Background: Photochromism is the reversible transformation of a chemical material to another form by the absorption of electromagnetic radiation (light), where the two metastable forms have distinct absorption spectra and other properties. Photochromism in materials allows for the switching of their function solely based on irradiation with light. Polymers are used frequently as the building blocks for materials as they are versatile, multifunctional, can carry charge and be processed by solution-based deposition methods. Supramolecular polymers share the same definition as polymers with the exception that they are held together by reversible and directional non-covalent interactions such as hydrogen bonds. Synthesizing …

Additive-Free Palladium-Catalyzed Decarboxylative Cross-Coupling Of Aryl Chlorides, Ryan A. Daley, En Chih Liu, Joseph J. Topczewski
Chemistry Faculty Research & Creative Works
The cross-coupling of sodium (hetero)aryl carboxylates with (hetero)aryl chlorides proceed with 1 mol % palladium catalyst and does not require inorganic base, silver salts, or copper salts. This coupling uses two low energy partners, and the only stoichiometric byproducts are carbon dioxide and sodium chloride. The substrate scope includes less activated aryl chlorides and carboxylates (>25 examples). The palladium loading could be reduced to 0.1 mol %, and Buchwald-style precatalysts could be used.
Chemosensors Based On Higher Energy Gap Control Of Fluorescence In Conjugated Polymers, Chun-Han Wang
Chemosensors Based On Higher Energy Gap Control Of Fluorescence In Conjugated Polymers, Chun-Han Wang
LSU Doctoral Dissertations
Conjugated polymers (CPs) have been widely investigated for their remarkably high sensitivity towards various chemical detection applications. CPs can exhibit effective transduction of certain analyte binding events through changes in fluorescence. These changes include amplified fluorescence quenching ("turn-offmechanism") or an appearance of a fluorescent emission ("turn-onmechanism"). Whereas turn-offsensors can be readily designed and are widely used, amplifying turn-onsensors, from a practical standpoint, are more convenient to use but are more challenging to design.
This dissertation primarily focuses on the development and study of conjugated polymer based amplifying turn-onfluorescence chemosensors utilizing the novel “higher …

Kinetic Resolution Of Cyclic Secondary Azides, Using An Enantioselective Copper-Catalyzed Azide-Alkyne Cycloaddition, Juliana R. Alexander, Amy A. Ott, En Chih Liu, Joseph J. Topczewski
Chemistry Faculty Research & Creative Works
An enantioselective copper-catalyzed azide-alkyne cycloaddition (E-CuAAC) is reported by kinetic resolution. Chiral triazoles were isolated in high yield with limiting alkyne (up to 97:3 enantiomeric ratio (er)). A range of substrates were tolerated (>30 examples), and the reaction was scaled to >1 g. The er of a triazole product could be enhanced by recrystallization and the recovered scalemic azide could be racemized and recycled. Recycling the azide allows efficient use of the undesired azide enantiomer.
Protocol Measuring Horizontal Gene Transfer From Algae To Non-Photosynthetic Organisms, James Weifu Lee
Protocol Measuring Horizontal Gene Transfer From Algae To Non-Photosynthetic Organisms, James Weifu Lee
Chemistry & Biochemistry Faculty Publications
Horizontal gene transfer (HGT) is a natural process for an organism to transfer genetic material to another organism that is a completely different species, for example, from a blue-green alga to a non-photosynthetic bacterium. The phenomenon of HGT is not only of an interest to the science of molecular genetics and biology, but also to the biosafety issue of genetic engineering. The novel protocol reported here for the first time teaches how to measure HGT from a genetically engineered (GE) blue-green alga (gene donor) to wild-type E. coli (recipient). This novel protocol can be used to measure HGT frequency for …

