Identification Of Areas Of Weakness In Forensic Footwear Research, Kathryn Roy

Forensic footwear examinations have been carried out for almost a century, but the foundational validity of identifying a questioned impression to a particular shoe has been questioned by the National Research Council and the President’s Council of Advisors on Science and Technology. The focus of this paper was to identify the current weaknesses present in forensic footwear impression research. Additionally, a future experiment was proposed to address these concerns and improve the foundational validity of footwear impression comparisons. Protein tyrosine phosphatases (PTPs) regulate cell signaling by removing phosphate groups from tyrosine residues. Malfunction of these enzymes can lead to diseases such as diabetes and cancer. A conserved feature among PTPs is the catalytic acid loop, essential for phosphate removal. Computational analysis of PTP structures revealed that ancestral PTPs have considerable variation in conformations of this loop, sparking interest in their further study. The Asgard group, identified as ancestral to eukaryotic PTPs, was successfully expressed and purified under optimized conditions. Current efforts focus on optimizing crystallization to obtain larger crystals for structural analysis. This project will investigate the allosteric …

Highly Regenerable Magnetic Sulfonated Chitosan Crosslinked With Glutaraldehyde Composite Beads (Iron Sand/Naso3-Chi-G) For Aqueous Mercury Removal, Fathurrahmi Fathurrahmi, Rahmi Rahmi, Lelifajri Lelifajri, Anggun Sixthia Wulan Ayu, Muhammad Iqhrammullah

Karbala International Journal of Modern Science

We developed a novel adsorbent from sulfonated, glutaraldehyde-crosslinked chitosan embedded with magnetic iron sand. The adsorbent was synthesized through a two-step process: (1) sulfonation with N(SO₃Na)₃ to introduce sulfonate groups, and (2) crosslinking with glutaraldehyde to enhance structural stability. The optimal formulation, containing 43.5% iron sand and crosslinked with 0.17 M glutaraldehyde, exhibited the highest Hg²⁺ adsorption capacity (30.74 mg/g) at pH 3. The adsorbents were characterized using Scanning Electron Microscopy (SEM), Fourier transform infra-red spectroscopic (FT-IR), and X-ray diffraction (XRD) techniques. Adsorption equilibrium was achieved within 60 minutes, and isotherm modeling showed that the process followed the Freundlich model …

Protein-protein interactions (PPI) are fundamental to cellular function, influencing processes such as signaling, structural organization, and disease progression. Disruptions in PPIs are implicated in cancer and neurodegenerative diseases, highlighting their importance as therapeutic targets. However, traditional methods for studying human PPIs, such as yeast two-hybrid and co-immunoprecipitation mass spectrometry, are costly and technically demanding. This dissertation explores the LexA-based bacterial two-hybrid (LexA-B2H) system as a cost effective and accessible alternative for investigating human PPIs.

The production of RNA is vital for many commercial and research purposes, with one of the more notable reasons being vaccine production. The RNA polymerase, derived from T7 bacteriophage, is commonly used for RNA production due to its ability to produce full-length RNA transcripts. However, researchers have faced limitations in optimizing this polymerase. For example, the polymerase generates heterogeneous RNA products that require additional purification steps during RNA production. Here, we selected three amino acid point substitutions based on a recent publication from Moderna that showed increased RNA purity compared to the wild type T7 RNA polymerase sequence. We created …

Tuberculosis (TB) remains a significant public health challenge worldwide. Currently, Bacillus Calmette-Guerin (BCG) is the only vaccine available for TB prophylaxis. However, the efficacy of the BCG vaccine against adult pulmonary TB is considered inconsistent. This condition encourages researchers to look for more effective options, such as subunit vaccines. This condition requires the development of a more effective subunit vaccine to protect active TB in productive and adult ages. The ethylene-forming enzyme (EFE) from the fungus Penicillium digitatum strain Pd1 was heterologously produced in Escherichia coli and its properties were compared to the extensively characterized bacterial enzyme from Pseudomonas savastanoi strain PK2. Both enzymes catalyze four reactions: the conversion of 2-oxoglutarate (2OG) to ethylene and CO2, oxidative decarboxylation of 2OG coupled to L-arginine (L-Arg) hydroxylation, uncoupled oxidative decarboxylation of 2OG, and the production of 3-hydroxypropionate (3-HP) from 2OG. Homeodomain transcription factors (TFs) bind to specific DNA sequences to regulate the expression of target genes. Structural work has provided insight into molecular identities and aided in unraveling structural features of these TFs. However, the detailed affinity and specificity by which these TFs bind to DNA sequences is still largely unknown. Silver nanoparticles (AgNPs) were synthesized via an innovative green synthesis method using amygdalin (Am) as a reducing agent and the Nd: YAG laser as a catalyst. We studied the properties of the nanoparticles using X-ray diffraction (XRD), field emission scanning electron microscopy (FESEM), energy dispersive X-ray spectroscopy (EDX), atomic force microscopy (AFM), ultraviolet-visible spectroscopy (UV), and Fourier transform infrared spectroscopy (FTIR) techniques. 1,8-Cineole is a naturally occurring chemical molecule predominantly found in fragrant plants, particularly Myrtle. Its aroma is distinctive and has been the subject of numerous investigations due to its various biological actions. This study examines the characterization of 1,8-Cineole derived from Myrtle and investigates its antibacterial and antioxidant properties. This study seeks to compare 1,8-Cineole with antibiotics like Amoxicillin and Tetracycline, and moreover, to investigate its antioxidant capabilities against diverse bacterial strains (both Gram-positive and Gram-negative). 1,8-Cineole exhibits the most effective antibacterial properties, demonstrating an inhibition zone of 12.00 mm against Staphylococcus aureus and 10.00 mm against Pseudomonas aeruginosa. …

