Metalloglycomics: Investigating The Interactions Of Metal Complexes With Heparan Mimetics, Wyatt Johnson
Metalloglycomics: Investigating The Interactions Of Metal Complexes With Heparan Mimetics, Wyatt Johnson
Theses and Dissertations
Proteoglycans containing Heparan Sulfate (HS), a sulfated glycosaminoglycan (GAG), play a major role in the cell signaling process, interacting with many different proteins. HS is over expressed on the surface of many cancer cells. Enzymatic cleavage of HS-GAGs by heparanase causes release of angiogenic growth factors leading to tumor cell migration. Heparanase is also over-expressed in tumors with significant correlation between metastatic potential and heparanase activity. Proteoglycans and their associated enzymes are thus significant drug targets of high biological relevance.
A functional consequence of strong PPC-HS binding has been shown in proof-of-concept studies confirming inhibition of the model pentasaccharide, Fondaparinux, …

There has been an increase in use of Traditional Chinese Medicine (TCM) in the United States because they are less expensive and believed to be more effective with less adverse effects in comparison to traditional pharmaceutics. Therefore, sales have increased in the US, despite articles and case studies demonstrating the dangers, such as injury and death, related to TCM, stemming from improper labelling, toxic contaminants, and, in some cases, the presence of pathogenic bacteria. The aim of this study was to perform a survival experiment to demonstrate the importance of proper herbal brewing technique and to conduct a molecular and …

Use Of Surface Enhanced Raman Spectroscopy For The Detection Of Bioactive Lipids, Christopher M. Ohlhaver
Theses and Dissertations
The detection and analysis of lipids in biological matrices for clinical applications poses many challenges, but rapid and reliable detection will prove invaluable for clinical diagnosis. Herein, we report the application of drop-casted Ag nanoplatelets as surface enhanced Raman scattering (SERS) substrates for qualitative detection of 20-hydroxyeicosatetraenoic acid (20-HETE), which is a potential biomarker for diagnosis of hypertensive disorders. Biomarker peaks of 20-HETE can be reliably detected and differentiated from those of the structurally similar lipids (arachidonic acid, eicosapentaenoic acid, and docosahexaenoic acid) commonly found in human blood, even 1 pM concentrations. Additionally, one study mixed 20-HETE with three structurally …

Theses and Dissertations
Nanomaterials, typically less than 100 nm size in any direction have gained noteworthy interest from scientific community owing to their significantly different and often improved physical properties compared to their bulk counterparts. Semiconductor nanoparticles (NPs) are of great interest to study their tunable optical properties, primarily as a function of size and shape. Accordingly, there has been a lot of attention paid to synthesize discrete semiconducting nanoparticles, of where Group III-V and II-VI materials have been studied extensively. In contrast, Group IV and Group IV-V based nanocrystals as earth abundant and less-non-toxic semiconductors have not been studied thoroughly. From the …
Metal-Organic Frameworks And Graphene-Based Support Materials For Heterogeneous Catalysis, Andrew Lin
Metal-Organic Frameworks And Graphene-Based Support Materials For Heterogeneous Catalysis, Andrew Lin
Theses and Dissertations
Nanoparticles are involved in a broad range of applications, including heterogeneous catalysis. Nanoparticles tend to quickly lose their well-defined shapes and facets due to aggregation under duress such as heat.
A series of highly studied materials are explored as support materials for nanoparticle supports. These supports include metal-organic frameworks (MOF), graphene oxide (GO), and a MOF-PRGO (partially reduced graphene oxide) hybrid. The inclusion of a support with the palladium increased lifespan of the catalyst by separation of nanoparticles. The choice of support material not only allowed for supporting of palladium nanoparticles, but allowed for rational catalyst synthesis in order to …
Heteroatom Doped Porous Carbon For Alternative Energy Conversion And Storage Systems, Fatema A. Choudhury
Heteroatom Doped Porous Carbon For Alternative Energy Conversion And Storage Systems, Fatema A. Choudhury
Theses and Dissertations
Abstract
The electrocatalysis of oxygen plays a significant role in several electrochemical energy storage and conversion systems including metal−air batteries, fuel cells, electrocatalytic and photocatalytic water splitting. The sluggish kinetics and complex reaction mechanism of this cathodic oxygen reduction reaction (ORR) affect the performance and practical application of such renewable energy technologies. To address this limiting factor, a suitable electrocatalyst is required for ORR. In general, platinum or highly dispersed Pt-based nanoparticles on carbon black are considered as the best ORR catalyst. But platinum being very scarce and expensive tends to increase the cost. Moreover, platinum-based catalysts are prone to …
Water Dynamics And The Effect Of Static And Alternating Electric Fields, Mohammadmahdi Shafiei Alavijeh
Water Dynamics And The Effect Of Static And Alternating Electric Fields, Mohammadmahdi Shafiei Alavijeh
Theses and Dissertations
Having a net dipole moment, water molecules tend to align with an external electric field. The re-orientation of water molecules to align with the field direction can result in structural and dynamic changes in liquid water. Studying these changes can help us to understand the role of an E-field in many biological systems, chemical reactions, and many technological advancements.
In short, the application of static electric fields causes molecules to stay aligned with the field, so, fewer hydrogen bonds break, and molecules have slower dynamics. This type of field can be used when the mobility of water molecules needs to …
The Investigation Of Oxidative Addition Reactions Of Metal Complexes In Cross-Coupling Catalytic Cycles Based On A Unique Methodology Of Coupled Ion/Ion-Ion/Molecule Reactions, Mariah L. Parker
Theses and Dissertations
Popular catalytic cycles, such as the Heck, Suzuki, and Negishi, utilize metal centers that oscillate between two oxidation states (II/0) during the three main steps of catalysis: reductive elimination, oxidative addition, and transmetallation. There has been a push to use less toxic, cheaper metal centers in catalytic cycles, leading to interest in first-row transition metals, such as nickel and cobalt. With these metals, the cycles can potentially pass through the +1 oxidation state, which acts as reactive intermediates, undergoing oxidative additions to form products, potentially with radical characteristics. The oxidative addition steps of catalytic cycles are critical to determining overall …

