Sex Differences In Peripheral Fat Detection And Metabolism, Naima Dahir
Sex Differences In Peripheral Fat Detection And Metabolism, Naima Dahir
Electronic Theses and Dissertations, 2020-2023
The current epidemic of obesity is driven, at least in part, by excessive caloric intake and fat intake has been implicated as a major contributing factor. The mammalian taste system displays marked plasticity, and it has been shown that the detection of fatty acids, the prototypical fat taste stimulus, leads to reduced sensitivity upon prolonged exposure to a high-fat diet. Increased obesity and metabolic disorders in postmenopausal females indicate that the female steroid hormone, estrogen, may contribute to metabolic homeostasis. Sex differences in the taste system were explored using a multidisciplinary approach to examine the ways in which estrogen influences …
Anti-Map Triple Therapy Supports Immunomodulatory Therapeutic Response In Crohn's Disease Through Downregulation Of Nf-Kb Activation In The Absence Of Map Detection, Erij Elkamel
Electronic Theses and Dissertations, 2020-2023
The triple antibiotic formulation, known as anti-MAP therapy, exhibits unique synergistic antimicrobial activity and should be effective for treatment of Crohn's disease (CD) associated with Mycobacterium avium subspecies paratuberculosis (MAP). The absence of MAP detection in some CD cases may be linked to poor diagnostics or lack of association with the disease. To understand the therapeutic response of some CD patients to anti-MAP therapy in absence of MAP detection, the immunomodulatory potency of anti-MAP therapy and its major ingredients, clarithromycin (CLA) and rifabutin (RIF), in THP-1, Caco-2, and Jurkat T-cells were investigated. Anti-MAP formulation at 2.0 µg/mL decreased MAP viability …
Knowledge And Attitudes Of Nurse Practitioner's Assessment Of Adults With Signs And Symptoms Of Posttraumatic Stress Disorder, Kelli Lipscomb
Knowledge And Attitudes Of Nurse Practitioner's Assessment Of Adults With Signs And Symptoms Of Posttraumatic Stress Disorder, Kelli Lipscomb
Electronic Theses and Dissertations, 2020-2023
As the numbers of primary care physicians decline, nurse practitioners (NPs) are filling this gap as primary care providers in many settings. The number of adults presenting to their primary care provider with signs and symptoms of posttraumatic stress disorder (PTSD) each year is unknown. The devastating effects of untreated PTSD include high rates of homelessness, violence, drug abuse, suicidality and a decreased quality of life. Little is known as to the breadth of NPs knowledge of PTSD sequelae, epidemiology and screening protocols. Increased perceived self-efficacy, tied to positive attitudes about PTSD knowledge and screening, result in overall higher rates …
Reassessing The Transcriptional Regulation Of Protective Cd4 T Cell Responses Against Influenza A Virus, Kunal Dhume
Reassessing The Transcriptional Regulation Of Protective Cd4 T Cell Responses Against Influenza A Virus, Kunal Dhume
Electronic Theses and Dissertations, 2020-2023
An attractive strategy to improve efficacy of current Influenza A Virus (IAV) vaccines is to promote protective CD4 T cell responses. Antiviral CD4 T cell responses are predominantly classified as strong IFN-gamma producing T helper type 1 (Th1) cells. Yet, the role of IFN-gamma in protection against IAV is still unclear, suggesting absolute Th1 polarization may be expendable for effective IAV immunity. Here we test this hypothesis using models that restrict the deficiency of known transcriptional regulators of Th1 immunity in only CD4 T cells, avoiding indirect impact on other immune cell responses. We find the 'master regulator' of Th1 …

Development And Evaluation Of Wound Healing Property Of In-Situ Hydrogel Formulation For Topical Delivery Of Short-Chain Hyaluronic Acid In Dental Use, Porniweat Rosaming
Chulalongkorn University Theses and Dissertations (Chula ETD)
Regeneration of damaged periodontal tissues becomes one of the biggest challenges that is hard to overcome, even without the presence of any pathogens. Tissue engineering was proposed as a possible technique for the lost tissues’ restoration. In this study, methacrylated hyaluronic acid (MeHA) hydrogel was developed with different concentrations of low molecular weight (short-chain) hyaluronic acid to achieve the hydrogels with suitable physical properties and biological properties for dental tissue regeneration. The results revealed that short-chain hyaluronic acid (sHA) could interfere with some behaviors of networking systems; however, overall properties (including gelation time, mechanical properties, swelling and degradation profiles, and …

