It has long been known that a hot environment negatively influences milk production in cattle. Although molecular mechanisms underlying this phenomenon to a great extent remain to be recognized, they include alterations in lipid metabolism. DGAT1 plays a key role in the synthesis of triacylglycerols and polymorphism of this gene has been proven to have a pronounced influence on milk production. The aim of this study was to establish if the effect of DGAT1:p.K232A on milk-related traits is dependent on environmental thermal conditions. Previously, we reported the effects of fescue toxicosis on developing Angus-cross steer growth, carcass, hepatic mRNA, and protein expression profiles of selected serum proteins, and blood clinical and chemical profiles, after summer-long grazing (85 days) of high endophyte (HE)- vs. low endophyte (LE)-infected fescue pastures. We now report the temporal development of acute, intermediate, and chronic responses of biochemical and clinical blood analytes determined at specified time intervals (period 1, day 0–36; period 2, day 37–58; and period 3, day 59–85). Shiga toxin-producing Escherichia coli (STEC) are enteric pathogens of humans. Cattle serve as a reservoir and harbor STEC in their intestines. Intimin-positive STEC are referred to as enterohemorrhagic E.coli (EHEC). Seven serogroups (O26, O45, O103, O111, O121, O145, and O157) account for the majority of illness due to STEC and are hereafter referred to as STEC/EHEC-7. To improve detection, enrichment broths were compared for supporting growth of STEC-7 and STEC O104:H4 (STEC-8). In pure culture, STEC enriched in trypticase soy broth (TSB) had significantly greater growth compared to TSB containing antimicrobials. In fecal samples, E. coli broth enrichment yielded …

This study examined the effects of increasing energy availability from both dextrose and lipid treatments on the proinflammatory response to LPS in Holstein steers. Steers were randomly assigned to one of three groups: saline at 0.5 ml/ kg body weight (Control) or 50% dextrose [0.5 ml/kg body weight (Dextrose) to mimic calm cattle’s response to LPS] administered immediately prior to LPS (0.5 mg/kg body weight at 0 h) or continuous lipid emulsion infusion from –1 to 6 h [Intralipid 20% (Baxter, Deerfield, IL USA); 0.5 ml/kg/hr (Lipid) to mimic temperamental cattle]. The hypothalamic gonadotropin-releasing hormone receptor (GnRHR) plays an essential physiological role in reproductive function. In this investigation, we studied genetic variation in the entire coding region of GnRHR gene using PCR-SSCP technique in 250 Bos taurus (Holstein Friesian, Jersey) and Bos indicus (Malnad Gidda, Deoni) bulls. The genetic variants in GnRHR gene were determined by PCR-SSCP technique by using 5 sets of primers. In this study, the presence of paratuberculosis was investigated by serological and molecular methods in herds of dairy cattle. Blood, milk, and stool samples of 147 cows aged 2 years old or older with chronic diarrhea were collected. A California mastitis test (CMT) was performed on milk samples. Indirect paratuberculosis enzyme-linked immunosorbent assay (ELISA) test was used for serological investigation. Polymerase chain reaction (PCR) was utilized for molecular identification of Mycobacterium avium subsp. paratuberculosis (MAP) from milk and stool samples. Swamp buffaloes have been important to Thai countryfolk for centuries. However, it’s meat consumption has increased continuously. Since meat and offal of cattle are very similar, pancreas might be used to differentiate buffalo offal from cow offal. Pancreases from 10 adult buffaloes of both sexes were obtained and studied anatomically. Studies revealed that, unlike human and other animals, buffalo pancreas had a unique F-shape with left lobe, right lobe, body, and uncinate process. There was no main pancreatic duct. Leptospira is an emerging global public health problem which involves both animals and human. The aim of this study was to appraise the presence of Leptospira DNA in blood samples from cattle and sheep via Polymerase Chain Reaction (PCR) method in Chaharmahal VaBakhtiari and Isfahan provinces of Iran. The aim of the present study was to compare the effectiveness of cefuroxime, herd-specific autovaccine, and cefuroxime/herd-specific autovaccine treatments in eliminating S. aureus from the milk of 45 cows with subclinical mastitis. Prior to this study, cows with S. aureus mastitis were divided into 3 groups of 15 cows each. Group 1 received cefuroxime, group 2 received the autovaccine, and group 3 received cefuroxime and the autovaccine. This study was conducted to determine the relationship between the degrees of anemia and blood gases in cattle with theileriosis. It included 28 cattle with theileriosis and 7 healthy animals as the control group. The cattle with theileriosis were divided into 4 groups according to their hematocrit (Hct) values: cattle with Hct of >26 were considered as nonanemic (Group 1), Hct of 20-26 as mildly anemic (Group 2), Hct of 14-19 as moderately anemic (Group 3), and Hct of 10-13 as severely anemic (Group 4). In the present study, an 84-bp insertion/deletion polymorphism of the sterol regulatory element-binding protein 1 (SREBP1) gene was investigated. The analysis included 6 Polish cattle herds consisting of beef (Limousin, Angus, Charolais), dairy (Holstein-Friesian, Jersey) and dual-purpose (Montbeliard) breeds. The polymorphism was found only in Limousin and Montbeliard animals, in which both the short (S)- and long (L)-type alleles occurred. The genotype distribution in Montbeliard cattle was as follows: 0.038, 0.406, and 0.556 for SS, LS, and LL, respectively. The aim of this study was to determine the possibility of yielding more transferable embryos from cows superovulated with/without exogenous progesterone (ear implant and intravaginal device). Two experiments were conducted in Holstein Friesian cows to evaluate the effect of exogenous progesterone on the yield of corpus luteum (CL), total embryo and ova, and transferable embryos before superovulation. A superovulation program using 2 different progesterone resources was applied (G1 = intravaginal device, G2 = ear implant). Both cattle and dogs were examined in modern and rural dairy farms that had a history of abortion over 5%. The blood samples were collected from 427 aborted cattle and the sera were tested using a commercial ELISA test kit. Additionally, a necropsy procedure was carried out on the fetuses and calves dead within 2 months after birth; the tissue samples were evaluated by histopathologic, immunoperoxidase, and PCR techniques. Recent progress in genomic analysis and other biochemical methods has led to the discovery of a large population of microRNAs (miRNAs), which have been demonstrated to play important roles in diseases and a wide range of developmental processes. Characterization of miRNA expression profiles in different stages of ovarian follicular development and early embryogenesis has suggested the potential roles of miRNAs in follicular development, maturation of oocytes, and preimplantation embryonic development. This review focuses on the current studies of miRNAs involved in ovarian and early embryonic development in cattle.