The Other Sio2: Investigating Oxidation Of Alcohols Using (Nh4)2cr2o7 In Sand, John Lepore, Emily Toy, Robert Torregrosa
McNair Scholars Program
Oxidation of alcohols to aldehydes and ketones is one of the classical reactions in organic chemistry which is utilized frequently in medicine, industry, and pharmaceuticals. Classical effective reagents in oxidation include pyridinium chlorochromate, potassium permanganate, Jones reagent, bleach, and Swern conditions. There has been an increasing interest throughout the years to use supported-based reagents in tandem with these reagents, particularly with silica gel especially towards Cr-based oxidizing agents. Despite the abundance of reported reactions in the literature pertaining to silica gel, little is known about sand, despite both are silicon dioxide and exhibit similar porosity and crystallinity. It is not …

The structure of zymonic acid (systematic name: 4-hydroxy-2-methyl-5-oxo-2,5-dihydrofuran-2-carboxylic acid), C6H6O5, which had previously eluded crystallographic determination, is presented here for the first time. It forms by intramolecular condensation of parapyruvic acid, which is the product of aldol condensation of pyruvic acid. A redetermination of the crystal structure of pyruvic acid (systematic name: 2-oxopropanoic acid), C3H4O3, at low temperature (90 K) and with increased precision, is also presented [for the previous structure, see: Harata et al. (1977). Acta Cryst. B33, 210–212]. In zymonic acid, the hydroxylactone ring …
Expanding The Toolbox With Site-Specific Methods Of Bioconjugation, Tiauna S. Howard
Expanding The Toolbox With Site-Specific Methods Of Bioconjugation, Tiauna S. Howard
Seton Hall University Dissertations and Theses (ETDs)
Bioconjugation is an important tool for studying complex biological systems, with site-specificity being the major challenge. Reactions based on amino acid derived organocatalysts have been widely used in organic synthesis, particularly for the asymmetric synthesis of small molecules but this concept has not been vastly explored on biomolecules. To combat these limitations, two chemical strategies are developed to effectively attach synthetic molecules site specifically to proteins. First, a protein modification technique based on conjugation at a non-native functional handle, an aldehyde or ketone, is developed. This functional handle is chemically introduced onto the biomolecules before undergoing the organocatalyzed aldol reaction, …
Nontraditional Hydrogen Bonding In Asymmetric Lewis Acid Catalysis, Brandon Vernier
Nontraditional Hydrogen Bonding In Asymmetric Lewis Acid Catalysis, Brandon Vernier
Electronic Theses and Dissertations
In the field of asymmetric induction, there is a shift from the synthesis of reaction
specific chiral auxiliaries towards a broader mechanistic approach. Our approach is to
develop a theory of asymmetric catalyst design from first principles. The Diels-Alder
reaction of 2-methacrolein and 1,3-cyclopentadiene in the presence of 15 mole % lmenthoxy
aluminum dichloride, reported by Koga, achieved the (S)-exo-Diels-Alder
cycloadduct with 72% ee (0% ee Endo for acrolein). The dramatic change from 72% to 0%
ee is a significant fact that has been overlooked in practical organic synthesis.
In the first phase of this work, the conformational landscape of …

Natural products are widely used in the pharmaceutical industry, in agriculture, and as specialty chemicals. Methodology development focuses on optimizing the key organic reactions to access these natural products while trying to limit the overall number of synthetic steps. Key bond forming strategies are sought to provide new ways to address carbon-carbon or carbon-heteroatom bonds. The advancement of new asymmetric reactions to generate enantiopure products from achiral starting materials is a vital area of research. The objectives addressed in this dissertation include: 1) the development of a general reductive conversion of esters to ethers with a broad substrate scope accessing …

