Livestock face complex foraging options associated with optimizing nutrient intake while being able to avoid areas posing risk of parasites or disease. Areas of tall nutrient-rich swards around fecal deposits may be attractive for grazing, but might incur fitness costs from parasites. We use the example of dairy cattle and the risks of tuberculosis transmission posed to them by pastures contaminated with badger excreta to examine this trade-off. A risk may be posed either by aerosolized inhalation through investigation or by ingestion via grazing contaminated swards. We have investigated the detection, confirmation, and metabolism of the beta-adrenergic agonist ractopamine administered as Paylean to the horse. A Testing Components Corporation enzyme-linked imunosorbent assay (ELISA) kit for ractopamine displayed linear response between 1.0 and 100 ng/ml, with an 1-50 of 10 ng/ml, and an effective screening limit of detection of 50 ng/mL. The kit was readily able to detect ractopamine equivalents in unhydrolyzed urine up to 24 h following a 300-mg oral dose. Amitraz (N′-(2,4-dimethylphenyl)-N-[[(2,4-dimethylphenyl)imino] methyl]-N-methyl-methanimidamide) is an alpha-2 adrenergic agonist used in veterinary medicine primarily as a scabicide- or acaricide-type insecticide. As an alpha-2 adrenergic agonist, it also has sedative/tranquilizing properties and is, therefore, listed as an Association of Racing Commissioners International Class 3 Foreign Substance, indicating its potential to influence the outcome of horse races. We identified the principal equine metabolite of amitraz as N-2,4-dimethylphenyl-N′-methylformamidine by electrospray ionization(+)-mass spectrometry and developed a gas chromatographic-mass spectrometric (GC-MS) method for its detection, quantitation, and confirmation in performance horse regulation. The phosphoprotein (P) of vesicular stomatitis virus (VSV) is an essential subunit of the viral RNAdependent RNA polymerase (RdRp) complex. It is phosphorylated at two different domains. Using defective interfering (DI) RNA or minigenomic RNA templates, we previously demonstrated that phosphorylation within the amino-terminal domain I is essential for transcription, whereas phosphorylation within the carboxyterminal domain II is necessary for replication. For the present study, we examined the role of the phosphorylation of residues in these domains in the life cycle of VSV. This study occurred in the Rural Academic Unit-Tiahuanaco installations, of the Bolivian Catholic University-La Paz, Bolviia, with the objective of evaluating macro- and microscopic characteristics of sperm ejaculation from bulbourethrectomized llamas. Six q'ara-variety male llamas of 3, 4, and 5 years of age were used over 8 weeks during which they were fed with natural and cultivated pastures. Ejaculate was collected with an artificial vagina with stimulation (libido) of male llamas by female llamas, for macro- and microscopic evaluation (volume, pH, color, appearance, motility, concentration, and sperm vitality). Isoxsuprine is used to treat navicular disease and other lower-limb problems in the horse. Isoxsuprine is regulated as a class 4 compound by the Association of Racing Commissioners, International (ARCI) and, thus, requires regulatory monitoring. A gas chromatography-mass spectrometry method utilizing electron impact ionization was developed and validated for the quantitation of isoxsuprine in equine plasma or equine urine. The method utilized robotic solid-phase extraction and tri-methyl silyl ether products of derivatization. Products were bis-trimethylsilyl (TMS) isoxsuprine and tris-TMS ritodrine, which released intense quantifier ions m/z 178 for isoxsuprine and m/z 236 for ritodrine that were products of C-C cleavage. …

Reviewing the mare reproductive loss syndrome (MRLS), it is proposed that the fundamental mechanism of this syndrome, which includes early fetal loss, late fetal loss, uveitis, pericarditis, and encephalitis, is tissue penetration by septic barbed setal fragments (septic penetrating setae) from Eastern tent caterpillars (Malacosoma americanum). Once ingested, these barbed setal fragments migrate through moving tissues, followed by rapid hematogenous spread of bacteria, bacterial emboli, and/or septic fragments of setae (septic penetrating setal emboli), collectively referred to as septic materials. Pathogenic bacteria, therefore, enter the horse as hitchhikers on or in the caterpillar setal fragments, and MRLS is caused by …

