The vitamin folic acid (folate, FA) has been extensively explored as a targeting ligand to deliver a variety of diagnostic/prognostic/therapeutic agents into various tumors through the assistance of its receptor – the folate receptor (FR). FR is over-expressed in many types of human cancer and can mediate internalization of FA-conjugates through an endocytic pathway. The discovery of small interfering RNA (siRNA), which is cable of inducing potent gene silencing in a sequence-specific manner, provides an excellent molecular tool to suppress aberrant gene expression in malignancies, and therefore siRNA has the potential to revolutionize cancer therapeutics. Semiconductor quantum dots (QDs) have recently been used to deliver and monitor biomolecules, such as drugs and proteins. However, QDs alone have a low efficiency of transport across the plasma membrane. In order to increase the efficiency, we used synthetic nona-arginine (SR9), a cell-penetrating peptide, to facilitate uptake. We found that SR9 increased the cellular uptake of QDs in a noncovalent binding manner between QDs and SR9. Further, we investigated mechanisms of QD/SR9 cellular internalization. Low temperature and metabolic inhibitors markedly inhibited the uptake of QD/SR9, indicating that internalization is an energy-dependent process. Results from both the pathway inhibitors and …

Vitamin B12 (B12)-based bioprobes were produced and investigated in vitro for studying the B12 uptake pathway in certain cancer cell lines and their target-specific capabilities as imaging and/or contrast agents in vivo. An intrinsic factor (IF)-bound B12 conjugate (B12-ReBQBA) was produced containing rhenium(I) for in vitro analysis of the cubilin-expressing placental choriocarcinoma cell line. B12-ReBQBA has millimolar toxicity in vitro and excitation and emission wavelengths in the visible region, 488 nm and 560 nm, respectively. The rhenium metal in this bioprobe can be readily exchanged for the metastable radioactive metal technetium-99m …

Librarians from Utah State University explain recent efforts to encourage subject librarians to take a more holistic view of their roles. We are shifting from a traditional emphasis primarily on collection development and refocusing on natural connections between collections, instruction, liaison, and reference service. Mutations in the human ether-a-go-go-related gene (hERG) cause long-QT syndrome type 2 (LQT2). We previously described a homozygous LQT2 nonsense mutation Q1070X in which the mutant mRNA is degraded by nonsense-mediated mRNA decay (NMD) leading to a severe clinical phenotype. The degradation of the Q1070X transcript precludes the expression of truncated but functional mutant channels. In the present study, we tested the hypothesis that inhibition of NMD can restore functional expression of LQT2 mutations that are targeted by NMD. We showed that inhibition of NMD by RNA interference-mediated knockdown of UPF1 increased Q1070X mutant channel protein expression and hERG current …

Mutational Studies Uncover Non-Native Structure In The Dimeric Kinetic Intermediate Of The H2a–H2b Heterodimer, Matthew R. Stump, Lisa M. Gloss

The folding pathway of the histone H2A–H2B heterodimer minimally includes an on-pathway, dimeric, burst-phase intermediate, I2. The partially folded H2A and H2B monomers populated at equilibrium were characterized as potential monomeric kinetic intermediates. Folding kinetics were compared for initiation from isolated, folded monomers and the heterodimer unfolded in 4 M urea. The observed rates were virtually identical above 0.4Murea, exhibiting a log-linear relationship on the final denaturant concentration. Below ∼0.4 M urea (concentrations inaccessible from the 4-M urea unfolded state), a rollover in the rates was observed; this suggests that a component of the I2 ensemble contains non-native structure that …

The method of electrochemical pretreatment of platinum electrode with the goal of standardizing the initial state of electrode surface and its open-circuit potential (OCP) in the blood and other biological media is proposed. The platinum electrode potential is measured in 0.14 M Na2SO4 aqueous solution, in the blood and blood plasma and serum. By the examples of OCP measured in the blood serum of patients with acute poisoning, acute cerebral pathology and patients treated by the method of hyperbaric oxygenation, it was found that the values of blood serum OCP were different for studied pathological states and healthy people.

