide, α-iron-cementite, and α-iron-iron oxide was accomplished through reverse micelle routes and high temperature decomposition of iron pentacarbonyl in various media. Structural analysis to confirm the structures was performed using extended x-ray absorption fine structure (EXAFS) techniques. A series of 1,3-di-substituted-imidazolium lithium phthalocyanines, in which the substituents on the imidazolium nitrogens were combinations of methyl, ethyl, pentyl, hexyl, isopropyl, adamantyl or 2,4,6-trimethylphenyl groups, was synthesized. The cation exchange of a single lithium ion of dilithium phthalocyanine for a 1,3-disubstituted-imidazolium ion was performed by mixing their salts in common organic solvents under ambient conditions. This afforded a number of imidazolium lithium phthalocyanines in moderate yields. They exhibited poor solubility in most solvents. Their composition and purity were initially verified by 1H and 13C-NMR and elemental analysis. The 1H-NMR spectra also indicated that the imidazolium and lithium phthalocyanine ions …

The synthesis of new organic materials that have the potential to be utilized in a thermoelectric application are in demand because they are able to convert thermal energy directly into electrical energy. The "end-on" sulfoxide and sulfone derivatives of Benzo[1,2-c:3,4-c':5,6-c'']trithiophene 1 were synthesized for this reason. The "end-on" mode sulfoxide derivative 13 of Benzo[1,2-c:3,4-c':5,6-c'']trithiophene 1 was prepared by utilizing 3-Carboxypyridinium Chlorochromate (CPCC) 16 and AlCl3, as an oxidizing system. The 1H NMR (DMSO-d6) spectrum displayed a singlet peak at 5.03 ppm, and the IR spectrum displayed an absorption at 1059.33cm-1, which confirmed its …

Evidence shows that a gradient like copolymer can be synthesized using nucleophilic aromatic substitution, NAS, chemistry. Using the 4-chloro-3-fluoro-4'hydroxydiphenylsulfone monomer, 5, it is possible to control the ratios of the leaving groups, in this case the fluorine and the chlorine, with the right solvent and temperature. Alternating the temperatures will alter the substitution of the leaving groups, thus forming a polymer backbone of gradient characteristics. Initial reactions to determine the relative rates of displacement of the chlorine and fluorine atoms were carried out using a model compound, 1, which possessed the chlorine and fluorine leaving groups, but not the phenol …

A series of new sulfonated poly(arylene ether)s containing one sulfonic acid group, located on a pendant phenyl sulfonyl moiety, at every repeat unit was synthesized using 3-sulfonated-3', 5'-difluorophenyl Sulfone, 4, and a variety of bisphenols. A conventional nucleophilic aromatic substitution (NAS) was utilized for the homopolymerization and copolymerization reactions. Polymerization reactions of 4 in NMP at 180 °C provided the corresponding linear sulfonated poly(arylene ether)s, s-PAEs, with number average molecular weight, Mn, ranging from 10,000 to 38,000 Daltons. The present study was aimed at providing data to be used at predicting exposure-based effects of 2,4,6-trinitrotoluene (TNT) aged in soil on endpoint organisms representing two trophic levels. These data can be used to define criteria or reference values for environmental management and conducting specific risk assessment. Long-term exposure tests were conducted to evaluate sublethal toxicity and uptake of aged soil-based explosives, with TNT as the main contaminant. In these tests, plants were exposed for 55 d, and biomass and explosives residues were determined. Worms were exposed for 28 and 42 d, and biomass, number, and tissue residues were determined. …

Loess is one of the most extensive surficial geologic deposits in mid-continental North America, particularly in the central Great Plains region of Nebraska. Last-glacial-age loess (Peoria Loess) reaches its greatest known thickness in the world in this area. New stratigraphic, geochronologic, mineralogic, and geochemical data yield information about the age and provenance of Peoria Loess, as well as evaluation of recent climate models.

Thin films of amphiphilic vinylidene fluoride oligomers prepared by Langmuir–Blodgett deposition on silicone substrates were investigated by comparing experimental and theoretical mid-infrared (IR) spectra. The experimental spectra were obtained using infrared spectroscopic ellipsometry. Theoretical spectra were calculated using density functional theory. Excellent correspondence of major IR bands in both data sets shows that the molecular backbone is oriented with the long axis normal to the substrate plane. This is in contrast to poly vinylidene fluoride[1] LB films, in which the polymer chains are parallel to the substrate.

