Ovulation has been hypothesized as an inflammatory process. Interleukin(IL)-1$\alpha$, IL-1$\beta$ and tumor necrosis factor(TNF)-$\alpha$ are potent cytokines produced from macrophages and various other cell types, and are pivotal components of inflammation. Although previous studies have investigated cytokine activities in the reproductive system, there is little information on their precise localization and activities during the periovulatory period. To investigate the role of cytokines in ovulation, experiments were designed to determine the immunohistochemical localization and time specific production of cytokines IL-1 and TNF-$\alpha$ using a mouse model at 36h, 12h, 6h, 2h before ovulation, and at 6h and 18h after ovulation in …

Using conventional microelectrodes, I studied the electrical basis for determining the resting V$\sb{\rm m}$ in intact EC's from hamsters. The resting V$\sb{\rm m}$ were found to be $-$40 mV for aortic EC's and $-$43 mV for vena caval EC's. The contributions of ions to the resting V$\sb{\rm m}$ of aortic EC's were compared in terms of the transference number (t$\sb{\rm ion}$). To develop a technique for in situ monitoring changes in V$\sb{\rm m}$ of postcapillary venular EC's in the hamster mesentery, a voltage-sensitive fluorescent probe, bisoxonol, was used to load the cells and the fluorescence signals were analyzed under an …

The present body of knowledge concerning the results of central nervous system injury is incomplete and controversial. The role of oligodendrocytes, the cells involved in phagocytosis, the stages of demyelination, and the evidence for remyelination are of interest. Following stab wounds to rat brain cortex, corpus callosum and hippocampus, the animals were examined at 4,7,11,18,23,29,45,60 and 70 days postoperation (dpo). To help in the identification of cells immunocytochemical markers for astrocytes and immature oligodendrocytes were used. These were labels for glial fibrillary acidic protein (GFAP) and myelin/oligodendrocyte specific protein (MOSP) respectively. Little is known of the effects of proton irradiation on neural tissue. A 1.0 mm microbeam was used on the cat lateral geniculate nucleus (LGN) to quantify the short term (< 3 months) and long term (< 9 months) effects of radiation damage. The LGN is a large structure with well defined afferent, efferent, and receptive field properties.
Electrophysiological and histological techniques were used to examine the effects of the microbeam at 60, 40, and 16 Gy, administered as a single bolus. Recordings of light evoked responses in the LGN were obtained using microelectrodes in nine animals within 12 weeks of irradiation, and 6 animals within 36 weeks of irradiation. Receptive fields were mapped onto a tangent screen using standard receptive field techniques. Histological measures included afferent …

Depression of the cellular immune response to Toxoplasma gondii has been reported in both mice and humans. The present study was undertaken to determine the kinetics and mechanism of the observed downregulation of interleukin 2 (IL-2) production during experimental murine toxoplasmosis. For these investigations, the cell-mediated immune response to the wild type (PTg) was compared with that to the less-virulent mutant parasite (PTgB), which is deficient in the major surface antigen, p30 (SAG-1). Spleen cells from infected A/J mice failed to proliferate in response to Toxoplasma antigens during the first week of infection. Both PTg- and PTgB-infected A/J mice exhibited …

Denervation Supersensitivity Of The Rat Vas Deferens: A Role For Protein Kinase C, Sonny T. Abraham

A role for protein kinase C (PKC) in the denervation-induced supersensitivity of the rat vas deferens was investigated. Chronic, surgical denervation of the rat vas deferens (up to 8 days) resulted in tissues that produced enhanced contractile responses to norepinephrine (NE) in isolated organ baths. Single challenges of NE (10 $\mu$M) produced 0.6 $\pm$ 0.1 g of maximal tension in the control vas whereas in the paired, denervated tissue 2.2 $\pm$ 0.3 g of tension was recorded (n = 6). Cumulative concentration-effect curves to NE produced in the denervated vas deferens were shifted 18-fold to the left of the control …

The vascular endothelial cell (EC) plays an important role in regulating vascular tone and local blood flow by sensing chemical and mechanical stimuli on the vascular wall and releasing a host of vasoactive substances upon activations of endogenous or exogenous vasoactive substances. The central hypothesis is that local control of blood now and autoregulatory behavior in the microcirculation is distinctive at different levels of the vasculature and is dependent on the cellular activities of the EC and its interaction with the local environment. Although tachykinins have potent vascular actions, their effect on renal resistance blood vessels is currently unknown. The vasoactive properties of tachykinins and related analogs were assessed in isolated perfused rat kidney. At a basal perfusion pressure (PP) of 75 $\pm$ 6 mm Hg (n = 5), bolus injections of substance P (SP) had no significant vasoactive effect. Following a sustained increase in baseline PP (134 $\pm$ 10 mm Hg) produced by phenylephrine (1 $\mu$M), SP evoked a dose-dependent increase in PP. The largest dose of SP increased PP by 60 $\pm$ 5 mm Hg. The vasoconstrictor response to SP was …

