Selenium methionine (Se Met) is an essential micronutrient required for normal body function and is associated with additional health benefits. However, oral administration of Se Met can be challenging due to its purported narrow therapeutic index, low oral bioavailability, and high susceptibility to oxidation. To address these issues, Se Met was entrapped in z e i n-coated nanoparticles made from chitosan using an ionic gelation formulation. The high stability of both the Se Met and s e l e n o methionine nanoparticles (Se Met-NPs) was established using cultured human intestinal and liver epithelial cells, rat liver homogenates, and rat …

Transmission Competence Of B. Burgdorferi Infected Ixodes Ticks And Development Of An Ospc-Based Lyme Disease Vaccine, Samanta Kamalika
Theses and Dissertations (ETD)
Introduction. Lyme disease, otherwise called Lyme borreliosis, is a vector-borne infectious disease caused by the spirochete Borrelia burgdorferi (Bb). Studying the increase in overall Lyme disease incidence requires understanding the role of the vector Ixodes scapularis (I. scapularis) as the agent that carries the spirochete which is important to limit the occurrence of Lyme disease. Once we understand the vector-pathogen response, it also becomes important to determine the remedies for Lyme disease. Lyme disease can be treated with antibiotics if detected early, but the major obstacle lies in the early detection process. Therefore, it is important to investigate alternative preventive …
Modeling The Bidirectional Relationship Between Shared-Patient Physician Networks And Patient Longitudinal Treatment Patterns: Application To Physician Risky-Prescribing, Xin Ran
Dartmouth College Ph.D Dissertations
Risky-prescribing is a pressing public health concern in the United States. Opioids, benzodiazepines, and non-benzodiazepine sedative-hypnotics (sedative-hypnotics) are three commonly-prescribed but potentially risky drug groups, prescribed alone or in combination. Physician shared-patient networks provide a unique perspective in studying physician network characteristics and structures, as well as their association with the delivery of health care. Understanding how physician shared-patient networks are related to their prescribing may inform network-based interventions targeting risky-prescribing, which is yet to be fully studied.
We investigated patient receipt of risky prescriptions and physician risky-prescribing intensity through the scope of shared-patient networks. We used retrospective Medicare insurance …

Despite being an ancient disease caused by Mycobacterium tuberculosis (Mtb) and after decades of research, tuberculosis (TB) still affects millions of people every year worldwide. In 2018, the World Health Organization (WHO) reported that 10 million people developed tuberculosis and 1.5 million died of the disease. With the increase in the multidrug-resistant (MDR) and extensively drug-resistant (XDR) cases, the treatment for TB with the standard first and second-line therapy is becoming increasingly difficult. Therefore, there is an urgent need to find new anti-TB drugs as combination partners of existing and experimental classes of antimicrobial agents to shorten and simplify the …

Organ transplants are a vital intervention for many diseases that result in end stage organ failure. Currently, the donation pool is not meeting the demands of the transplant list. Expanding this pool to include donation after cardiac death (DCD) is highly sought-after. However, the use of DCD livers can lead to increased odds of graft failure and ischemic cholangiopathy. The loss of epithelialization and fibrosis that occurs during ischemic cholangiopathy is characteristic of these cells undergoing epithelial-to-mesenchymal transition (EMT). The biological changes the cell experiences enhance migratory capacity, invasiveness, and increased resistance to apoptosis. Our earlier studies have shown differential …
The Impact Of Hiv-1 Tat And Morphine On Spatial Distribution Of Drugs In The Brain., Austin M. Jones
The Impact Of Hiv-1 Tat And Morphine On Spatial Distribution Of Drugs In The Brain., Austin M. Jones
Theses and Dissertations
Despite combination antiretroviral therapy effectively suppressing HIV within the periphery, the central nervous system (CNS) remains affected by the virus. Approximately half of people living with HIV will experience HIV-associated neurocognitive disorders (HAND) or neuroHIV. Concurrent opioid use exacerbates neuroHIV by promoting neuroinflammation, viral replication, and potentially altering the antiretroviral concentrations within the brain.
Using a transgenic mouse that expresses the HIV-1 Tat protein, we examined the effects of Tat and morphine on antiretroviral accumulation and distribution and the effects of Tat on morphine accumulation within the brain using infrared matrix-assisted laser desorption electrospray ionization with mass spectrometry imaging (IR-MALDESI-MSI). …

Development Of A Chemical Biology Approach To Uncover The Influence Of Sequence Variations On Ces1 Activity In Live Cells, Samuel James Knebel
Masters Theses
Drug metabolism is the biochemical process of modifying drugs to detoxify and remove them through enzymatic transformations. These biotransformation’s occur primarily in the liver and are critical to understanding how pharmaceutical compounds are chemically altered inside the human body. Human carboxylesterases (CESs) catalyze the hydrolysis of esters, amides, thioesters, and carbamates. CES-mediated hydrolysis plays an important role in the metabolism of many drugs including the first FDA approved antiviral treatment for COVID-19, remdesivir (Veklury), the seizure control medication rufinamide (Banzel), and the flu antiviral drug oseltamivir (Tamiflu). CES activity is known to be influenced by a variety of factors including …

Modified Tlr7/8 Adjuvanted Liposomes For Improved Intranasal Delivery Of Subunit Vaccines, Elizabeth Brander Lorentz
Graduate Student Theses, Dissertations, & Professional Papers
Intranasal vaccines hold enormous potential for improving vaccine efficacy against respiratory pathogens by activating the mucosal immune system. The urgency for novel approaches in vaccine development is amplified by the recent COVID-19 pandemic. Most intranasal vaccines that were developed are either live-attenuated or inactivated vaccine candidates that demonstrated poor safety profiles. Protein-based subunit vaccines represent a safe and cost-effective alternative, but they are often weak immunogenically. TLR7/8 adjuvants such as INI-4001 have shown promise for enhancing the humoral and cellular immunogenicity of protein-based subunit vaccines. It is theorized that poor immunogenicity of intranasal protein subunit vaccines is due to low …

