School of Graduate Studies Faculty Publications

Development of intranasal vaccines for HIV-1 and other mucosal pathogens has been hampered by the lack of adjuvants that can be given safely to humans. We have found that an intranasal Shigella vaccine (Invaplex) which is well tolerated in humans can also function as an adjuvant for intranasal protein and DNA vaccines in mice. To determine whether Invaplex could potentially adjuvant similar vaccines in humans, we simultaneously administered a simian immunodeficiency virus (SIV) envelope (Env) protein and DNA encoding simian-human immunodeficiency virus (SHIV) with or without Invaplex in the nasal cavity of female rhesus macaques. Animals were intranasally boosted with …

Trends In Hysterectomy In Hospital And Ambulatory Settings In Kentucky 2017-2020., Felicia A. Pugh

Electronic Theses and Dissertations

Hysterectomy is the most common treatment for benign uterine conditions in the United States. Data suggest that one in nine women will have the procedure in their lifetime (1). Previous studies indicate that abdominal hysterectomy rates for non-malignancies are declining as alternative methods are increasing for decades; however, those trends do not apply for all women in the country (2-5). In this cross-sectional study, hysterectomy procedures completed between 2017-2020 are described in Kentucky Cabinet for Health and Family Services data. Multivariable logistic regression is used to assess the odds of abdominal hysterectomy for benign indications in Kentucky. Conclusion: Rates of …

Development Of 3d-Bioprinted Scaffolds For Oral Probiotic Application., Jhanvi Patel

Electronic Theses and Dissertations

Background: Porphyromonas gingivalis adheres to and invades gingival epithelial cells, resulting in decreased cell viability. Previous studies have indicated that probiotics are effective against dental pathogens; however, few approaches provide sustained-delivery of active agents in the oral cavity. Hypothesis: Probiotics will limit P. gingivalis effects on Telomerase Immortalized Gingival Keratinocyte (TIGK) cells and 3D-printed scaffolds will prolong probiotic release. Methods: Lactobacillus acidophilus, Lactobacillus reuteri, and Bifidobacterium bifidum were assessed with adhesion and antibiotic protection assays to limit P. gingivalis effects on TIGKs. Scaffolds were printed with a select probiotic and evaluated for release kinetics. Results: Free L.a., …

Telehealth Assessment In The Elderly Using The 4m'S, Janet Obonyo Aug 2021

Telehealth Assessment In The Elderly Using The 4m'S, Janet Obonyo

Nursing Capstones

COVID-19 has accelerated an already urgent need for telehealth in the United States. While health care systems have been providing telehealth for years, the number of providers and patients who need to participate in telehealth has ballooned overnight. Though telehealth has been in use for a while, the geriatric population seems to be disadvantaged, especially those who reside in rural settings. Due to increased COVID 19 cases among the elderly population in North Dakota, the Geriatric Workforce Enhancement Program (GWEP) allocated a grant to the University of North Dakota to promote telehealth in the elderly receiving homecare services in rural …

Predictors Of Aggression Among Adolescent Girls, Sarah Joy Park Aug 2021

Predictors Of Aggression Among Adolescent Girls, Sarah Joy Park

Loma Linda University Electronic Theses, Dissertations & Projects

Women and girls will often deny what they feel and think to maintain their relationships. Silencing the self can be detrimental to the physical and mental health of teenage girls. This stifling of the self is related to depression, earlier first intercourse, eating disorders, and less relationship satisfaction. Girls who silence their voices are also likely to have difficulty expressing themselves when they feel angry. In this study we explored the relationship between silencing the self and physical, verbal, and indirect aggression. Our sample consisted of 88 teenage girls from two high schools, with a mean age of 16. They …

Epithelial Memory Of Resolved Inflammation Limits Tissue Damage While Promoting Pancreatic Tumorigenesis, I-Lin Ho Aug 2021

Epithelial Memory Of Resolved Inflammation Limits Tissue Damage While Promoting Pancreatic Tumorigenesis, I-Lin Ho

Dissertations and Theses (Open Access)

Inflammation is a major risk factor for pancreatic ductal adenocarcinoma. When occurring in the context of pancreatitis, mutations of KRAS accelerate tumor development. We discovered that long after its complete resolution, a transient inflammatory event primes pancreatic epithelial cells to subsequent transformation by oncogenic KRAS. Upon recovery from acute inflammation, epithelial cells of the pancreas display an enduring adaptive response associated with sustained transcriptional and epigenetic reprogramming. Such adaptation enables the prompt reactivation of acinar-to-ductal metaplasia (ADM) upon subsequent inflammatory events, thus efficiently limiting tissue damage via rapid decrease of zymogen production. We propose that since activating mutations of KRAS …

Manipulating Enhancer Rna Activity To Study Their Roles In Gene Regulation And Cancer, Zian Liao Aug 2021

Manipulating Enhancer Rna Activity To Study Their Roles In Gene Regulation And Cancer, Zian Liao

Dissertations and Theses (Open Access)

Enhancers are essential regulatory elements that precisely control gene transcription during development and disease. Especially in cancer, enhancers modulate tumor gene expression, and in some cases directly drive tumorigenesis. With the recent advances in next-generation sequencing, researchers discovered that active enhancers are pervasively transcribed into RNAs, which are named enhancer RNAs (eRNAs). Studies have suggested that eRNAs have a functional role in the enhancer action mechanism. Studying their roles will provide a new understanding of gene regulation and cancer development. In my MS study, I focused on using novel methods to study eRNA functions. Using a breast-cancer-specific eRNA, I have …

Effect Of Lactoferrin To Increase Drug Permeability Of Primary Pulmonary Mycobacterial Granulomas, Thao Nguyen, Thao Kt Nguyen Aug 2021

Effect Of Lactoferrin To Increase Drug Permeability Of Primary Pulmonary Mycobacterial Granulomas, Thao Nguyen, Thao Kt Nguyen

Dissertations and Theses (Open Access)

Despite extensive research and worldwide eradication efforts, Mycobacterium tuberculosis (Mtb) remains a major infectious pathogen to the human population with about 10 million cases of infection per year globally. The host-pathogen interaction, pulmonary granuloma formation, and Mtb adaptions result in increased complexity of the disease. Granulomas are formed by active immune responses generated during Mtb infection, and serve to contain and limit bacterial dissemination. The major mycobacterial surface mycolic acid, trehalose 6,6'-dimycolate (TDM), functions in multiple ways to enhance immune cell recruitment of sites of infection, to induce inflammation and granulomatous responses, and to initiate survival strategies for …

Abdominal Vascular Injuries- What General/ Trauma Surgeons Should Know?, Zia Ur Rehman Aug 2021

Abdominal Vascular Injuries- What General/ Trauma Surgeons Should Know?, Zia Ur Rehman

Department of Surgery Docs

Abdominal vascular injuries are the common cause of death post-trauma. These are challenging injuries to manage due to severe haemodynamic instability, other associated injuries and difficulty in accessing and controlling these deep-seated vessels. Early control of bleeding can decrease mortality in these patients. Abdominal vasculature is divided into three zones, each requiring different operative strategy for exposure. Standard vascular surgery principles of achieving proximal and distal control before exploring any haematoma are followed when managing these injuries. Resuscitative endovascular balloon occlusion of aorta is a minimally invasive method of achieving proximal aortic occlusion. This also acts as bridge for definitive …