Mycobacterium bovis infection in wildlife and feral species is a potential source of infection for livestock and a threat to protected and endangered species. The aim of this study was to identify Spanish wild animal species infected with M. bovis through bacteriological culture and spacer oligonucleotide typing (spoligotyping) of isolates for epidemiological purposes. To determine the ability of experimentally inoculated white-tailed deer (Odocoileus virginianus) to transmit Mycobacterium bovis to naive deer through the sharing of feed, four deer were intratonsillarly inoculated with 4x105 colony-forming units of M. bovis. On a daily basis, feed not consumed by inoculated deer after approximately 8 hr was offered to four naıve deer maintained in a separate pen, where direct contact, aerosol transmission, or transmission through personnel were prevented. After 150 days, naıve deer were euthanized and examined. All naıve deer had lesions consistent with tuberculosis and M. bovis was isolated from various tissues. …

Tuberculosis due to Mycobacterium bovis in captive Cervidae was identified as an important disease in the United States in 1990 and prompted the addition of captive Cervidae to the USDA Uniform Methods and Rules for eradication of bovine tuberculosis. As well, M. bovis infection was identified in free-ranging white-tailed deer in northeast Michigan in 1995. Tuberculosis in both captive and free-ranging Cervidae represents a serious challenge to the eradication of M. bovis infection from the United States. Assisted reproductive technologies developed in domestic cattle serve as a starting point in similar studies on nondomestic bovids. The common eland is a useful model species for studies on rare tragelaphine antelopes. In Chapter 3 of the present study, effects of components/attributes of protein-free embryo culture media on the in vitro development of in vitro-derived bovine embryos were evaluated. During the course of their disease, leprosy patients may experience two types of inflammatory reactions- erythema nodosum leprosum (ENL) or reversal reaction (RR). Thalidomide is effective treatment for ENL, but not for RR. Using concentrations of thalidomide similar to that achieved in the treatment of ENL, we investigated thalidomide’s effect on reactions, viability of M. leprae, and integrity of plasma membranes. These studies were conducted to elucidate the regional analgesic effect that is observed clinically after treatment of orthopedic disorders with application of extracorporeal shock waves in horses. Regional analgesia after treatment with extracorporeal shock waves presents a concern because it may eliminate protective limiting mechanisms and may place equine athletes with predisposing lesions at risk of sustaining career- or life-ending injuries. Direct percutaneous application of non-focused extracorporeal shock waves to palmar digital nerves in the pastern area of horses resulted in decreased sensory nerve conduction velocities compared with untreated control nerves at 3, 7, and 35 days after treatment. Cranial cruciate ligament (CCL) rupture is a common injury in the dog and major cause of degenerative joint disease. The pathophysiology of CCL rupture in the dog is well described. Osteoarthritis secondary to CCL rupture causes severe pain and lameness. There are many surgical techniques accepted for dogs with CCL rupture. The purpose of this study was to investigate methods to induce earlier onset of cyclicity in transitional mares. Two experiments were conducted evaluating the effect of follicular aspiration to advance the onset of cyclicity, more succinctly define criteria for selection of mares for follicular aspiration and to compare aspiration to deslorelin treatment for initiating cyclicity in transitional mares. In Experiment 1, anestrous mares were assigned to control (n=6) or follicular aspiration (n=11). The control mares were monitored twice weekly, until ovulation was detected. Brucellosis and pseudorabies lead to abortion in pregnant sows and are perpetuated by feral swine reservoirs. A multivalent oral vaccine for these diseases would improve vaccination and eradication programs worldwide. Previous studies have shown that the rough attenuated Brucella strains RB51 and VTRS1, when administered subcutaneously to swine, stimulate host immune responses, transiently colonize tissues, and provide partial protection against virulent B. suis infection in pregnant sows. An emerging epizootic of mycobacteriosis currently threatens striped bass Morone saxatilis populations in Chesapeake Bay, USA. Several species of