Nuclear receptor binding set domain protein 2 (NSD2) plays a key role in chromatin regulation and is associated with different cancers and other developmental problems. Berries are rich in major secondary metabolites with anticancer properties. Here, we virtually screened 145 berry phytochemicals as putative NSD2 inhibitors via structure-based virtual screening, molecular docking, MD simulations, and DFT and ADMET analyses. Among them, α-carotene had the maximum docking score of -9.9 kcal/mol, followed by sulfuretin and β-amyrin. Obesity is caused by an energy imbalance that increases chronic low-grade inflammation, including macrophage cell infiltration. Phenolic compounds from olive mill wastewater (PCO) of Algerian origin were used to produce sodium alginate-based active films using the casting method. The effects of adding various concentrations of PCO (0%, 0.1%, and 0.2% w/v) were evaluated regarding the molecular, morphological, thermal, physicochemical, optical, barrier, biodegradability, antimicrobial, and antioxidant properties of the alginate films. The FTIR and SEM results elucidated the development of a coherent cross-linked structure attributed to hydrogen bonding interactions between PCO and alginate chains. Cold atmospheric plasma (CAP) was evaluated for degrading organophosphate pesticide (OP) residues (acephate, malathion, and dimethoate) from Kevlar fabrics. Fourier transform-Infrared (FTIR) spectroscopy, scanning electron microscope (SEM) imaging and tensile strength testing confirmed that CAP treatment preserved Kevlar's structural integrity and mechanical properties. Results show degradation efficiency increased with higher power, shorter discharge gaps, and longer exposure duration. Hyperspectral imaging supported the detection of plasma-induced spectral changes, reinforcing the presence of reactive species. The strong oxidative potential of CAP facilitated the rapid breakdown of OPs into nontoxic byproducts. These findings demonstrate the potential of CAP as a portable, energy-efficient solution …

This study employed high-time-resolution systems to examine the transient properties of aerosols and gases emitted from electronic cigarette (EC) puffs. Using a fast aerosol sizer, we measured particle size distributions (PSDs) across various EC brands (JUUL, VUSE, VOOPOO), revealing sizes ranging from 5 to 1000 nm at concentrations of 107 to 1010 cm-3. Most aerosols were found to be in the ultrafine range (below 100 nm), with JUUL-, VUSE-, and VOOPOO-producing aerosols with geometric mean sizes of 19.9, 47.3, and 29.4 nm, respectively. Bone tissue engineering (BTE) aims to repair bone defects using biocompatible materials with tailored geometries and pore structures, providing appropriate mechanical support and control over biodegradation kinetics to promote bone growth. In this study, we utilized digital light processing (DLP) 3D printing to fabricate scaffolds with varying pore sizes using polymer–ceramic slurries composed of polylactic acid (PLA) as the main polymer matrix, incorporated with hydroxyapatite (HA) and bioactive borate glass (BBG) at various ratios. Light is critical for the survival of most shallow water scleractinian corals. Corals are mixotrophs, acquiring energy via both feeding on water column plankton and photosynthesis via a symbiotic relationship with dinoflagellates living within their tissues. Corals rely heavily on photosynthetically derived metabolites through this symbiotic relationship, intrinsically linking their biology to light availability. This research aims to develop formative assessment media assisted by conceptual scaffolding to improve students’ understanding of temperature and heat, test its validity and practicality, and gauge student responses. Utilizing the ADDIE model, the research follows five stages: Analysis, Design, Development, Implementation, and Evaluation. In the Analysis stage, a literature review was conducted to identify critical components, including formative assessment, conceptual scaffolding, concept mastery, and previous research. The Design stage involved identifying users, defining competencies and indicators, creating a storyboard, integrating assessment components, and determining mastery levels. The study investigates the performance of students in constructing Concept Maps (CM) related to environmental pollution material during the COVID-19 pandemic. The data reveals that the overall achievement in CM construction was suboptimal, with no component reaching more than half of its maximum potential. The highest achievement was recorded in the valid relationship component, which attained 9.75% of its maximum possible score. This was followed by the hierarchy component, achieving 7.34%, and the branching component at 3.42%. The aim of this study is to identify students’ problem-solving skills in Newton’s laws through the implementation of the 5E learning cycle in the classroom. A pre-experimental design with a one-group pretest-posttest approach was employed. Data on problem-solving skills were collected from students’ pre-test and post-test responses to two essay questions. The indicator with the highest percentage of problem-solving skills was useful description (83% of students), while the indicator with the lowest percentage was specific application of physics (33% of students).