Cell death is a common feature of neural development in all vertebrates. The bcl-2 proto-oncogene has been shown to protect a variety of cell types from programmed cell death. We have examined the distribution of bcl-2 protein in the developing and adult nervous systems. bcl-2 protein is widespread during embryonic development. Proliferating neuroepithelial cells of ventricular zones as well as the postmitotic cells of the cortical plate, cerebellum, hippocampus and spinal cord express bcl-2. Postnatally, bcl-2 is principally retained in the granule cells of the cerebellum and dentate gyrus of the hippocampus. bcl-2 expression in the CNS declines with aging. …

The following review of the p53 tumor suppressor gene will be discussed with particular attention to its role in human gliomas, as well as the various advances that have brought this molecule to the forefront of cancer research. A review of tumorigenesis focuses on the molecular mechanisms that convey neoplastic characteristics upon a normal cell. It is discussed how the coordinate advances in chromosome analysis and molecular techniques enabled the p53 gene to be categorized as a "tumor suppressor gene" and applied to various forms of cancer, including colorectal and lung carcinoma. Radiosurgery using the Linear Accelerator or the Gamma Knife has proven to be an effective treatment modality for malignant brain tumors. In comparison to other treatments, radiosurgery can be performed on an outpatient basis and is noninvasive (Table 5). Due to the functional properties of radiosurgical devices, they are ideal for patients who are unable to undergo surgical removal of their brain tumors. The sharp dose drop—off beyond the tumor margin allows for high dosage tumor irradiation while sparing normal brain tissue. Blood pressure, heart rate, and renal sympathetic nerve responses were measured in 9-13 week old male spontaneously hypertensive rats (SHR) and compared to those in age and sex-matched Wistar-Kyoto (WKY) rats following intravenous injection of the neuropeptide substance P (SP), the nicotinic stimulant 1,1-dimethyl-4-phenylpiperazinium (DMPP), and the adrenoceptor stimulant norepinephrine (NE). Charles River Sprague-Dawley (CD) rats were used in some studies to develop methodologies. Measurements were made in control rats and also following sinoaortic denervation, pithing, ganglion blockade, or adrenoceptor blockade. The perinea! gland in the hispid cotton rat, Sigmodon hispidus, is a seasonally cyclic, subcutaneously located organ which is associated with the reproductive system. On gross dissection, the gland possesses a strong attachment to the penis with only loose fascia! connections to the rectum and surrounding muscle and skin. Histologic exam confirms this connection to the reproductive tract and shows the organ to be a compound tubuloacinar gland with a projection leading into the penis. The cyclic hypertrophy and regression of the gland closely parallels that of the testes and seminal vesicles indicating that this cyclicity may be under …

Pollinator preferences were observed for the six species of section Erythranthe of the genus Mimulus using greenhouse-grown plants placed in a meadow in the Red Butte Canyon Natural Area, Salt Lake County, Utah. The principal pollinators were hummingbirds and bumble bees. Hummingbirds preferred the species with the most reflexed tubular flowers regardless of color, whereas bumblebees preferred pink. lavender, or yellow flowers to red flowers regardless of shape. Results for the six species were confirmed by observations of F2 hybrid recombinant plants selected such that flower color could be held constant and flower shape varied and vice versa.

The cranial nerves are the nerves that leave the brain directly. These nerves control various actions and sensations such as sight, smell, .chewing, and swallowing, and will be the focus of this week's lab.

While it is well known that the different sensory modalities are primarily represented at separate locations within the central nervous system, there are numerous sites in which information from the different sensory modalities converge. Perhaps the best known structure where inputs from different sensory modalities converge is the superior colliculus. Here, not only are visual, auditory and somatosensory inputs present, but they also form organized representations of auditory and visual space as well a map of the body surface in addition, neurons that receive inputs from more than one sensory modality (i.e. multisensory) are found here in abundance. The osteoclast, a multinucleated giant cell, has as its role the resorption of bone matrix with consequent release of calcium. The osteoclast is thought to derive from a pluripotent stem cell which also gives rise to monocytes and macrophages. The development, activation, and inhibition of osteoclasts are influenced by a number of systemic and local factors. The release of these factors is influenced by changes in the dietary status of the animal. Calcium deficiency is known to stimulate the recruitment and activity of large numbers of osteoclasts. To study disturbed muscle activation patterns in hemiparetic stroke, the EMG activity in spastic-paretic and contralateral elbow muscles of 10 stroke subjects were compared during graded directionally varying voluntary isometric contractions. We observed significant shifts in the peak EMG angle and increases in the angular range of EMG activity in the impaired upper limb. Furthermore, we observed stereotypic coactivation of many muscles acting at the same joint or at different joints across all subjects.
The effect of elbow angle changes on muscle coactivation patterns was studied by examining isometric torque-angle relations under passive and active conditions in 8 stroke subjects. …