In this study, we compared 2 methods, liquid-phase blocking ELISA (LPBE) and solid-phase competition ELISA (SPCE), for the detection of antibodies to the structural proteins of foot-and-mouth disease virus (FMDV). These methods were compared using sera collected from cattle (n = 30) without a history of foot-and-mouth disease infection or vaccination, cattle (n = 180) vaccinated with oil-adjuvanted bivalent vaccine, and international reference sera (positive, weak positive, and negative) for FMDV serotypes O and A. Endometritis In Simmental Cows: Incidence, Causes, And Therapy Options, Robert Zobel

The objectives of this study were to evaluate endometritis incidence and possible causes, and to assess the different therapy options. From a total of 1300 cows, endometritis was diagnosed in 23.07%, with clinical endometritis in 15.31% and subclinical endometritis in 7.77%. Previous dystocia resulted in 40.59% of the clinical endometritis and 47.52% of the subclinical endometritis (P < 0.05). Placental retention caused clinical and subclinical endometritis in 43.18% and 37.50% of the cows, respectively (P < 0.05). Body condition score 2 resulted in the highest endometritis rate and body condition score 3-4 resulted in the lowest endometritis rate (P < 0.05). Corpus luteum was found in 9.67% of the cows suffering from endometritis, while the rest developed various types of subfertility (P < 0.05). Three groups of cows (A, B, and C) with an almost equal number of clinical and subclinical endometritis diagnoses were formed. All of the groups received prostaglandin analogues (PGF Veyx Forte®, Veyx Pharma) intramuscularly 3 times at 11 day intervals. In group A, only prostaglandins were used; in group B, cephapirin (Metricure®, Intervet Schering-Plough) was administered into the corpus uteri simultaneously with the first 2 doses of prostaglandins; and in group C, the first 2 doses of prostaglandins were followed by the ozone product Riger Spray (Novagen, Italy) applied into the uterus. The criteria applied for the therapy success were the number of cows conceived after the first artificial insemination and the number of days open. The best therapy results were achieved in groups C and B (P < 0.05), with the advantage of no milk and meat withdrawing period in the animals of group C.