Protecting Life And Lung: Protected Areas Affect Fine Particulate Matter And Respiratory Hospitalizations In The Brazilian Amazon Biome., Derek Michael Sheehan
Protecting Life And Lung: Protected Areas Affect Fine Particulate Matter And Respiratory Hospitalizations In The Brazilian Amazon Biome., Derek Michael Sheehan
Graduate Student Theses, Dissertations, & Professional Papers
I assessed the impacts of upwind protected area coverage on local respiratory health within the Brazilian Amazon. A hypothesized mechanism is the legal prohibition of human ignited fires within protected areas, reducing particulate matter pollution, impacting respiratory health downwind. The connection between fires and respiratory diseases in the Amazon is well established (Smith et al. 2014; Rangel and Vogl 2019; Rocha and Sant’anna 2020). What is not well understood is the potential that government policies aimed at preventing ecosystem loss may also promote health and wellbeing, combining the UN sustainable development goals 3 and 15. Protected areas currently dominate government …

College Students' Mental Health And Drug Use Outcomes During The Stay-At-Home Order, Alexander T. Hain
Cal Poly Humboldt theses and projects
The COVID-19 pandemic has affected people of all ages causing increased drug usage and worsening mental health in addition to hospitalization and death. The current study investigated how the United States’ stay-at-home orders affected the mental health and drug use of young adult college students. This population is of particular interest because young adults are at most risk of drug use developing into addiction. Two hypotheses related to the self-medication hypothesis were investigated: (1) there will be a significant increase in drug use during the stay-at-home order when compared to drug use prior to the stay-at-home order and (2) feelings …
Effects Of A Dual-Task Paradigm On Tandem Gait Performance, Courtney Angela Perry
Effects Of A Dual-Task Paradigm On Tandem Gait Performance, Courtney Angela Perry
Cal Poly Humboldt theses and projects
In concussion management, a variety of cognitive and motor control tests, like the tandem gait, have been used for assessing the presence of a concussion injury at the sideline and through recovery. More recently, research suggests that introducing a secondary cognitive retention task during tandem gait (i.e., dual task) may provide a better assessment of concussion. The purpose of this study was to determine if a tandem gait test paired with a 6-digit retention is a valid and reliable tool. Participants completed three trials of both the single-task (ST) and dual-task (DT) tandem gait tests, and their average (MEAN) and …

Muscle-invasive bladder cancer (MIBC) is a fast-growing and aggressive malignant tumor in urinary system. Since chemotherapy and immunotherapy are only useable with a few MIBC patients, the clinical treatment of MIBC still faces challenges. Here, we examined the feasibility of plasma-activated saline (PAS) as a fledgling therapeutic strategy for MIBC treatment. Our data showed that plasma irradiation could generate a variety of reactive oxygen species (ROS) and reactive nitrogen species (RNS) in saline. In vivo tests revealed that pericarcinomatous tissue injection with PAS was effective at preventing subcutaneous bladder tumor growth, with no side effects to the visceral organs after …

The Gram-positive anaerobic bacterium Cutibacterium acnes (C. acnes) is a commensal of the human skin, but also an opportunistic pathogen that contributes to the pathophysiology of the skin disease acne vulgaris. C. acnes can form biofilms; cells in biofilms are more resilient to antimicrobial stresses. Acne therapeutic options such as topical or systemic antimicrobial treatments often show incomplete responses. In this study we measured the efficacy of nanosecond pulsed electric fields (nsPEF), a new promising cell and tissue ablation technology, to inactivate C. acnes. Our results show that all tested nsPEF doses (250 to 2000 pulses, 280 ns pulses, …

Collagen microfiber-based constructs have garnered considerable attention for ligament, tendon, and other soft tissue repairs, yet with limited clinical translation due to strength, biocompatibility, scalable manufacturing, and other challenges. Crosslinking collagen fibers improves mechanical properties; however, questions remain regarding optimal crosslinking chemistries, biocompatibility, biodegradation, long-term stability, and potential for biotextile assemble at scale, limiting their clinical usefulness. Here, we assessed over 50 different crosslinking chemistries on microfluidic wet-extruded collagen microfibers made with clinically relevant collagen to optimize collagen fibers as a biotextile yarn for suture or other medical device manufacture. The endogenous collagen crosslinker, glyoxal, provides extraordinary fiber ultimate tensile …

