Observations Related To The Mechanisms Of Tetracycline-Induced Lipid Accumulation In A Strain Of Mouse Fibroblasts Cultivated In Vitro, Dale William James Jan 1971

Observations Related To The Mechanisms Of Tetracycline-Induced Lipid Accumulation In A Strain Of Mouse Fibroblasts Cultivated In Vitro, Dale William James

University of the Pacific Theses and Dissertations

Intravenous administration of tetracycline has been known to result in fatty degeneration of the liver. This study was undertaken in order to elucidate some of the actions of tetracycline on mammalian cells in culture and to attempt to relate these findings to the clinical condition of fatty degeneration of the liver mentioned above.

It is suggested that lipid accumulation occurring after tetracycline administration is possible the result of a decrease in B-lipoprotein synthesis and enzyme inhibition. These factors could result from the high blood levels of tetracycline seen after large and rapid intravenous injections of the drugs into normal patients …

A Pharmacologic Investigation Of The Antiinflammatory Activity Of Cryogenine, Donald Saadia Kosersky Jan 1971

A Pharmacologic Investigation Of The Antiinflammatory Activity Of Cryogenine, Donald Saadia Kosersky

University of the Pacific Theses and Dissertations

This study was conducted in an attempt to define the anti inflammatory efficacy of cryogenine, the major alkaloid of Heimia salicifolia Link and Otto. The anti inflammatory effects produced by cryogenine, phenylbutazone, indomethacin, mefenamic acid, paramethasone, hydrocortisone and 6-mercaptopurine were compared considering the sequential changes in various motameters during the course of adjuvant-induced polyarthritis in rats. Polyarthritis was induced by the subplantar injection of a suspension of heat-killed Mycobacterium butyricus in mineral oil into the right hind paw of Sprague-Dawley rats. The manifestations of the diseases were monitored in terms of hind paw swelling, subjective appearance of secondary lesions, impairment …

Antibradykinin Evaluation Of Cryogenine And Selected Other Agents, Ralph William Trottier Jr. Jan 1971

Antibradykinin Evaluation Of Cryogenine And Selected Other Agents, Ralph William Trottier Jr.

University of the Pacific Theses and Dissertations

One intent of this study was to detect and to define any antibradykinin capacity that cryogenine may have using both in vivo and in vitro pharmacological models. The activity of cryogenine in these tests was compared to a variety of clinically-proven anti-inflammatory compounds as well as to other agents known to possess anti-inflammatory activity in certain experimentally-induced inflammations. Cryogenine is an unusual anti-inflammatory agent since it is an alkaloid - all of the commonly used anti-inflammatories (aspirin, phenylbutazone, indomethacin, etc.) are organic acids.

The Effects Of Indomethacin On A Strain Of Fibroblasts Cultivated In Vitro, John Thomas Mccloskey Jan 1971

The Effects Of Indomethacin On A Strain Of Fibroblasts Cultivated In Vitro, John Thomas Mccloskey

University of the Pacific Theses and Dissertations

Man has always searched for greater understanding of himself and his environment. As a result, much scientific investigation has been directed towards the understanding of the life process. In recent years, such scientific inquiry has become increasingly focused on the fundamental biochemical events taking place at the cellular, subcellular, and molecular levels. In the course of investigation into the nature of living things, research has often necessitated the development of new techniques and investigational tools to serve as experimental systems. Very often such new methods have made possible the discovery of previously unknown processes. One very interesting and useful investigational …

The Dialysis Of Caffeine Through Selected Semi-Permeable Membranes, Paul James Perry Jan 1971

The Dialysis Of Caffeine Through Selected Semi-Permeable Membranes, Paul James Perry

University of the Pacific Theses and Dissertations

Until the past few years, cellulose derivatives, (e.g., cellophane, collodion, and parchment) and animal membranes (e.g., goldbeater’s skin) have been the only dialysis membranes employed commercially. Cellophane has been used as the dialysis membrane in the artificial kidney since the machine’s inception in 1914. It continues to serve in this capacity, even though, in the last few years, attempts have been made to develop better films.