mycobacteria, including Mycobacterium marinum, species resembling M. avium, M. gordonae, M. peregrinum, M. scrofulaceum and M. terrae, and the new species M. shottsii have been isolated from diseased and healthy bass. In this study, we describe the ultrastructure of developing M. marinum granulomas in experimentally infected bass over a period of 45 wk. The leading viral killer of commercially produced channel catfish is Channel Catfish Virus (CCV). Studies conducted to evaluate DNA vaccination against CCV compared encoded gene, dose, multiple DNA vaccines, and immune response to vaccination. Genes were selected (ORFs 1 and 3 [immediate early genes], ORFs 6, 19, and 46 [membrane genes], and ORF59 [putative major envelope glycoprotein gene], cloned into a plasmid, and expressed in mammalian and fish cell culture to detect predicted molecular weight proteins. TRANSMISSIBLE spongiform encephalopathies, of which ovine scrapie is a prototype, are neurodegenerative disorders characterised by the accumulation of an abnormal isoform (PrP-scrapie or PrPSc) of a normal cellular protein (PrPC) (Prusiner 1982). Characteristics of the disease include long incubation periods, and a progressive and chronic clinical course resulting in death (Fraser 1976). In this study, 212 Acanthobrama marmid Heckel, 1843 individuals were caught at monthly periods from Keban Dam Lake between January 1997 and December 1998 and transported live to the laboratory. After haematological analysis was carried out on the blood samples taken from their hearts, total lengths and weights were measured. The sex of the fish was identified and their ages were determined from the scales. The total amounts of erythrocytes, leucocytes, thrombocytes, haemoglobin and haematocrits and MCV, MCH and MCHC parameters were determined from blood samples. This study aimed to investigate the critical control points for the contamination in a factory experiencing a problem with coliform bacteria and Escherichia coli, and to prevent economic losses and also to protect public health by facilitating production under hygienic conditions. Samples were taken from a total of 20 control points, which involved every stage of production from the beginning to the end, including milk, all equipment, surfaces, air, hands of staff and water; yoghurt samples were taken on the same day. The sex composition, age, length and body mass relationship, Von Bertalanffy growth equation and condition factor of 342 individuals were investigated to determine some population parameters of goldfish (Carassius auratus L., 1758) between June 1998 and May 1999 in Lake Eğirdir. Sex composition was 32.75 % females and 67.25 % males and the fish were 0-IV years old. The length-body mass relationship was W = 0.0210 L^{3,0598}, the Von Bertalanffy growth equation was Lt = 122.7 [1-e^{-0.033(t+3.74)}] and the average condition factor was 2.520 ± 0.012. The aim of this study was to evaluate the effects of the addition of 0.5% urea or 0.5% urea plus 4% molasses into some sorghum varieties (Gözde, P-988, Grazer and Grass-II) harvested at the dough stage on silage quality, in vitro dry matter (DM) digestibility and digestible DM yields. Triplicate silage samples from each variety and treatment group, a total of 36 silage samples, were prepared in 1 l mino-silos and incubated for 90 days. This study was carried out to determine the cardiac reference parameters of Karabash dogs, a breed specific to Turkey, using M-mode and 2-D echocardiography techniques. The material of the study comprised 25 male and 25 female Karabash dogs aged between 2 and 6 and weighing between 30 and 51 kg. The spermatological characteristics of merino rams' semen collected with an artificial vagina (AV) and electro-ejeculator (EE) were compared. The semen was evaluated for volume (ml), mass activity (0-5, no activity to rapid wave), motility (%), sperm concentration (x 10^9/ml) and abnormal spermatozoa percentage. This study was conducted to determine the effects of the solar eclipse of August 11, 1999, on the behavior of birds (chickens, Pekin ducks, gulls, crows and sparrows), cattle, bees and horses. The animals were observed for 6, from 11 am to 5 pm. Abnormal and extreme behaviors were determined at the observation times. The present study was conducted to investigate the effects of probiotics on the live weight, live weight gain, feed consumption, feed efficiency and health condition of calves. Nineteen Holstein calves, divided into 2 similiar groups in terms of age, sex and live weight, were used in the trial.
