Control of Bovine Viral Diarrhea Virus (BVDV) relies on resource-intensive individual animal sampling to detect and remove persistently infected (PI) cattle. Herd-level surveillance tools would be useful for herds with unknown BVDV status and for monitoring herds with BVDV-free status. The overall objective of this thesis is to explore the viability of BVDV surveillance at a herd-level using samples collected without handling individual animals. The first objective was to determine the feasibility of using stable flies as a sampling tool to detect BVDV. A form of congestive heart failure is increasingly reported as a cause of death in feedlot cattle located at moderate altitude (≤ 1,524 m.). Significant knowledge gaps exist in the epidemiology and management of this form of bovine congestive heart failure (BCHF) regarding frequency and timing of BCHF cases and ways in which beef cattle producers and veterinarians can mitigate this condition. The aim of this study was to evaluate the serological diagnostic potential of the Brucella recombinant outer membrane (rOMP25, rOMP31) and periplasmic proteins (rBP26, rSOD) in a comparative way using an indirect enzyme-linked immunosorbent assay (i-ELISA). Rabbit and/or mouse antibodies to Brucella whole cell and/or soluble protein preparations recognized all recombinant proteins used, which confirms the expression of target antigens in E. coli in active form. The recombinant proteins showed different antigenicity to antibodies of cattle kept on a brucellosis-affected (endemic) farm and/or a new focus of infection. Anaplasmosis has become a major concern in the cattle industry throughout the world due to its great economic impact. In this case report, specific antivenin treatment for Viperidae snake envenomation in two cattle was defined for the first time. The diagnosis of snake envenomation was made according to the history and clinical findings. Clinical examination revealed systemic (tachycardia, dyspnea, cyanosis, hypersalivation, ruminal hypotony) and local (swelling of the leg and head) findings depending on the location of snake bites. Laboratory analysis determined granulocytic leukocytosis, monocytosis, decreased iron binding capacity, and increased CK and LDH enzyme activities. In the specific treatment of cases, equine-derived polyvalent antivenin containing antitoxic immunoglobulins against Viperidae species was administered. A total of 20 domestic cattle (Bos taurus L.) and 15 water buffalo (Bubalis bubalis L.) skulls were analyzed in this study. All of the specimens belonged to female individuals. Using a total of 27 craniometric measurements from each of the skulls, 9 indices were calculated. Although there were statistically significant differences between the linear measurements of the skulls of both species, while calculating the indices with their ratios, the values of the measurements of the facial area were determinative in the craniology. Among these indices, the facial index 1 value was statistically significant (P < 0.01) in the comparison of these two species. On the other hand, while considering the orbit and foramen magnum measurements, in contrast with the transversal measurement, the height was more determinant for their index and form.

A total of 20 microsatellite DNA markers were used for genetic characterization and determination of phylogenetic relationships of native cattle breeds of Turkey, including the Anatolian Grey (AG), Anatolian Black (AB), South Anatolian Red (SAR), East Anatolian Red (EAR), Southern Anatolian Yellow (SAY), and Zavot (ZAV). DNA samples were isolated from 271 blood samples using an organic method. Amplified polymerase chain reaction products were separated by capillary electrophoresis and genotypes were determined for 20 microsatellites. A total of 269 different alleles were determined. The present study was conducted to optimize oral doses of fat- and water-soluble vitamins in cattle using in vitro gas production model and to ascertain their effects on dry matter intake (DMI), nutrient digestibility, and rumen microbial protein (RMBP) synthesis. Doses optimized using the in vitro model were 3.66, 6.78, 16.3, 41.1, 3.87, 0.323, 4.12, and 0.055 mg/kg DMI, respectively, for thiamine, riboflavin, niacin, pantothenic acid, pyridoxine, biotin, folic acid, and B12 and 500 IU/kg DMI for vitamin K. This study was designed to evaluate serum and electrocardiographic (ECG) changes in cattle with foot-and-mouth disease (FMD). Thirty-two cows with clinical signs of FMD and 13 healthy cows were randomly enrolled. After blood sampling, ECG was performed on 23 of the 32 ill cows. Serum circulatory troponin I (cTnI), CPK, LDH, and CK-MB activities were determined in the patient and control groups. Arrhythmias including ventricular tachycardia (VT), sinus tachycardia (ST), atrial fibrillation (AF), and premature ventricular complex (PVC) were seen in 4, 4, 2, and 1 of the patients, respectively. The purpose of this