Examining Electrochemical Properties Of O-Quinone And Sulfur Adducts Using Bisphenol A-3,4-Quinone Platform, Barbara Mullen
Theses/Capstones/Creative Projects
o-Quinones are toxic electrophiles that can disrupt cellular macromolecules. Whether endogenous or xenobiotic, o-quinones are reactive compounds that are difficult to isolate. This work uses the o-quinone of bisphenol(BPA), bisphenol A-3,4-quinone(BPAQ), which can be efficiently synthesized and isolated, to study the electrochemical properties of o-quinones and their sulfur adducts in order to understand their metabolic fate. Cyclic voltammetry and equilibrium analysis were used to determine the relative oxidation potential of the BPA catechol and adducts, formed by reacting glutathione(GSH) and N-acetylcysteine(NAC) with BPAQ. BPAQ-sulfur adducts with 5-position substitution displayed lower oxidation potentials when compared to the …

Molecular Machines: Synthesis, Modeling, And Applications, Syd Kotar
Student Scholar Symposium Abstracts and Posters
While some of the smallest, most useful machines known to science are the biological molecules that keep living organisms alive, there is a growing advancement of the creation of artificial molecules that have the same function and are more efficient. Molecular machines were invented in 1983 when scientists in France created a machine formed of two interlinked molecular rings. In 2016, the Nobel prize in chemistry was awarded to Feringa, Stoddart, and Sauvage who finally documented molecular machines and put them into energy‐filled states in which their movements could be controlled. From this, molecular machines became known as a group …
A Study On The Modifications Of [2.2]Paracyclophane And Their Effects On The Enantioselectivity Of The Copper Catalyzed Cyclopropanation Reaction, Yousef Dawoud
Honors Theses
The use of chiral [2.2]paracyclophane Schiff-base ligands has been shown to induce enantioselectivity in the copper catalyzed cyclopropanation of styrene with diazoesters. 1 Previous research by Dr. Masterson tested Schiff-base ligands based on 4- amino[2.2]paracyclophane with moderate enantioselectivity. In this project, the Schiff-base N- salicylidene-4-amino[2.2]paracyclophane, was modified by way of Grignard addition to the imine carbon and its enantioselective properties were explored with the copper catalyzed cyclopropanation. The methyl Grignard addition to N- salicylidene-4-amino[2.2]paracyclophane was successful in producing the amino alcohol and the amino alcohol was characterized. The amino alcohol was produced from the R enantiomer of the Schiff-base and …

Electronic Theses and Dissertations
β-lactam antibiotics have been the most widely used drug of choice to combat infectious disease caused by bacteria. Unfortunately, their effectiveness is drastically threatened by bacterial β-lactamases. β-lactamases is responsible for the resistance to most antibiotic drugs. For decades, β-lactam β-lactamases inhibitors have been used to reduce bacterial resistance; however, in this study 1,2,4-oxadiazolidinone derivatives as a non-β-lactam β-lactamases inhibitor against TEM-1 and P99 β-lactamases. The significance of oxadiazolidinone is the prominent five-membered ring scaffold in its structure, which is configurationally stable and present in other biologically active compounds such as linezolid and avibactam. Oxadiazolidinones were synthesized by treating nitrones …

Preparation Of Enantioenriched Alkyltin Species And Their Application In Stereospecific Transformations, Glenn O. Ralph
Preparation Of Enantioenriched Alkyltin Species And Their Application In Stereospecific Transformations, Glenn O. Ralph
Dissertations, Theses, and Capstone Projects
Organometallic reagents containing the tin-carbon bond are used extensively in modern synthetic chemistry for the formation of new bonds to carbon. Over recent decades, transition metal catalyzed cross coupling reactions between two C(sp2) centers have been widely developed. However, the introduction of a C(sp3) center complicates the catalytic cycle, and opens unproductive chemical pathways which lead to isomerization, elimination, and racemization. Our lab has developed a modified-Stille reaction to combat the deleterious effects of β-hydride elimination. Our protocol enables unactivated 2° alkyl organotin nucleophiles to undergo efficient cross coupling reactions with C(sp2) electrophiles, avoiding …
Mild Synthesis Of Perylene Tetracarboxylic Monoanhydrides With Potential Applications In Organic Optoelectronics, Xizhe Zhao
Dissertations, Theses, and Capstone Projects
Perylene tetracarboxylic derivatives are considered good n-type semi-conductors. In past decades, there has been extensive study on their synthesis and electronic properties. Because of the high electron affinity, the ability to form π-π stacks, they have been widely utilized in organic photovoltaic solar cells, field-effect transistors and light-emitting diodes. Many of those applications prefer unsymmetrically substituted perylene tetracarboxylic derivatives. Perylene monoanhydrides are the most versatile intermediates for the preparation of unsymmetrically substituted perylene tetracarboxylic derivatives. In this thesis, I focused on introducing new synthesis methods for perylene monoanhydrides with labile functional group and their potential applications in organic optoelectronics.