Personal Hardiness As An Unexamined Component Of The Healthy Volunteer Effect, Angelique E. Blann
Walden Dissertations and Doctoral Studies
Clinical trials are the standard for approving medicines for public use. To conduct trials, researchers depend on a declining number of volunteers. The healthy volunteer effect, a phenomenon in which participants demonstrate better outcomes than their peers, limits the generalizability of trial outcomes. The healthy volunteer effect does not fully explain the outcome difference between volunteers and nonvolunteers. The purpose of this cross-sectional quantitative study was to examine personal hardiness as an unexplored component of the healthy volunteer effect and determine whether personal hardiness is associated with willingness to participate. Hardiness was a personality construct that represented resilience to stress. …

The participants were five independent pharmacy managers who implemented strategies to mitigate the …
Role Of Cysteine Redox State In Altering The Epithelial-Mesenchymal Transition In Lung Cancer, Md Anisur Rahman
Role Of Cysteine Redox State In Altering The Epithelial-Mesenchymal Transition In Lung Cancer, Md Anisur Rahman
University of the Pacific Theses and Dissertations
Epithelial-mesenchymal transition is a process by which cancer cells increase their capability for metastasis. EMT involves conversion of epithelial cells into a mesenchymal phenotype which makes cancer cells more invasive and motile. Lung cancer has the highest mortality rate among different types of cancers globally. Lung cancer exists in various types, the most prevalent of which is non-small cell lung cancer (NSCLC). Several factors, including oxidative stress, can induce EMT in cancer cells. However, the mechanism of how reactive oxygen species (ROS)-induced changes in cysteine redox state affect EMT remains elusive. Our studies aim to understand this mechanism by inducing …

Investigating Bzip Recognition Of Dna Sequences Through A Knob-Socket Perspective, Aaron Tran
University of the Pacific Theses and Dissertations
To investigate whether higher order packing interactions confer protein-DNA specificity, a modified Knob-Socket (KS) model was used to analyze the interface of bZIP-DNA crystal structures. The KS analysis identified a nine-residue quadripartite recognition core consisting of four contiguous KS pockets P1, P2, N3, and N4 that each pack one of the four DNA half-site bases in the target sequence. Only one base per base pair packs, and these interactions are split across the DNA strands: the first two positive strand positions 1p and 2p pack into P1 and P2 while the last two negative strand positions 3n and 4n pack …

UROP Posters
Purpose: Systemic treatment of colorectal cancer involves chemotherapeutic agents which elicit serious and negative side effects from the toxicity of the drug. To address this issue, we are testing dietary supplements for their efficacy against human colon cancer cell lines and also their potential synergistic effects when combined with conventional chemotherapy. Dietary supplements (specifically sesamol and black rice extract) exhibit anticancer, anti-inflammatory, and chemo-preventive properties. Meanwhile, one of the cancer resistance mechanisms is the upregulation of drug elimination mechanisms, leading to multi-drug resistance. We hypothesize that dietary compounds will act as chemo-enhancers, thus enhancing potency of the chemotherapy drug(s) on …

Protective Effect Of Naringenin Against Indomethacin-Induced Lysosomal Toxicity, Kaitsuda Saiprom
Protective Effect Of Naringenin Against Indomethacin-Induced Lysosomal Toxicity, Kaitsuda Saiprom
Chulalongkorn University Theses and Dissertations (Chula ETD)
An increase in lysosomal membrane permeabilization (LMP) impaired lysosome integrity causing release of cathepsin B, and subsequently may contribute to cell death. Indomethacin, which is a non-steroidal anti-inflammatory drugs (NSAIDs), caused apoptosis through generation of cell stress, and disruption of mitochondria and lysosome functions. Recently, it was reported that naringenin, a natural bioactive flavonoid, elicited cytoprotection against indomethacin toxicity in Caco-2 cells. This study investigated the effect of naringenin on indomethacin-induced lysosome toxicity in human colon carcinoma Caco-2 cells. Cells were pretreated with naringenin (100 µM) for 24 h before incubating with indomethacin for another 48-72 h. At the end …

A simple, accurate, and low-cost effective method has been introduced in this study for separation of valsartan enantiomers using ligand-exchange chromatography as chiral mobile phase additives. The optimized method was found using a mobile phase system of 57%(v/v) methanol, adding 1.5 mM of L-isoleucine as the chiral selector along with 0.5 mM of cupric sulfate and 10 mM of sodium acetate, adjusted to pH 6.1. The flow rate was 0.8 mL/min with a temperature of 400C. The injection volume was 10 µL and detected at 210 nm. The stationary phase was a reversed-phase C18 Zorbax Eclipse Plus (250 mm x …

Respiratory Syncytial Virus (RSV), a highly contagious virus known to cause severe respiratory illnesses, poses a significant health threat, particularly impacting vulnerable populations such as young children, the elderly, and immunocompromised individuals. It is a leading cause of lower respiratory tract infections, often resulting in serious complications like pneumonia and bronchiolitis. Despite its substantial impact, there is currently only an approved RSV vaccine specifically designed for the elderly, emphasizing the urgency of continued research in this area for developing broader coverage of the population. The plant expression platform for producing therapeutic proteins offers a cost-effective and scalable solution, presenting a …
