The criminal use of explosives has increased in recent years. Political instability and the wide spread access to the internet, filled with "homemade recipes," are two conjectures for the increase. C-4 is a plastic bonded explosive (PBX) comprised of 91% of the high explosive RDX, 1.6% processing oils, 5.3% plasticizer, and 2.1% polyisobutylene (PIB). C-4 is most commonly used for military purposes, but also has found use in commercial industry as well. Although environmental monitoring of polycyclic aromatic hydrocarbons (PAH) is an essential step to prevent human exposure to contaminated sites, it provides little information on the actual human uptake and subsequent risks. To this end, urine analysis of short-term biomarkers such as PAH metabolites fill an important niche. The general approach follows the sequence of urine hydrolysis, sample clean-up and pre-concentration, chromatographic separation and determination. Whereas chromatographic methods are based on well established laboratory techniques, the development of easy-to-use, cost-effective and large sample throughput techniques is becoming increasingly relevant to investigate adverse PAH effects on large human populations. Heavy metal contamination of soils, sediments and groundwater presents an ongoing source of hazardous and persistent environmental pollution. How best to remediate these contaminants is the impetus of continuing research efforts. Methods include containment, ex situ and in situ techniques. A successful in situ method utilizing a combination of emulsified liquid membranes, ELM, and zero-valent metal, ZVM, and bimetals has demonstrated impressive heavy metal reduction in 100 ppm solutions of Cd, Cu, Ni, Pb, Cr and U. Elemental analysis in forensic laboratories can be tedious and many trace evidence items are not analyzed to determine their elemental composition. Presently, scanning electron microscopy-energy dispersive x-ray spectroscopy (SEM-EDS) is the primary analytical tool for determining the elemental composition of trace evidence items. Three-dimensional (3D) optical data storage based on two-photon processes provides highly confined excitation in a recording medium and a mechanism for writing and reading data with less cross talk between multiple memory layers, due to the quadratic dependence of two photon absorption (2PA) on the incident light intensity. The capacity for highly confined excitation and intrinsic 3D resolution affords immense information storage capacity (up to 1012 bits/cm3). Fire incidents are a major contributor to the number of deaths and property losses within the United States each year. Fire investigations determine the cause of the fire resulting in an assignment of responsibility. Current methods of fire debris analysis are reviewed including the preservation, extraction, detection and characterization of ignitable liquids from fire debris. Leak rates were calculated for the three most common types of fire debris evidence containers. The consequences of leaking containers on the recovery and characterization of ignitable liquids were demonstrated. Simplified Low Copy Number Dna Analysis By Post Pcr Purification, Pamela Smith

Frequently evidentiary items contain an insufficient quantity of DNA to obtain complete or even partial DNA profiles using standard forensic gentotyping techniques. Here, various methods of post PCR purification were evaluated for their effects on the sensitivity of fluophore-based allelic detection. A method of post PCR purification is described which increases the sensitivity of standard 28 cycle PCR such that low copy number DNA templates (<100 pg DNA) can be analyzed. Full profiles were consistently obtained with as little as 20 pg template DNA without increased cycle number. In mock case type samples with dermal ridge fingerprints, genetic profiles were obtained by amplification with 28 cycles followed by post-PCR purification whereas no profiles were obtained without purification of the PCR product. Allele drop-out, increased stutter, and contamination (allele drop-in) typical of LCN analysis were observed. A single incident of contamination was observed in a reagent blank (not duplicated upon re-amplification) however, no contamination was observed in negative amplification controls.

Forensic fiber examination is an important part of trace evidence analysis. Fibers may be recovered from a crime scene that could link a particular suspect to the scene. Clothing fibers are most frequently encountered but automobile carpeting fibers may also be recovered. An understanding of the frequency of occurrence and the discrimination power of different analytical techniques is needed in order to better establish the evidentiary value of automobile carpet fiber evidence. Seventy-five automobile carpet fiber samples were analyzed using a series of techniques ranging from nondestructive to destructive. It is routine for the forensic scientist to obtain a genetic profile of an individual from DNA recovered from a biological stain deposited at a crime scene. In contrast, only a limited number of laboratories in the United States have the capability of performing Y-STR analysis in casework. Waste streams of hydrogen and helium mixtures are produced at the Kennedy Space Center during purging of the hydrogen systems and supply lines. This process is done prior to and after hydrogen servicing. The purged waste gases are lost to the atmosphere, resulting in an annual loss of 2 million and 0.1 million standard cubic meters of helium and hydrogen, respectively. Sediments can act as both a sink for pollutants and a source for aquatic contaminants. Natural and human disturbances of the sediments can release the contaminants to the overlying water where bottom dwelling, or benthic, organisms may be exposed through direct contact, ingestion of sediment particles, or uptake of dissolved contaminants present in the water. Remediation of trichloroethylene (TCE) and other chlorinated solvents is of great concern due to their toxicity and their persistence in the environment. Iron has been used extensively in the past decade as a subsurface reactive agent for the remediation of dense, nonaqueous-phase liquids (DNAPLs). Permeable reactive barrier walls (PRBW) have been installed at many sites around the country to treat contaminated plumes resulting from the presence of DNAPL pools. The goal of part I of this thesis was to isolate pure anti cancer compounds from the Russian plant Sanguisorba officinalis. This plant was selected for investigation because it showed 100% activity against leukemia (L1210 mouse leukemia cells) during the preliminary screening of approximately 100 Russian plant extracts. This work has resulted in the isolation of novel compound 11 using traditional chromatography techniques. Although modern medicine made great steps toward curing most diseases considered deadliest, yet cancer remains one of the major public health problems. The first part of this thesis investigates a new source of selective compounds that are potential candidates against cancer. Fifteen Russian compounds were tested in order to establish their efficiency against two types of cancer: human breast SK-Br-3 and colorectal carcinoma HT-29. Photodynamic therapy (PDT) has been investigated over the past three decades and is currently an approved therapeutic modality for skin cancer, the treatment of superficial bladder, early lung and advanced esophageal cancers, and age-related macular degeneration in a number of countries. In PDT, the absorption of light by a chromophore generates cytotoxic species such as reactive singlet oxygen, leading to irreversible destruction of the treated tissue. Novel bifunctional five-membered cyclic dithiocarbonates (1,3-oxathiolane-2-thione)s were synthesized by the reactions of the corresponding bifunctional oxiranes (epoxides) with carbon disulfide at room temperature with lithium bromide as catalyst. Full characterization of these monomers was performed including elemental analysis, proton and carbon nuclear magnetic resonance (NMR) spectroscopy, gas chromatography-mass spectroscopy, and Fourier transmission infrared (FTIR) spectroscopy. The polyaddition polymerization of 1,3-oxathiolane-2-thione with 1,4-diaminobutane at room temperature resulted in a poly(thiourethane) material. The latter undergoes crosslinking due probably to the autooxidation of the product and formation of disulfide linkages. As a class, amides are of great interest in biological studies and pharmaceutical application. In this work, podocarpic acid, a natural tricyclic diterpene, derived from Podocarpus species, has been employed to form a novel family of amide derivatives which will later be studied for their potential as new drug leads. Novel amide derivatives of podocarpic acid were synthesized from podocarpic acid in three steps. The first step involved methylation with dimethylsulfate to form methyl-O-methylpodocarpate. This step was followed by iodination with iodine to give iodomethyl-O-methylpodocarpate. Finally amidation with various aliphatic amides using a copper catalyst yielded four amide derivatives of …