Electronic Theses and Dissertations
Thalidomide analogues were synthesized utilizing optimized click conditions from 3-azidoglutarimide and an array of arylacetylenes or N-ethynyl/N-propargyl phthalimide derivatives. The intermediate, 3-azidoglutarimde, was pivotal and yielded a new and scalable synthesis. The reaction conditions utilized a copper sulfate/sodium ascorbate system in aqueous tetrahydrofuran to propagate the dipolar cycloaddition reactions between the azidoglutarimide and the alkynyl coupling partners. The first explored substrates were substituted arylalkynes to optimize the click reaction. Arylalkynes selected were to exhibit similar electron deficient rings to mimic phthalimide. Along with the synthesis of click thalidomide analogues, click analogue of the antiangiogenic and more potent …
An Investigation Of Alkylboron Transmetallation In Palladium-Catalyzed Suzuki Reactions, Benjamin Murray
An Investigation Of Alkylboron Transmetallation In Palladium-Catalyzed Suzuki Reactions, Benjamin Murray
Dissertations, Theses, and Capstone Projects
Through the past 40 years, carbon-carbon cross-coupling reactions have greatly enhanced the ability of chemists to synthesize C(sp2)-C(sp2) bonds and more recently C(sp2)-C(sp3) bonds. In particular, the Suzuki reaction has proven to be invaluable with its high yields, good functional group tolerance, and low toxicity of reagents. One of the component steps of the catalytic cycle of this reaction is transmetallation, in which the nucleophilic species transfers its organic component to the metal center (generally palladium). The mechanism of transfer from primary alkylboron nucleophiles was studied in the 1990's via use of …
Bridging The Molecular/Material Divide: An Investigation Into The Properties Of Polyesters, Terra Marie M. Jouaneh
Bridging The Molecular/Material Divide: An Investigation Into The Properties Of Polyesters, Terra Marie M. Jouaneh
Senior Honors Projects
By definition, polymers consist of large molecules composed of small, repeating units called monomers. These monomers give characteristic material properties to polymers, whether they be the melting point, the folding character, or the inherent stability. Many of the objects we encounter on a daily basis are composed of polymers. These polymers may be synthetic, such as the plastic in disposable bags, or natural, like glycogen in the liver. Recent developments in biotechnology have used synthetic polymers in drug delivery, contact lenses, and even organ transplants. Polyesters, in particular, have been utilized pharmacologically due to their biodegradable properties. Understandably, not all …
Heavy-Atom Tunneling In The Planar Bond Shifting Of [16]Annulene, Philip Lampkin, Jon Shezaf
Heavy-Atom Tunneling In The Planar Bond Shifting Of [16]Annulene, Philip Lampkin, Jon Shezaf
Creative Activity and Research Day - CARD
Mid-sized annulenes are known to undergo rapid π-bond shifting. Given that heavyatom tunneling plays a role in planar bond shifting of cyclobutadiene, we computationally explored the contribution of heavy atom tunneling to the planar bond shifting in the major (CTCTCTCT) and minor (CTCTTCTT) isomers of [16]annulene. (U)M06-2X/cc-pVDZ calculations yield bond-shifting barriers of ca. 10 kcal/mol. The results also reveal extremely narrow barrier widths, suggesting a high probability of tunneling for these bond-shifting reactions. Rate constants were calculated using canonical variational transition state theory (CVT) as well as with small curvature tunneling (SCT) contributions, via direct dynamics. For the major isomer, …