study was to examine resistance against cadmium (Cd$^{+2}$), copper (Cu$^{+2}$), lead (Pb$^{+2}$), mercury (Hg$^{+2}$), and manganese (Mn$^{+2}$) in Escherichia coli isolates recovered from cattle stool using phenotypic (agar dilution) and genotypic [polymerase chain reaction (PCR)] methods. In addition, the isolates were genotyped via enterobacterial repetitive intergenic consensus polymerase chain reaction (ERIC-PCR). Bovine mammary gland biopsies allow researchers to collect tissue samples to study cell biology including gene expression, histological analysis, signaling pathways, and protein translation. This article describes two techniques for biopsy of the bovine mammary gland (MG). Three healthy Holstein dairy cows were the subjects. Before biopsies, cows were milked and subsequently restrained in a cattle chute. The aim of this study was to isolate the antimicrobial-resistant enterobacteria from cattle feces and to identify the extended-spectrum beta-lactamase (ESBL) and carbapenemase gene among them. A total of 70 cattle fecal swab samples were collected from Durg, Chhattisgarh, India, and cultured in nutrient broth. Bacterial isolation recovered 120 gram-negative bacteria. Antimicrobial susceptibility testing showed 90 (75%) coliform isolates (66 Escherichia coli, 20 Klebsiella sp., and two each of Enterobacter sp. and Citrobacter sp.) resistant to amoxicillin, amoxicillin-clavulanic acid, cefixime, ceftazidime, imipenem, or meropenem. The present case report deals with the morphological (gross and histopathological) and histochemical characterization of Johne?s disease in a cow. The carcass was presented in a hidebound condition with sunken eyes and with a history of persistent and nonresponsive diarrhea. Necropsy revealed generalized gelatinization of the visceral and peripheral fat. Thorough gross examination unveiled major pathological changes in the digestive tract. Effects of enzymatically hydrolyzed yeast (EHY) and supplementation frequency (FREQ) on immune parameters among beef cows and calves was evaluated. Eighty multiparous (parity = 4.2 ± 0.3) cows were fed a common brome hay-based diet (CP = 8.0 ± 0.17%). Cows were blocked by expected calving date and stratified by body condition score (BCS) before random assignment of treatment. Beginning 88 ± 5 d prior to parturition, cows were provided 1 kg daily or 3 kg every 3 d of a soybean hull-based supplement (CP = 34.0, % of DM) that contained 0 or 3 g/kg EHY. The main aim of this study was to identify the association between the single nucleotide polymorphism (SNP) in the bovine interleukin-17A (IL-17A) gene and mastitis resistance. Additionally, the connection between the identified polymorphism and the somatic cell score (SCS) was tested. Two hundred and six Polish Holstein?Friesian cows were genotyped using the PCR-RFLP method. One SNP mutation (c.126G>A) was identified within the coding sequence of the analyzed gene. Results of logistic regression analysis displayed no significant association between the IL-17A genotype and clinical mastitis occurrence (Wald chi-square test = 0.74; P = 0.69). The study aimed to investigate the presence of coccidial oocysts and pathological changes in a dairy herd with the history of coccidial outbreak in Faisalabad District. Fecal samples were collected from 541 animals, including cattle (379) and buffaloes (162), and examined for protozoal infection using flotation techniques. Results revealed 2.9% presence of coccidial oocysts in the samples collected. No significant difference was found in coccidial oocysts count in species of the cattle (2.3%) and buffaloes (4.3%), sex and age group. The aim of this study was to look at four ultrasonographic approaches that diagnose acquired reticular diaphragmatic hernia (RDH) using either left or right caudal thoracic approaches (4th/5th intercostal space) and compare them to those made more cranially (1st/2nd intercostal space). Each sonographic observation was made in 27 unsedated, standing animals (22 buffaloes and 5 cattle) that were surgically confirmed for RDH. The purpose of the study was to use decision trees to predict increased levels of somatic cells in cow's milk. The material for the study comprised data collected in 2012-2014 from five farms in Poland equipped with an automatic milking system. Data on 803 Polish Holstein-Friesian cows were collected. In order to predict somatic cell count data mining techniques were used to build a graphical model of a decision tree. Bovine skin and teat papillomas were examined for immunohistochemical expression of bovine papillomavirus (BPV), proliferating antigens (PCNA, Ki-67), CD3+ T lymphocytes, and some of the type III intermediate filaments. Papillomas showed epidermal hyperplasia, koilocytes, and hyperkeratosis with prominent dermal connective tissue. Immunohistochemistry found the presence of BPV antigens in the nuclei of granulosa cells. PCNA and Ki-67 revealed positive nuclear staining in the dermis