An appreciation of both the “solution theory” and the “pore theory” is in order for this discussion. By incorporating the dynamics of these theories in the techniques of membram formulation, improved membrane performance can …

The Attempted Synthesis Of 4,4-Dimethyl-3-Dimethylamino-2,6-Cyclohexanedione Carboxamide, Roger Leith Cockerline Jan 1971

The Attempted Synthesis Of 4,4-Dimethyl-3-Dimethylamino-2,6-Cyclohexanedione Carboxamide, Roger Leith Cockerline

University of the Pacific Theses and Dissertations

The use of boron-containing amines for the amino alkylation of amides has been investigated in our laboratories with the aim of preparing compounds which could be used in neutron capture therapy. The purpose of this procedure is to achieve selective irradiation of a neoplactic growth by inducing radioactivity throughout that tissue. Although the results obtained in destroying tumors have not always been encouraging, they have been such - especially in cases not amenable to treatment by surgery or chemotherapy - that further research is warranted in order to develop drugs that can be used effectively in neutron capture therapy (Farr, …

Differential Spectrophotometric Analysis Of Intravenous Admixtures Containing Metaraminol With Selected Corticosteroids, Frederick Erin Turner Jan 1970

Differential Spectrophotometric Analysis Of Intravenous Admixtures Containing Metaraminol With Selected Corticosteroids, Frederick Erin Turner

University of the Pacific Theses and Dissertations

Intravenous admixtures consisting of one or more drug formulations, diluted in a large volume of intravenous solution, are used extensively in current medical practice. Development of reliable methods to predict the compatibility of the admixture components is essential as a guide for their selection.

Because the majority of the reports published, to date, have used a visible change as the criterion for judging compatibility, it was the purpose of this study to demonstrate the use of spectrophotometric analysis to detect the occurrence of chemical interactions between two components of an intravenous admixture.

Interactions Of Sodium Ethacrynate In Intravenous Admixture With Selected Cardiovascular And Psychotherapeutic Agents, Patrick N. Catania Jan 1970

Interactions Of Sodium Ethacrynate In Intravenous Admixture With Selected Cardiovascular And Psychotherapeutic Agents, Patrick N. Catania

University of the Pacific Theses and Dissertations

The possibility of drug incompatibilities is a matter of serious concern when administering two or more therapeutic agents. This is especially true in the case of parenteral administration when one must be aware not only of therapeutic interactions but also chemical and physical reactions that might occur.

In order to avoid incompatibilities, drug manufacturers have suggested that parenteral solutions be used immediately after reconstitution and that admixtures of parenteral products not be administered wherever possible (1,2). Physicians have also stressed the importance in avoiding multiple drug therapy (3,4). In spite of these suggestions, the practice of multiple drug therapy is …

An Investigation Of The Release Of 5-Fluorouracil From Ointment Bases, Vance Leroy Grainger Jan 1969

An Investigation Of The Release Of 5-Fluorouracil From Ointment Bases, Vance Leroy Grainger

University of the Pacific Theses and Dissertations

During the several years in which this therapy has been developed and evaluated, as standard dermatological preparation has not been available. The objective of the present work was to study the release of 5-FU from various ointment bases and to attempt to determine the type of base which would release the compound most satisfactorily.

The methods chosen were designed to determine the release and penetration of 5-FU both by in vitro and in vivo methods. A modified agar plate method was chosen as the in vitro test for measuring release and subsequent diffusion into the agar. The penetration of the …

A Comparison Of The Fluidized Bed Drying Technique With Conventional Methods Of Drying Tablet Granulations, Kit Michael Mills Jan 1969

A Comparison Of The Fluidized Bed Drying Technique With Conventional Methods Of Drying Tablet Granulations, Kit Michael Mills

University of the Pacific Theses and Dissertations

Drying procedures, like other important unit operations in pharmacy, have received little attention in pharmaceutical literature. Within recent years, however, growing interest in these pharmaceutical engineering areas has become apparent.