Assessment Of Oxidative Stress In The Livers Of Rats Exposed To 2-Aminoanthracene For Twelve Weeks, John C. Stagg
Honors College Theses
Diabetes is a chronic illness that affects many people, and it’s thought that environmental exposure to certain compounds may increase its proliferation. In this study, we examine the dietary effects of 2-aminoanthracene (2AA), a polycyclic aromatic hydrocarbon (PAH), in rats, specifically as it relates to oxidative stress. Oxidative stress refers to the imbalance of reactive oxygen species (ROS) and antioxidants in living systems; it has been shown to contribute to type-1 diabetes. To accomplish the study objectives rats were divided into three dietary treatment groups, control (0 mg/kg-2AA), low dose (50 mg/kg-2AA) and high dose (100 mg/kg-2AA). Rats were fed …

Best Organic Chemistry Practices In The Midwest, Claire Van Der Bosch
Honors Theses
Organic Chemistry is a gateway course to a handful of courses in higher chemical education. Unfortunately, it is notorious for its high-fail, high-withdrawal, and high-drop rate and has a reputation for being time-consuming, with difficult concepts that are oftentimes new to students. It is often taught in a two-semester sequence, with the courses named Organic I and Organic II, and both courses have a lab component. At Western Michigan University, the fail rate for these two courses are 53.5% and 36.9%, respectively. Most higher education institutions are certified with the American Chemical Society (ACS) and given a skeleton of a …

Analysis Of Dl-Amino Acid Ratios In Eggshells Using Reverse Phase-High Pressure Liquid Chromatography, Gergana Milkova
Honors Theses
HPLC methodology was developed to determine the concentrations and ratios of D to L amino acids in emu and ostrich eggshells treated with heat at different temperatures . We aimed to determine an internal standard and how ratios were affected under different conditions. An HPLC method was determined that gave consistent retention times and satisfactory enantioseparation . Calibration curves for each amino acid were developed from single and multi-amino acid containing dilution series producing a model that most closely replicates the eggshell-extracted amino acids.
Application Of The Picoloyl Group In Carbohydrate Chemistry, Michael Mannino
Application Of The Picoloyl Group In Carbohydrate Chemistry, Michael Mannino
Dissertations
Stereocontrol of glycosylation reactions is a constant struggle in the field of synthetic carbohydrate chemistry. The application of the picoloyl (Pico) substituent can offer numerous stereocontrolling avenues. The most popular application is the Hydrogen-bond-mediated Aglycone Delivery (HAD) method that provides excellent selectivity in the glycosylation of a variety of sugar substrates. The HAD method relies on the formation of an intermolecular hydrogen bond between the nitrogen atom of the Pico substituent on the glycosyl donor with the hydroxyl group of the glycosyl acceptor. This interaction provides a facial preference for the nucleophilic attack and hence provides powerful stereocontrol for the …

Enantioselective Copper Catalyzed Alkyne-Azide Cycloaddition By Dynamic Kinetic Resolution, En Chih Liu, Joseph J. Topczewski
Chemistry Faculty Research & Creative Works
The copper(I) catalyzed alkyne-azide cycloaddition (CuAAC), a click reaction, is one of the most powerful catalytic reactions developed during the last two decades. Conducting CuAAC enantioselectively would add a third dimension to this reaction and would enable the direct synthesis of α-chiral triazoles. Doing so is demanding because the two precursors have linear geometries, and the triazole product is a flat heterocycle. Designing a chiral catalyst is further complicated by the complex mechanism of CuAAC. We report an enantioselective CuAAC (E-CuAAC), enabled by dynamic kinetic resolution (DKR). The E-CuAAC is high yielding and affords up to 99:1 er. The E-CuAAC …
