The main purpose of this research was to develop a gel assay for analysis of casein samples for ultimate application for studies of in vitro models for stomach and intestinal processing of dairy products in adults and infants. It is proposed that casein completely dissolves and is digestible in simulated gastric, and is somewhat soluble and digestible in simulated intestinal environments of adults and infants. The following experimental methods were devised to thoroughly examine behavior and properties of casein originating from different sources in range of solvents and varying pH values. Here, we aim to further the potential use of surface-enhanced Raman spectroscopy (SERS) in analyzing hair dyes for use in forensic investigations. Current methods include light microscopy and thin-layer chromatography (TLC). However, these techniques can be destructive to the hair samples, do not have high discriminatory power, and have had problems with giving inconclusive results. Raman spectroscopy, on the other hand, is a fast, simple technique with a high discriminatory power. This study investigates the interactions of both purine (adenine and guanine) and pyrimidine (cytosine, thymine, and uracil) nucleobases with a pair of silver atoms (Ag2). Full geometry optimizations were performed on several structures of each nucleobase/Ag2 complex and the corresponding isolated monomers using the M06-2X density functional theory (DFT) method with a correlation consistent triple-ζ basis set augmented with diffuse functions on all atoms and a relativistic pseudopotential on Ag (aug-cc-pVTZ for H, C, N, and O and aug-cc-pVTZ-PP for Ag; denoted aVTZ). Harmonic vibrational frequency computations indicate that each optimized structure corresponds to a minimum on the M06-2X/aVTZ potential …

O-linked β-N-acetylglucosamine (O-GlcNAc) post-translational modification (O- GlcNAcylation), plays a vital role in many metabolic pathways, including those of numerous diseases. New research methods and basic functional roles of O-GlcNAc post- translational modification on metabolism, human health, and cancer are quickly developing, but a vast amount of research remains on the specificity of this pathway towards specific proteins. This investigation studies the interactions between various nitroaromatic high energy density materials (HEDMs) and a pair of silver atoms (Ag2). Telomeres are repeated nucleotide sequences that cap the ends of each chromosome in eukaryotes. Telomeres are among the earliest genomic sequences to be degraded during apoptosis, potentially providing a biomarker of cell death. To study this, we quantified telomeres in serum from Glioblastoma and non-cancer patients. Cell-free DNA was isolated by centrifugation to remove intact cells, and purified using a QIAamp DNA Blood Midi Kit (Qiagen). Cell-free telomeric (cf-tel) and cell-free actin (cf-actin) DNA were analyzed with quantitative PCR. Total cell-free DNA was measured with PicoGreen assays. 1,2,3-triazole is used as a building block in many pharmaceutical drugs. The molecule has several derivatives that are useful in other areas including treating fungal infections and in dealing with some environmental issues. One of its derivatives, benzotriazole, is commonly used as an inhibitor of corrosion through the formation of a copper-benzotriazole complex. One area of particular interest in studying triazoles is the ability of these molecules to tautomerize. Calmodulin is an intensely studied, intracellular, calcium-binding protein that is involved in diverse physiological processes. When bound to calcium, calmodulin undergoes a conformational change that allows it to bind specific target peptides with high affinity, one being the calmodulin binding region of myosin light chain kinase, herein called M13. This high affinity, calcium dependent association has made it a desirable choice for use in a biomaterial for drug delivery for non-steroidal anti- inflammatory drugs (NSAIDs). Pain, although subjective, is an unpleasant sensation caused by a noxious stimulus, that effects millions of people every year. Currently, the most frequently prescribed class of analgesics for moderate to severe acute and sometime chronic pain are opioid analgesics, most commonly ?-opioid receptor (MOP) agonists, such as oxycodone, morphine, and fentanyl. Although these drugs provide significant pain relief, they are also associated with unwelcome side effects such as constipation, addiction, physical dependence, respiratory depression, and physiological tolerance that leads to hyperalgesia. Statin drugs are commonly prescribed to lower cholesterol levels in patients, but recently statins have been under investigation as a potential anti-cancer drug. Statins are HMG-CoA reductase inhibitors and subsequently inhibit the mevalonate pathway and cholesterol synthesis. Since cholesterol trafficking is vital in the growth and metastasis of cancer, statin drugs are being studied to explore their effects on cancer cells. The free radical chlorine dioxide (ClO2) is a strong oxidizing agent that is used to purify drinking water and medical equipment. The existing literature