Experimental Evaluation Of Uranyl Transport Into Mesoporous Silica Gel Using Fluorescence, Brandon M. Dodd
Theses and Dissertations
This research investigated parameters that can affect the use of nanoporous silica gel as a media for accumulating a detectable amount of uranium. The unique fluorescence of the Uranyl (UO22+) ion was used to evaluate the transport kinetics and accumulation within silica gel in a static fluid and under pressure driven flow. The addition of fluid flow decreased the time constant from on the order of an hour to approximately 2s with a very low fluid velocity of 0.36cm/s. The 0.36cm/s fluid velocity was found to be the critical velocity above which there was no gain in …

Nitrogen Rich Porous Organic Frameworks: Proton Conduction Behavior Of 3d Benzimidazole And Azo-Linked Polymers, Michael J. Anhorn
Theses and Dissertations
Nitrogen-rich porous organic frameworks show great promise for use as acid-doped proton conducting membranes, due to their high porosity, excellent chemical and thermal stability, ease of synthesis, and high nitrogen content. Aided by very high surface area and pore volume, the material has the ability to adsorb high amounts of H3PO4 into its network, which creates a proton rich environment, capable of facile proton conduction. The morphology and chemical environment, doping behavior, and proton conduction of these materials were investigated. With such high acid-doping, ex-situ studies revealed that under anhydrous conditions, PA@BILP-16 (AC) produced a proton conductivity …

The formation of boron–nitrogen (B–N) bonds has been widely explored for the synthesis of small molecules, oligomers, or linear polymers; however, its use in constructing porous organic frameworks remains very scarce. In this study, three highly porous diazaborole-linked polymers (DBLPs) have been synthesized by condensation reactions using 2,3,6,7,14,15-hexaaminotriptycene and aryl boronic acids. DBLPs are microporous and exhibit high Brunauer–Emmett–Teller surface area (730–986 m2 g−1) which enable their use in small gas storage and separation. At ambient pressure, the amorphous polymers show high CO2 (DBLP-4: 4.5 mmol g−1 at 273 K) and H2 (DBLP-3: 2.13 wt% at 77 K) uptake while …

Current atherosclerosis treatment strategies primarily focus on limiting further cholesteryl esters (CE) accumulation by reducing endogenous synthesis of cholesterol in the liver. No therapy is currently available to enhance the removal of CE, a crucial step to reduce the burden of the existing disease. Given the central role of hepatic cholesteryl ester hydrolase (CEH) in the intrahepatic hydrolysis of CE and subsequent removal of the resulting free cholesterol (FC), in this work, we applied galactosefunctionalized polyamidoamine (PAMAM) dendrimer generation 5 (Gal-G5) for hepatocyte-specific delivery of CEH expression vector. The data presented herein show the increased specific uptake of Gal-G5/CEH expression …

In this article, we report the lowest-temperature CO oxidation catalyst supported on metal–organic frameworks (MOFs). We have developed a facile, general, and effective approach based on microwave irradiation for the incorporation of Pd nanoparticle catalyst within Ce-MOF. The resulting Pd/Ce-MOF material is a unique catalyst that is capable of CO oxidation at modest temperatures and also of efficient uptake of the product CO2 gas at low temperatures. The observed catalytic activity of this material toward CO oxidation is significantly higher than those of other reported metal nanoparticles supported on MOFs. The high activity of the Pd/Ce-MOF catalyst is due to …

Roles Of Free Electrons And H2o2 In The Optical Breakdown-Induced Photochemical Reduction Of Aqueous [Aucl4]-, Victoria K. Meader, Mallory G. John, Collin Rodrigues, Katharine M. Tibbetts
Chemistry Publications
Free electrons and H2O2 formed in an optical breakdown plasma are found to directly control the kinetics of [AuCl4]− reduction to form Au nanoparticles (AuNPs) during femtosecond laser-assisted synthesis of AuNPs. The formation rates of both free electrons and H2O2 strongly depend on the energy and duration of the 800 nm laser pulses over the ranges of 10−2400 μJ and 30−1500 fs. By monitoring the conversion of [AuCl4]− to AuNPs using in situ UV−vis spectroscopy during laser irradiation, the first- and second-order rate constants in the autocatalytic rate law, k1 and k2, were extracted and compared to the computed free …