A stable intracellular ionic environment is necessary for hepatocytes to function normally. Thus, during hypotonic shock or L-alanine uptake, hepatocytes swell and then exhibit a regulatory volume decrease (RVD), which comprises an increase in K$\sp+$ conductance (G$\sb{\rm K}$), an increased K$\sp+$ efflux, and a hyperpolarization of transmembrane potential (V$\sb{\rm m}$). Since hepatocyte intracellular Cl$\sp-$ has been demonstrated to distribute passively with V$\sb{\rm m}$, this study is designed to test the hypothesis that the hypotonic shock- or L-alanine uptake-induced hyperpolarization of V$\sb{\rm m}$ might provide an electromotive force for the efflux of hepatocyte intracellular Cl$\sp-$, which in turn would contribute osmotically …

Red Butte Canyon is a protected, near pristine canyon entering Salt Lake Valley, Utah. It contains a well-developed riparian zone and a perennial stream; hillside vegetation ranges from grasslands on the lower limits to Douglas-fir and aspen stands at the upper elevations. In this paper we describe the history of human impact, natural history aspects of climate, geology, and ecology, and faunal and floral information for key species in the canyon. This study examined the R value at VO2 max during maximal graded treadmill exercise tests to determine if it was a reliable indicator of VO2 max and could therefore be used as a test termination criterion. The subjects in this study were 160 coronary artery diseased (CAD) patients and 170 nondiseased individuals. Data were collected from existing stress test records. The R value was also analyzed to determine if proportions of diseased and nondiseased subjects were the same. The time it took 100 CAD patients to reach VO2 max and exhaustion once R rose above 1.00 was also examined.

Declining aircrew performance during periods of sustained flight operations (SUSOPs) has underscored the need to develop effective countermeasures. This paper reports on the ability of the central nervous system (CNS) stimulant d-methamphetamine to alleviate the detrimental effects of a simulated SUSOPs on subjective fatigue. Subjective fatigue was repeatedly measured by three questionnaires. The simulated SUSOP started at 1800 and consisted of a 9-h planning session followed by 4 h of rest and a 14-h mission. After 6 h of sleep, the 9/4/14 work/rest/work pattern was repeated. At 4 1/2 h into the second mission, 13 subjects were administered 10 mg …

The problem under investigation was to determine if strengthening the abductor and adductor muscle groups of the hip would increase hamstring flexion strength. Forty females from aerobic classes were put into two groups, control and treatment, after being pretested on the Cybex II Dynamometer. The control group participated only in aerobics, while the treatment group participated in the aerobics and treatment program. The treatment consisted of four exercises working abduction and adduction of the thigh with SPRI's Xercise Band.

Onset of puberty in male Djungarian hamsters is characterized by increased pituitary gonadotropin secretion and testes maturation, and is controlled by neurons within the brain which secrete gonadotropin-releasing hormone (GnRH). Puberty is associated with increased numbers of morphologically unipolar, but not bipolar, GnRH neurons in the medial preoptic area (MPOA) and diagonal band of Broca (DBB). To test the hypothesis that delayed sexual maturation arrests this increase, males were exposed to short days or administered melatonin. Males with delayed puberty had significantly fewer unipolar GnRH neuron numbers in the MPOA and DBB comparable to pubertal controls in long days. Unipolar …

We have adapted techniques developed for the extraction of DNA from formalin-fixed, paraffin-imbedded human tissues for use on preserved fish tissues. DNA was successfully extracted and the d-loop region of mitochondrial DNA was amplified with the polymerase chain reaction (PCR). The sequences of the amplified DNA from preserved and modern samples were identical. These techniques were also applied to fin tissue treated with a variety of preservatives. Extraction of DNA from ethyl alcohol and air-dried fin tissues gave yields equivalent to those from frozen tissues. The purpose of this research was to determine the effects of cryopreservation on the physical, biochemical and cellular aspects of heart valve tissue. The present study demonstrated that radiolabeled proline accumulation provides a quantitative estimate of total cellular metabolic viability. Radiolabeled inulin was demonstrated to be a useful marker for extracellular tissue space. Assessment of metabolic viability of the cellular population of leaflet tissue as a function of procurement and processing revealed that proline accumulation declined with increasing warm and cold ischemic times. Amphotericin B and streptomycin were toxic to the cells. Cefoxitin, lincomycin, polymyxin B, vancomycin and penicillin had …