The purpose of this study was to establish the fertility trends in dairy cows in Central Croatia and compare the trends and causes of infertility and subfertility during a 5-year period. A total of 12,060 Simmental cows and heifers were included in the study. The animals were examined by vaginoscopy, transrectal palpation, and ultrasonography on a regular basis. Fertility problems that included infertility and subfertility were diagnosed in 56.61% of all observed animals. The total number of animals diagnosed with subfertility increased and almost doubled by the end of the study period. Subfertility issues associated with anovulatory oestrus, delayed ovulation, …

This study was performed via a survey conducted of a total of 112 cattle operations in Kars and the vicinity and included a total of 2317 head, of which 280 were determined to have foot problems. The financial loss incurred per operation was calculated in accordance with the data obtained. As a result, the prevalence of foot disease encountered in cattle in the region in 2010 was determined to be 12.08%, and although it varied depending on the severity of the foot diseases, the animals were healed in an average of 46.5 days. The aim of this study was to determine the normal conjunctival bacterial flora and the effects of sex and age variations on them in healthy cattle in Urmia, Iran. The animals (n = 50) were selected from 2 sexes and divided into 2 age groups. Swabs were taken from the inferior conjunctival sac of both eyes (n = 100) and were cultured on blood and MacConkey agar. All (100%) of the specimens yielded positive bacterial isolations. The bacterial isolates (in order of decreasing frequency) were Lactobacillus plantarum, Escherichia coli, Bacillus cereus, Proteus mirabilis, Enterobacter aerogenes, Staphylococcus epidermidis, and S. aureus. …

Enzootic bovine leukosis (EBL) is caused by bovine leukemia virus (BLV) infection. BLV was detected incattle using nested polymerase chain reaction (PCR) assay and were identified BLV infected cattle farms in fiveselected provinces in the Philippines. A total of 300 cattle blood samples were used. BLV Proviral DNA wasextracted and amplified using nested PCR assay targeting the BLV long terminal repeat (LTR). Results showed that11 samples (3.67%) of the 300 cattle blood samples used were positive for BLV infection. This study is considered firstreport of cattle EBL in the Philippines.

The seroprevalence of Leptospira interrogans serovars in cattle was determined by analyzing 385 serum samples from 8 Rural Development Districts (RDD), in Tamaulipas, Mexico. A microscopic agglutination test (MAT) including 9 serovars of L. interrogans as antigens was used. Serum samples were considered positive when 50% or more of agglutination in a dilution of ≥ l:100 was observed. A total of 70.4% animals were reported positive for one or more serovars. The most frequent antibodies reported were those against serovars tarassovi (53.25%) hardjo (23.64%) and canicola (15.32%), whilst serovar bataviae (0.52%) was found at lower frequency. To determine the seroprevalence of Anaplasma in Minnesota cattle, the database of the Veterinary Diagnostic Laboratory (VDL), University of Minnesota was searched over a 10 year period (2001 to July 2010). A total of 438,407 bovine serum samples were tested by complement fixation test (CFT) and competitive enzyme linked immunosorbent test (cELISA). The positive rate of seroprevalence by CFT was 2.4% out of 211,484 samples, with a 0.13% anticomplementary result. The VDL at the University of Minnesota started using cELISA after 2002, and CFT application was suspended as diagnostic test for anaplasmosis after 2005 due to its complexity. With cELISA, …

This study aimed to investigate the histopathological changes that occurred in the wound of the sole ulcer located on the hooves of dairy cattle. A total of 12 Holstein-Frisian dairy cows suffering from sole ulcer were studied. Sole ulcers in the lateral hoof of the left or right foot of hind legs containing the corium and a part of the epidermis were collected and examined histopathologically. There were subacute inflammatory changes in the superficial dermal vessels. A new type of bovine pestivirus, atypical pestivirus, was identified in natural infected calffirstly in Khon Kaen, Thailand in 2004. Mycoplasma wenyonii (M. Wenyonii) was studied by PCR and blood smear staining in 197 cows. Clinical signsand hematological parameters of all tested cows were collected. Results confirmed that 31% populations werepositive of M. wenyonii infection. Statistical analysis revealed that the cows of 2-4 years old were most susceptible. Noclinical sings were found to be significantly associated with the infection. The infection also did not result in anemiabased on hematological investigation that only showed elevation of WBC counts in infected cows. Sequence analysisof the partial 16S rRNA gene showed that three different representative strains were confirmed in the present study.

A total of 19,679 adult ticks were collected from 443 cattle in 9 villages in the West Aegean region of Turkey over 2 years (June 2006-May 2008). Thirteen out of the 19,679 adult ticks were identified as Hyalomma rufipes (12 adult males and 1 female). The ticks were identified according to their characteristic dark coloured conscutum and scutum along with the presence of small to medium but uniform and numerous punctations. The aim of this study was to establish the prevalence of repeat breeder syndrome (RBS) in different breeds and reproductive ages of cattle in central Croatia and to assess the effectiveness of hormone therapy as a treatment option. A total of 1088 dairy cows were observed, 218 heifers and 870 multiparous cows. After the third unsuccessful artificial insemination (n = 164) and a thorough clinical and gynecological examination, the cows were tested for sexually transmitted diseases. An RBS diagnosis was based on failure to conceive, negative laboratory tests for sexually transmitted diseases, and the absence of other reproductive disorders. The …