This can occur, in part, by modulation of glial function following activation by soluble or insoluble modified alphasynuclein (α-syn), a chief component of Lewy bodies that is released from affected dopaminergic neurons. α-Syn is nitrated during oxidative stress responses and in its aggregated form, induces inflammatory microglial functions. Elucidation of these microglial function changes in PD could lead to new insights into disease mechanisms. To this end, PD-associated inflammation was modeled by stimulation of microglia with aggregated and nitrated α-syn. MUC1, a transmembrane mucin, is a key modulator of several signaling pathways that affect oncogenesis, motility, and cell morphology. The interaction of MUC1 cytoplasmic tail (MUC1CT) with signal transducers and its nuclear translocation and subsequent biological responses are believed to be regulated by phosphorylation status, but the precise mechanisms by which this occurs remain poorly defined. We detected a novel association between the Met receptor tyrosine kinase and the MUC1CT. Met catalyzed phosphorylation of tyrosine at YHPM in the MUC1CT. In this study we explore the strategies that undergraduate and graduate chemistry students use when engaged in classification tasks involving symbolic and microscopic (particulate) representations of different chemical reactions. We were specifically interested in characterizing the basic features to which students pay attention when classifying chemical reactions at the symbolic and microscopic levels. We identified the categories that students create when classifying chemical reactions, and compared the performance in simple classification tasks of students with different levels of preparation in the discipline. A series of synthetic molecules combining a geranyl backbone with a heterocyclic or oxime head group are quorum-sensing molecules that block the yeast to mycelium transition in the dimorphic fungus Candida albicans. A number of the analogs have an IC50 ≤ 10 μM, a level of potency essentially identical to the natural quorum sensing signal, the sesquiterpene farnesol. Two of the most potent analogs, neither toxic toward healthy mice, display remarkably different effects when co-administered with C. albicans. While neither offers protection from candidiasis, one analog mimics farnesol in acting as a virulence factor, whereas the other …

The level of mycotoxin fumonisins in corn-based food and feed collected from Linxian County, a high-risk area for esophageal cancer in China, has been analyzed using high-performance liquid chromatographic coupled with evaporative laser scattering detector (HPLC-ELSD). A total of 104 corn kernel samples were obtained from local households, granaries, wholesale markets (central markets), and retail markets (stores and supermarkets). Fumonisin B1 (FB1) was detected in the samples from households, granaries, central markets, and stores, with a positive rate of 61.5%, 50%, 33.3%, and 17%, respectively. Evidence from a number of vertebrate taxa suggests that modifications of hemoglobin (Hb) function may often play a key role in mediating an adaptive response to high altitude hypoxia. The respiratory functions of Hb are a product of the protein’s intrinsic O2-binding affinity and its interactions with allosteric effectors such as protons, chloride ions, CO2, and organic phosphates. Here we review several case studies involving high altitude vertebrates where it has been possible to identify specific mechanisms of Hb adaptation to hypoxia. The respiratory functions of Hb are a product of the protein’s intrinsic O2-binding affinity and its interactions with allosteric effectors such as protons, chloride ions, CO2, and organic phosphates. Here we review several case studies involving high altitude vertebrates where it has been possible to identify specific mechanisms of Hb adaptation to hypoxia. This was the fourth in a series of symposia designed to strengthen the links between Latin American and US and European scientists in the areas of surface science and catalysis. The maturation of transfer RNA (tRNA) involves extensive chemical modification of the constituent nucleosides and results in the introduction of significant chemical diversity to tRNA. Many of the pathways to these modified nucleosides are characterized by chemically complex transformations, some of which are unprecedented in other areas of biology. To illustrate the scope of the field, recent progress in understanding the enzymology leading to the formation of two distinct classes of modified nucleosides, the thiouridines and queuosine, a 7-deazaguanosine, is reviewed. This study describes the antibacterial properties of synthetically-produced mixed aryl alkyl disulfide compounds as a means to control the growth of Staphylococcus aureus and Bacillus anthracis. Some of these compounds exerted strong in vitro bioactivity. Our results indicate that among the twelve different aryl substituents examined, nitrophenyl derivatives provide the strongest antibiotic activities. This may be the result of electronic activation of the arylthio moiety as a leaving group for nucleophilic attack on the disulfide bond. Small alkyl residues on the other sulfur provide the best activity as well, which for different bacteria appears to be somewhat dependent on …