Exposed along the southeast fl ank of the Colorado Front Range are rocks that beautifully illustrate the interplay of sedimentation and tectonics. Two major rangebounding faults, the Ute Pass fault and the Rampart Range fault, converge on the Garden of the Gods region west of Colorado Springs. Cambrian through Cretaceous strata upturned by these faults reveal in their grain compositions, textures, and bedforms radically different styles of sedimentation. The Cambrian/Ordovician marine transgressive deposits appear to have come to rest on a passive and tectonically inactive craton. In contrast, coarse-grained Pennsylvanian/Permian marine deposits of the Fountain Formation and Lyons Sandstone reveal …

This plan outlines strategies and methods used in an ongoing study initiated in 1999 to restore a selfsustaining population of the Plains gartersnake (Thamnophis radix) in Ohio. Restoring a self-sustaining population would require increases in the current population to where the ratios of T. radix to T. sirtalis are approximately 1:1 in multiple locations in Killdeer Plains Wildlife Area (KPWA). This ratio would be similar to what was seen earlier by Reichenbach and Dalrymple (1986) at one site in KPWA.

The human ether-a-go-go-related gene 1 (hERG1) encodes the pore-forming subunit of the rapidly activating delayed rectifier potassium channel. Several hERG1 isoforms with different N- and C-terminal ends have been identified. The hERG1a, hERG1b, and hERG1-3.1 isoforms contain the full-length C terminus, whereas the hERG1USOisoforms, hERG1aUSO and hERG1bUSO, lack most of the C-terminal domain and contain a unique C-terminal end. The mechanisms underlying the generation of hERG1USOisoforms are not understood. We show that hERG1 isoforms with different C-terminal ends are generated by alternative splicing and polyadenylation of hERG1 pre-mRNA. We identified an …

The Ecology Of The Peaks Of Otter Salamander (Plethodon Hubrichti) In Sympatry With The Eastern Red-Backed Salamander (Plethodon Cinereus), Norman Reichenbach, Andrew Kniowski

The Peaks of Otter Salamander, Plethodon hubrichti, is found along a 19 km length of the Blue Ridge Mountains, Virginia, USA, often in sympatry with the Eastern Red-backed Salamander, P. cinereus. In a sympatric area of Bedford County, Virginia, we conducted a mark-recapture study on a 10 × 10 m site. Surface densities of salamanders increased as the number of days without precipitation prior to a collection event increased. This suggests vertical movements in response to surface moisture. When salamanders returned to the surface after rain, individuals appeared to “shuffle” between rocks and likely, leaf litter. That is, we were …

Plant colonization can be limited by lack of seeds or by factors that reduce establishment. The role of seed limitation in community assembly is being increasingly recognized, but in early primary succession, establishment failure is still considered more important. We studied the factors limiting colonization on the foreland of Coleman Glacier, Washington, USA to determine the importance of seed and establishment limitation during primary succession. We also evaluated the effects of seed predation, drought and existing vegetation on establishment. Charting The Late Cretaceous Seas: Mosasaur Richness And Morphological Diversification, Marcus R. Ross

Abundant, readily identifiable, and biostratigraphically resolved specimens make mosasaurs ideal candidates to test fluxes in generic richness and morphological trends among marine vertebrates during the late Cretaceous. More than 1800 globally distributed mosasaur specimens are allocated to fifteen substage-level stratigraphically correlated assemblages. These data are quantitatively analyzed to illuminate trends within the group. Following their first appearance in the Cenomanian, mosasaurs experienced a significant radiation in the Coniacian and Santonian. Richness levels continued increasing into the Maastrichtian while mosasaurs incrementally exploited new predatory niches recognized via accumulation of novel dental morphologies. In spite of their complete lack of any structural features that characterize membrane proteins, cytosolic nucleoside-diphosphate kinases (NDPKs) have been found repeatedly to associate with membranes. In some instances the recruitment of cytosolic NDPKs to membranes was attributed to interactions with peripheral or integral membrane proteins, but in many cases the mechanism underlying the association of NDPKs with membranes remained unknown. We show here that cytosolic NDPKs bind directly to membrane lipids in a dynamic process that is controlled by its substrates, nucleoside tri- and diphosphates, and can be fully reconstituted with chemically defined, protein-free phospholipids and recombinant NDPK, or …

Detailed proteomic analyses of mammalian olfactory and rod photoreceptor sensory cilia are now available, providing an inventory of resident ciliary proteins and laying the foundation for future studies of developmental and spatio-temporal changes in the composition of sensory cilia. Cilia purification methods that were elaborated and perfected over several decades were essential for these advances. In contrast, the proteome of primary cilia is yet to be established, because purification procedures for this organelle have been developed only recently. In this chapter we review current techniques for the purification of olfactory and photoreceptor cilia, and evaluate methods designed for the selective …