Fluidization operations, although relatively unexplored in pharmacy, have been firmly established on a broad scale in other industries. Fluidization may be defined as the suspension and agitation of a bed of particulate solids by a vertically rising stream of gas. Each suspended particle is surrounded by the gas. This relative velocity of the gas and solid particles is high, although the gas velocity itself is relatively low. Heat is transferred …

Studies On The Localization And Retention Of Tetracycline Hydrochloride In Ehrlich Ascites Cells, Dale William James Jan 1969

Studies On The Localization And Retention Of Tetracycline Hydrochloride In Ehrlich Ascites Cells, Dale William James

University of the Pacific Theses and Dissertations

The antibiotics have become so widely and successfully used in the practice of medicine that they are now considered to be essential. Among the antibiotics, the tetracycline group has achieved an enviable reputation for dependable medication, In spite of this, there remain several unanswered questions, One of these pertains to the interesting phenomenon for almost selective localization of tetracycline in certain tumor cells or masses, Although there have been numerous publications dealing with this phenomenon, as the following paragraphs will indicate, there is little agreement as to the factors involved. Indeed, there are some sharply conflicting opinions.

The investigations described …

A Synthesis Of A Homologous Series Of Aryl Mono- And Diboronic Acids, Larry Milton Allen Jan 1967

A Synthesis Of A Homologous Series Of Aryl Mono- And Diboronic Acids, Larry Milton Allen

University of the Pacific Theses and Dissertations

Many attempts have been made to prepare aryl boronic acids and their esters which show a preferential affinity for tumor tissues for use in radiation therapy of neoplatic disorders (1,2). The data of Soloway, as extended and refined by Hansch and his coworkers, successfully correlates the low lipid solubility and electron releasing substituents of phyenl boronic acid derivatives with increased tumor concentration (3,4,5). However, most of these organoboron compounds do not produce a sufficiently high tumor-to-normal tissue boron concentration ratio for practical use in human cancer chemotherapy by the neutron capture technique. The knowledge that several chemical substances are preferentially …

Studies On The Essential Oil Of Anemopsis Californica, Ramesh Narmadashanker Acharya

University of the Pacific Theses and Dissertations

The roots and rhizomes of Anemopsis Californica (Nutt.) Hook and Arn. (family Saururaceae) have been used by the early Spanish Americans and Indians to treat a variety of ailments. The plant is commonly known as Yerba del Mansa, Yerba Mansa, or Manza.

We decided to investigate the essential oil of Anemopsis Californica in greater detail. The results of our investigation to date are included and described in this thesis.

Studies On The Metabolism And Excretion Of Chlorine Chloride, John Thomas Mccloskey Jan 1967

Studies On The Metabolism And Excretion Of Chlorine Chloride, John Thomas Mccloskey

University of the Pacific Theses and Dissertations

The organic base choline is of considerable interest to the physiologist because of its role in normal metabolic function, and to the pharmacologist because of its possible application in the treatment of several human diseases.

The overall function of choline in the biologic system must be seen as an equilibrium between its lipotropic activity as a whole molecule and its function as a source of methyl groups in biochemical reactions.

IV. Investigation Undertaken in This Study

Due to the long-standing and as yet unresolved question dealing with the relative merits of the path by which choline leaves the intestine, these …

A Study Of Several Factors Incident To The Absorption Of Choline From The Small Intestine Of The Albino Rat, Ralph E. Purdy Jan 1967

A Study Of Several Factors Incident To The Absorption Of Choline From The Small Intestine Of The Albino Rat, Ralph E. Purdy

University of the Pacific Theses and Dissertations

The substance choline, one of the quaternary nitrogen bases, has been the subject of a number of reviews and investigations. It has been described as a cholinergic agent, an inotropic agent, a vitamin, and perhaps in other ways. In spite of the interest shown in its action and uses, there is as yet very little information available as to the mechanism by which it is absorbed by the intestinal mucosa.

The studies referred to thus far leave the field open for additional investigation. No mention was found in the literature of studies on selective areas of absorption of choline in …

The Application Of A Modified Macmichael Viscometer In Rheologic Studies Of Montmorillonite Clays, Billy Edwin Farley Jan 1966

The Application Of A Modified Macmichael Viscometer In Rheologic Studies Of Montmorillonite Clays, Billy Edwin Farley

University of the Pacific Theses and Dissertations

Available instruments for viscosity and rheologic measurements necessitate disruption of the system under test prior to observations. Generally, the gel structure of a thixotropic clay must be disturbed by shaking, pouring into a container for testing, and again upon insertion into the sample of the bob, spindle or paddle of the instrument. Also, these instruments yield a single point reading which is only a final or average value, depending upon the instrument. The occurrences during the test itself cannot be observed. Accordingly? it seemed feasible to determine, if possible, what other rheologic factors are involved in the breakdown of ·the …