We report the synthesis and characterization of amino acid ester based chiral ionic liquids, derived from L- and D-alanine tert butyl ester chloride. The synthesis was accomplished via an anion metathesis reaction between commercially available L- and D-alanine tert butyl ester chloride using a variety of counterions such as lithium bis (trifluoromethane) sulfonimide, silver nitrate, silver lactate, and silver tetrafluoroborate. Both enantiomeric forms were obtained as confirmed by bands of opposite sign in the circular dichroism spectra. Polymer-based microelectrophoresis was investigated to analyze known (mutation detection) and unknown (mutation scanning) low-abundant mutations in genomic DNA with high diagnostic value for colorectal cancers. For our mutation detection assays, point mutations in the K-ras oncogene were identified using the ligase detection reaction (LDR). For the mutation scanning assay, which searches for sporadic mutations, an EndoV-LDR assay was utilized with mutations in the p53 tumor suppressor gene used as a model. A poly(methylmethacrylate), PMMA, microchip filled with a 4% linear polyacrylamide (LPA) gel was used to electrophoretically sort products formed from LDRs, which produced oligonucleotides <65 bp in length. Using microchip electrophoresis with the LPA, a 44 bp ligation product was resolved from a 100-fold molar excess of unligated primers (25 bp) in approximately 120 s, which was ~17 times faster than conventional capillary gel electrophoresis. In order to simplify the electrophoretic process and further reduce development time, the LDR products were sorted in the absence of the sieving gel using free solution conjugate electrophoresis (FSCE). FSCE incorporated polyamide “drag-tags” onto LDR primers, which provided DNA fragment mobilities in free solution that were dependent upon their size. LDR/drag-tagged (LDR-dt) products could be formed in a multiplexed format for mutant-to-wild-type ratios as low as 1 to 100 with single base resolution. Separations were conducted using capillary array electrophoresis (CAE) and PMMA microchips filled with only a TRIS buffer. Analysis times for the LDR-dt products were less than 11 min using CAE and ~85 s for PMMA microchips with high reproducible migration times within and between microchips. PMMA-based microchips were also evaluated for the identification of sporadic mutations using an endonuclease V – LDR (Endo V/LDR) technique. Endo V cleaves heteroduplexed DNA one base 3’ of single-base mismatched sites as well as nicking DNA at some matched sites as LDR reseals miscleaved sites to reduce false positive signals. Results suggested that Endo V/LDR products from p53 mutations could be successfully separated and detected using a PMMA microfluidic chip filled with a sparsely cross-linked replaceable polyacrylamide gel in less than 6 min, which was approximately 10-fold shorter compared to CAE.

The synthesis of novel multifunctional oligosaccharide tags with amino, azido, and alkyne termini is described. Tags with an amino terminus can be introduced into the carbohydrate at the reducing end through reductive amination, while 1,3-dipolar cycloaddition (“click chemistry”) can be used to label azido sugars with oligosaccharide tags having an alkyne terminus and vice versa.

We designed multiple Small Cluster Oligosaccharide Mimetics (SCOMs) - potential glycosidase inhibitors - to be metabolically stable and small enough to enter cells or bacteria. Therefore, minimal scaffolds (urea, amide, ammonia) or simply non-glycosidic linkages of carbohydrate structures were central to our synthetic strategy, including: (a) coupling of several natural carbohydrate precursors; (b) total syntheses of aminomethyl tetrahydropyrans and their chiral amides with quinic acid; (c) glycopyranosyl cyanide reduction to prepare crowded clusters on a urea scaffold; (d) total syntheses via cycloadditions leading to amide-linked C-glycosides; (e) reduction of nitromethyl C-glycosides; and (f) a synthesis of hydroxylated 1,2- cyclohexanedicarboxylic acids.

There is a global need to find a permanent and readily implemented solution to the problem of heavy metal pollution in aqueous environments. A dithiol compound, benzene-1,3-diamidoethanethiol (BDETH2), also known as N,N'-bis(2-mercaptoethyl) isophthalamide or N,N'-bis(2-mercaptoethyl)-1,3-benzenedicarboxamide, capable of binding divalent metal ions, has been synthesized and characterized. A broad range of BDET-metal compounds, spanning the periodic chart, has been prepared and characterized by IR, MS, EA, Raman, XAFS and TGA. The characteristics of the BDET-M compounds were determined through secondary reactions.