shows discrepancies in the explanation of the solvent dependent properties of ClO2. The following thesis investigates these properties by integrating integrates inorganic synthesis, spectroscopic analyses, and computational methods. ClO2/water hydrogen-bonded networks were computed with the DFT method B3LYP with 6-31G(d,p) and 6-311++G(2df, 2pd) basis sets. Raman frequency computations were performed on local minima geometries. With 65% of land area covered in forest, Mississippi is a leader in the timber industry. Whereas forestry provides numerous benefits for humans and wildlife, forest harvest has been hypothesized to increase mercury (Hg) mobilization to aquatic systems. In a forest, Hg mostly accumulates in the upper layer of soil, where associates with organic matter. This top soil is exposed after clear-cutting and thus more susceptible to runoff. Harvest may also change the net balance between Hg methylation and demethylation by microorganisms through changes in redox conditions and addition of carbon sources from decaying logging residues. A paper published by Joshi et al. showed lower than expected yields in mono- and di-iodination of 3,5-dichloroanisole in dichloromethane using silver salts. One possibility for this could be that the starting material was being tri-iodinated and precipitating out of the solution. We used similar reaction conditions in attempts to prepare and isolate triiodinated product, but attempts of recrystallization from ethanol, DMSO, and 2- methoxyethanol showed no signs of it. Removal of silver iodide from the solids formed during the reaction using aqueous potassium iodide resulted in complete dissolution of the entire crude product, suggesting no organic product as well. …

The ability of perylenebisimides to interrupt the structures of the G4 DNA on the end of telomeres has allowed for the observation of how the variable-regions of the perylenebisimide can have differing effects on anticancer properties. The purpose of this experiment was to synthesize a perylenebisimide with the intentions of observing its intercalating properties with G-quadruplex DNA. An unknown fungal species was cultured from necrotic Brachiaria eruciformis leaves found in Oxford, Mississippi. The fungus was identified via molecular techniques as Pyricularia grisea. This fungus was grown on Czapek Dox broth culture medium for 21 days under continuous light at 24 0C. The culture filtrate was extracted with ethyl acetate and concentrated. The major bioactive compound, namely pyrichalasin H, was isolated using TLC-guided fractionation and identified via spectroscopic techniques. The compound was then tested for bioactivity on monocotyledons, dicotyledons and agriculturally important fungal species. The isoenzymatic form of microsomal cytochrome P450, CYP2E1, is involved in the formation of oxygen free radicals, specifically hydroxyethyl ether. Chronic alcohol abuse has been found to induce larger levels of CYP2E1 in the liver, thus increasing production of these harmful free radicals. Generation of the hydroxyethyl ether oxygen free radicals occurs during the metabolism of ethanol in microsomes. Panobinostat is the first approved histone deacetylase inhibitor for the treatment of multiple myeloma, along with other drugs such as Velcade and Dexamethasone. Halogen bonding is a unique and relatively unexplored noncovalent interaction that has recently garnered considerable attention due to its applications in materials development. The thiolate-protected gold nanocluster (AuNP) Au38(SCH2CH2Ph)24 was studied using Raman spectroscopy with particular attention to the vibrational modes of the gold-sulfur interface. Such studies of AuNPs are non-trivial, as these materials are weak scatterers that burn and fluoresce readily. Previous works have successfully studied this class of materials using a rolling circle filter to subtract fluorescence backgrounds, but we set out to develop a method which would suppress fluorescence using more fundamental means. Cadherins are a family of transmembrane proteins that dimerize to form junctions between cells in the presence of calcium. Due to the necessity for proper cell adhesion during cell differentiation in development, we are examining whether binding of other metallic divalent cations would disrupt calcium-sensitive dimerization of neural cadherin in vitro. Specifically, we are studying the effect of cadmium(II) on the linkage between calcium binding, dimerization, and dimer disassembly. Cadmium toxicity became recognized during the Industrial Revolution, with particular attention gained during the Itai-Itai disease outbreak in China during the 1910s. Gold nanoparticles have wide range of applications in the field of sensing, imaging, cancer therapy, and other biomedical applications. It is desired to have monodisperse and atomically precise gold nanoparticles to have better control and effective utilization of that nanoparticle. The nanoparticles that are less than 2 nm in size with precise number of atoms and ligands (± 0 atoms) which exhibit unique physical, chemical and optoelectronic properties are called as gold nanomolecules.
