Oculomotor Deficits In Diseases Of The Basal Ganglia: Parkinson's And Huntington's Diseases, Todd B. Baird

Oculomotor deficits are now recognized as being present in several neurological diseases of the basal ganglia. The present report will focus primarily on those observed in Huntington's and Parkinson's diseases. Neuronal cell loss in the pars compacta of the substantia nigra, degeneration of the nigrostriatal pathway, and consequent depletion of the neurotransmitter dopamine is the most obvious etiological abnormality in Parkinson's disease. Huntington's disease, on the other hand, involves the selective genetically-driven atrophy of the striatum (caudate and putamen). Rhesus monkeys (Macaca mulatta) reared during the first year of life without social contact develop persistent stereotyped movements, self-directed behaviors, and psychosocial abnormalities, but neurobiological mechanisms underlying the behaviors of socially deprived (SD) monkeys are unknown. Monkeys were reared in total social deprivation for the first 9 months of life; control monkeys were reared socially (SR) with mothers and peers. Subjects were killed at 19-24 yr of age. Because the behaviors of SD monkeys are reminiscent of changes in striatal or amygdalar function, we used immunocytochemistry for substance P (SP), leutine-enkephalin (LENK), somatostatin, calbindin, and tyrosine hydroxylase (TH) to evaluate …

Acid demineralized bone (DMB) implanted in extraosseous sites induces bone formation. In vivo studies have shown that DMB stimulates the differentiation of mesenchymal cells into osteogenic cells. In this study an in vitro system was developed to demonstrate the osteoinductivity of DMB quantitatively. This in vitro working system was used to assess the osteoinductivity of variously processed DMB products as a means to evaluate their processing protocols. A human dermal fibroblastic (HDF) cell line was initiated from tissue explants as a working model, along with a human periosteal (HPO) cell line as a control. The effect of ethanol on bone histomorphology was studied using an isocaloric liquid pair feeding system to administer equal amounts of essential vitamins and nutrients to control and ethanol animals.
A bone remodeling sequence was induced in the lower right mandible by removal of the upper right molars. The bone remodeling unit, consisting mainly of the osteoclast and osteoblast, could then be studied microscopically to note any changes brought about by the administration of ethanol.

The purpose of this investigation was to determine the relationship between total blood cholesterol levels, blood triglyceride levels, and blood HDL levels in men and women 20 to 80 years of age. All subjects fasted for 12 hours prior to having their blood drawn.
The computer program SPSSX (SPSSX, Inc., 1988) was used along with subprograms CONDESCRIPTIVE, CORRELATION, and ANOVA. Pearson Product Moment Correlations were calculated for all subjects, for male subjects, and for female subjects.
The study revealed a low correlation coefficient between blood cholesterol levels and blood triglyceride levels for all subjects, female subjects and male subjects. There …

Meissner corpuscles in forepaw digital pads of albino mice were examined by light and electron microscopy to determine structural age-changes. After 1.5 months of age, no intraepidermal nerve fibers were seen extending from corpuscles. From young (1.5-6 months) to middle age (9-15 months), corpuscles became larger and more complex, gaining more horizontally arranged terminals, associated lamellae and connective tissue. At old age (18-26 months), corpuscles became small and lobulated, appearing disorganized. There was no loosening of the corpuscle-epidermal interface. An increase in collagenous connective tissue and basal lamina duplication occurred with advancing age. Ultrastructural age-changes consisted of disorganization of the …

When defined in terms of markers for normal cell lineages, most invasive breast cancer cells correspond to the phenotype of the common luminal epithelial cell found in the terminal ductal lobular units. Luminal epithelial cells cultured from milk, which have limited proliferative potential, have now been immortalized by introducing the gene encoding simian virus 40 large tumor (T) antigen. Infection with a recombinant retrovirus proved to be 50-100 times more efficient than calcium phosphate transfection, and of the 17 cell lines isolated, only 5 passed through a crisis period as characterized by cessation of growth. The density and distribution of labeled cortical cells was examined following discrete deposits of retrograde tracers into the superior colliculus (SC) of neonatal and adult rats to study the topographic order of SC afferents from the cortex during the initial phase of axon ingrowth. Small deposits of red and green latex microspheres (0.02-0.05ul) were placed into the SC of rats ranging in age from birth to adulthood using glass micropipettes (tip diameter 25-35um). There are no data available as to whether training with eccentric contractions are more effective than concentric contractions for producing skeletal muscle fiber hypertrophy. To better understand the effects of training with different contraction types two related studies were performed.
In the first study a device which is frequently used by clinicians for concentric and eccentric exercise, the Kin-ComR, was tested for accuracy and reliability. The measurements obtained from the force, angle, and speed transducers of this device were found to be accurate and reliable between days.

Post-surgical cold applications are beneficial to orthopedic surgery patients. However, their use is limited because of the difficulty in maintaining a constant temperature, increased need for attention, and unwanted leaks or condensation from ice bags. Twenty-one arthroscopic surgery patients were divided into three groups: a Dura*Kold cold pack group, an Aircast Cryo/cuff compression dressing group, and a control group. Six dependent variables were examined: subjective pain ratings three and 48 hours after surgery, a subjective rating of functional activity 48 hours after surgery, skin temperature for three hours after surgery, and pain medication consumed during three hours and 48 hours …

The electrophysiologic technique (Reuss, L., Proc. Natl. Acad. Sci. USA 82:6014, 1985) was modified to measure changes in steady-state hepatocyte volume during osmotic stress. Hepatocytes in mouse liver slices were loaded with tetramethylammonium ion (TMA$\sp{+}$) during transient exposure of cells to nystatin. Intracellular TMA$\sp{+}$ activity (a$\sp{\rm i}\sb{\rm TMA}$) was measured with TMA$\sp{+}$-sensitive, double-barreled microelectrodes. Loading hepatocytes with TMA$\sp{+}$ did not change their membrane potential (V$\sb{\rm m}$), and under steady-state conditions a$\sp{\rm i}\sb{\rm TMA}$ remained constant over 4 min in single impalements.