and epidermis. Perivascular cuffing in the dermis predominantly consisted of CD3+ T cells. Bovine parainfluenza virus type 3 (BPIV3) is a leading respiratory pathogen of cattle and is often implicated as a causative pathogen of bovine respiratory disease complex (BRDC). Three genotypes (BPIV3a-c) have been tentatively classified based on genetic and phylogenetic analyses. A BPIV3c isolate, designated NX49, was isolated using the Madin-Darby bovine kidney cell culture from cattle in western China, where feedlot cattle with severe respiratory disease symptoms were observed. Hemagglutination assay demonstrated that NX49 had low agglutination activity against guinea pig erythrocytes, whereas SD0835, the first BPIV3c strain reported in China, showed high agglutination activity. Ticks are the main hematophagous arthropods and obligatory ectoparasites that are considered a vector of serious pathogens for animals and humans. The aims of the present survey were to assess the prevalence and diversity of ticks in cattle in Southeast Iran and to determine Coxiella burnetii in the ticks. From May 2014 to April 2015, 583 ticks were collected randomly from 257 cattle. Nested trans-PCR was performed to detect C. burnetii. The overall prevalence of hard tick infestation on cattle was 56.8%. In vivo derived (IVD) and in vitro produced (IVP) embryo transfers are respectively the 2nd and 3rd generations of the reproductive biotechnologies. Commercial application of these technologies began in the seventies for the former and in the eighties for the latter. After 40 years, they have clearly proved their sustainability to the livestock industry. The present article deals with bovine embryos only. The numbers of embryos collected or produced and transferred worldwide are reported in the first part. About one million bovine embryos are transferred each year. Trophoblast cells from bovine somatic cell nuclear transfer (SCNT) conceptuses express major histocompatibility complex class I (MHC-I) proteins early in gestation, and this may be one cause of the significant first-trimester embryonic mortality observed in these pregnancies. MHC-I homozygous-compatible (n = 9), homozygous-incompatible (n = 8), and heterozygous-incompatible (n = 5) SCNT pregnancies were established. The control group consisted of eight pregnancies produced by artificial insemination. Uterine and placental samples were collected on Day 35 ± 1 of pregnancy, and expression of MHC-I, leukocyte markers, and cytokines were examined by immunohistochemistry. Microcystins are acute hepatotoxins of increasing global concern in drinking and recreational waters and are a major health risk to humans and animals. Produced by cyanobacteria, microcystins inhibit serine/threonine protein phosphatase 1 (PP1). A cost-effective PP1 assay using p-nitrophenyl phosphate was developed to quickly assess water and rumen content samples. Significant inhibition was determined via a linear model, which compared increasing volumes of sample to the log-transformed ratio of the exposed rate over the control rate of PP1 activity. Although their action in antagonizing arachidonic acid metabolism is a key antiinflammatory effect of conjugated linoleic acid (CLA) and omega-3 polyunsaturated fatty acids (n-3 PUFA), these molecules have other antiinflammatory effects that might occur upstream of altered eicosanoid production. We examined the effects of two CLA isomers (c9,t11 and t10,c12 isomers), n-3 PUFA (EPA and DHA), and ΚNF-ΚB inhibitor (ammonium pyrrolidine dithiocarbamate (PDTC)) on lipopolysaccharide (LPS)-induced secretion of prostaglandins (PGE2 and PGF2a) by bovine endometrial (BEND) cells. Mycoplasmas cause the most severe and economically costly diseases of cattle throughout the world. In this study, Mycoplasma species were isolated from calves and cows with suspected mycoplasmosis in Holstein dairy cattle farms within 7 geographical regions of Turkey between May 2010 and December 2015. Mycoplasma infections were positive in 17 (80.9%) of 21 dairy cattle farms and the overall percentage was calculated as 32.1%. The highest isolation rate occurred in the Southeastern Anatolia Region (42.8%), and the lowest was observed in the Mediterranean Region (19.6%). In the current study, 23 papilloma/tumor-like samples obtained from cattle having clinical lesions and 9 blood samples collected from healthy-appearing cattle in Turkey were examined for bovine papillomavirus (BPV) DNA using the degenerate primers FAP59/64, and the different types of BPV were distinguished by type-specific primer sets. Furthermore, histopathological studies of papillomavirus were performed. A total of 7 BPV types (BPVs 1, 2, 3, 4, 6, 8, 9), including genera Deltapapillomavirus, Xipapillomavirus, and Epsilonpapillomavirus, were identified. This study aimed to find whether there is any difference in blood parameters of cattle that are naturally infected with BVDV, BHV-1, and BHV-4 and Brucella-seronegative.