Chemistry Publications
Hierarchically porous silica monoliths are obtained in the two-step Nakanishi process, where formation of a macro microporous silica gel is followed by widening micropores to mesopores through surface etching. The latter step is carried out through hydrothermal treatment of the gel in alkaline solution and necessitates a lengthy solvent exchange of the aqueous pore fluid before the ripened gel can be dried and calcined into a mechanically stable macro mesoporous monolith. We show that using an ethanol water (95.6/4.4, v/v) azeotrope as supercritical fluid for mesopore etching eliminates the solvent exchange, ripening, and drying steps of the classic route and …

A high mobility group box 1 (HMGB1) protein has been reported to recognize both 1,2-intrastrand crosslinked DNA by cisplatin (1,2-cis-Pt-DNA) and monofunctional platinated DNA using trans-[PtCl2(NH3)(thiazole)] (1-trans-PtTz-DNA). However, the molecular basis of recognition between the trans-PtTz-DNA and HMGB1 remains unclear. In the present work, we described a hydrogen/deuterium exchange mass spectrometry (HDX-MS) method in combination with docking simulation to decipher the interactions of platinated DNA with domain A of HMGB1. The global deuterium uptake results indicated that 1-trans-PtTz-DNA bound to HMGB1a slightly tighter than the 1,2-cis-Pt-DNA. The local deuterium uptake …
Theoretical And Experimental Analysis Of Optical Properties Of Defects In Gan:, Ibrahima Castillo Diallo
Theoretical And Experimental Analysis Of Optical Properties Of Defects In Gan:, Ibrahima Castillo Diallo
Theses and Dissertations
Using the Heyd-Scuseria-Ernzherof (HSE06) hybrid functional method along with photoluminescence experimental measurements, we analyze the properties of intrinsic defects such as vacancies, interstitials, antisites, and common complexes. By using configurational coordinate diagrams, we estimate the likelihood of defects to be radiative or non-radiative. Our calculations show that gallium vacancies exhibit a large magnetic moment in the neutral charge state and are most likely non-radiative. We also investigate the correlation between the observed infrared PL bands created in 2.5 MeV electron-irradiated GaN samples and the formation of native defects. It is found that gallium-nitrogen divacancies are possible sources of the broad …

Permanent magnets are classified as hard magnetic materials with the main purpose of generating flux for applications such as electric motors, turbines, and hard drives. High coercivity, magnetic remanence, and saturation values with high stability are some of the requirements for permanent magnets. Rare-earth magnets including neodymium and samarium based magnets are known to have superior magnetic properties due to their high magnetocrystalline anisotropy. However, due to the price of rare-earth materials development of alternate permanent magnets composed of inexpensive materials is an ongoing process. Previously cobalt carbide (CoxC) have shown promise as a potential rare-earth free magnet …

Cancer chemotherapy results in systematic damage as the drugs used are also toxic to benign tissue. Sensitizing a cancer cell to therapy by interfering with the DNA repair mechanisms would decrease overall toxicity, as the necessary dosage of chemotherapy drugs would be lowered. The Hartman lab developed a peptide (8.6) that binds with a KD of 1 μM to the C-terminal domain of breast cancer associated protein (BRCA1), blocking homologous recombination. The crystal structure of the peptide shows the tyrosine and threonine residues are close together, suggesting that by cyclizing these positions, the peptide may already be constrained into its …

Theses and Dissertations
The use of porous sorbents for physisorptive capture of CO2 from gas mixtures has been deemed attractive due to the low energy penalty associated with recycling of such materials. Porous organic polymers (POPs) have emerged as promising candidates with potential in the treatment of pre- and post- fuel combustion processes to separate CO2 from gas mixtures. Concurrently, significant advances have been made in establishing calculation methods that evaluate the practicality of porous sorbents for targeted gas separation applications. However, these methods rely on single gas adsorption isotherms without accounting for the dynamic gas mixtures encountered in real-life applications. …

Gradient surfaces exhibit a variation in functionality along the length of the surface. One method for preparing gradients is controlled-rate infusion (CRI). In Part 1 of this work, CRI was used to prepare gradients for the purpose of separating transition and heavy metals. Initial work on this project was focused on controlling the retention of the metal ions by varying the number of amine groups, aminoalkoxysilane concentration, and the infusion time. The retention factors of four metal ions varied predictably with increasing number of amine groups, increasing aminoalkoxysilane concentration, and increasing infusion time, producing small but useful changes in the …

Surface chemical gradients are materials that exhibit continuous, gradually varying chemical or physical properties along and across the length of a substrate. As a result, each point on the gradient surface can represent an individual sample. They are broadly classified as chemical and physical gradients depending upon the properties that the gradient exhibits. A physical gradient involves a continuous variation of physical properties such as surface roughness and film porosity on the micrometer scale. Chemical gradients involve a gradual variation of chemical properties such as polarity, acidity and basicity, etc. Such gradients have found various applications in cell adhesion, nanoparticle …