Damage from myocardial infarction (MI) and subsequent heart failure are serious public health concerns. Current clinical treatments and therapies to treat MI damage largely do not address the regeneration of cardiomyocytes. In a previous study, we established that it is possible to promote regeneration of cardiac muscle with vascular endothelial growth factor B gene delivery directly to the ischemic myocardium. In the current study we aim to optimize cardioporation parameters to increase expression efficiency by varying electrode configuration, applied voltage, pulse length, and plasmid vector size. By using a surface monopolar electrode, optimized pulsing conditions and reducing vector size, we …

Exposure to hydrochloric acid (HCl) leads acutely to asthma-like symptoms, acute respiratory distress syndrome (ARDS), including compromised alveolo-capillary barrier, and respiratory failure. To better understand the direct effects of HCl on pulmonary endothelial function, we studied the characteristics of HCl-induced endothelial barrier dysfunction in primary cultures of human lung microvascular endothelial cells (HLMVEC), defined the involved molecular pathways, and tested the potentially beneficial effects of Heat Shock Protein 90 (HSP90) inhibitors. HCl impaired barrier function in a time- and concentration-dependent manner and was associated with activation of Protein Kinase B (AKT), Ras homolog family member A (RhoA) and myosin light …

We previously reported that female mice exhibit protection against chemically induced pulmonary fibrosis and suggested a potential role of estrogen. Phytoestrogens act, at least in part, via stimulation of estrogen receptors; furthermore, compared to residents of Western countries, residents of East Asian countries consume higher amounts of phytoestrogens and exhibit lower rates of pulmonary fibrosis. Therefore, we tested the hypothesis that dietary phytoestrogens ameliorate the severity of experimentally induced pulmonary fibrosis. Male mice placed on either regular soybean diet or phytoestrogen-free diet were instilled with 0.1 N HCl to provoke pulmonary fibrosis. Thirty days later, lung mechanics were measured as …

Gene-based approaches for protein replacement therapies have the potential to reduce the number of administrations. Our previous work demonstrated that expression could be enhanced and/or the applied voltage reduced by preheating the tissue prior to pulse administration. In the current study, we utilized our 16-pin multi-electrode array (MEA) and incorporated nine optical fibers, connected to an infrared laser, between each set of four electrodes to heat the tissue to 43 °C. For proof of principle, a guinea pig model was used to test delivery of reporter genes. We observed that when the skin was preheated, it was possible to achieve …

An estimated 350 million people are living with chronic Hepatitis B virus (HBV) worldwide. Preventative HBV vaccination in infants has reduced the disease burden; however, insufficient immunization programs and access obstacles leave vulnerable populations at risk for infection in endemic regions. Gene electrotransfer (GET) using a noninvasive multielectrode array (MEA) provides an alternative platform for DNA vaccination in the skin. DNA vaccines are nonlive and nonreplicating and temperature stable unlike their counterparts. In addition, their simple engineering allows them to be manufactured quickly at a low cost. In the current work, we present the combination of GET and moderate heating …

The ability to directly observe membrane potential charging dynamics across a full microscopic field of view is vital for understanding interactions between a biological system and a given electrical stimulus. Accurate empirical knowledge of cell membrane electrodynamics will enable validation of fundamental hypotheses posited by the single shell model, which includes the degree of voltage change across a membrane and cellular sensitivity to external electric field non-uniformity and directionality. To this end, we have developed a high-speed strobe microscopy system with a time resolution of ~ 6 ns that allows us to acquire time-sequential data for temporally repeatable events (non-injurious …

Cold atmospheric plasma, including plasma jet and surface plasma, can promote the apoptosis of cancer cells without causing significant damage to surrounding normal cells, which was hopeful to be applied to the clinical cancer therapy. However, experimental plasma devices used directly to clinical experiments has challenges in technology and methods, especially the difference in killing tumor cells efficiency of these two common plasma sources. Therefore, it is great necessity to explore the differences in treating tumors between different plasma sources. This paper achieved good killing efficiency by using two kinds of cold atmospheric plasma generating devices, namely plasma jet and …
