Excess Extracellular K+ Causes Inner Hair Cell Ribbon Synapse Degeneration, Hong-Bo Zhao, Yan Zhu, Li-Man Liu Jan 2021

Excess Extracellular K+ Causes Inner Hair Cell Ribbon Synapse Degeneration, Hong-Bo Zhao, Yan Zhu, Li-Man Liu

Otolaryngology--Head & Neck Surgery Faculty Publications

Inner hair cell (IHC) ribbon synapses are the first synapse in the auditory system and can be degenerated by noise and aging, thereby leading to hidden hearing loss (HHL) and other hearing disorders. However, the mechanism underlying this cochlear synaptopathy remains unclear. Here, we report that elevation of extracellular K+, which is a consequence of noise exposure, could cause IHC ribbon synapse degeneration and swelling. Like intensity dependence in noise-induced cochlear synaptopathy, the K+-induced degeneration was dose-dependent, and could be attenuated by BK channel blockers. However, application of glutamate receptor (GluR) agonists caused ribbon swelling but …

Benefits Of Phenolic Compounds Isolated From Olive Oil On Prevention Of Cancer, Aicha Hatimy, Nadia El Kadmiri

Health Sciences

The traditional Mediterranean Diet has been long known to have many health benefits. The diet is associated with a low percentage of certain types of cancers such as colorectal cancer, breast cancer, gastric cancer, and prostate cancer. The main source of fats in the Mediterranean diet is olive oil. In fact, the beneficial health effects of olive oil were attributed to its phenolic compounds like phenolic alcohols, hydroxytyrosol (3,4-dihydroxy phenyl ethanol), and their secoiridoid derivatives (oleocanthal). Several studies have demonstrated that hydroxytyrosol and oleocanthal are able to inhibit proliferation and induce apoptosis in different tumor cell lines. The aim of …

Development Of A Novel Orthotopic Gastric Cancer Mouse Model., Wonyoung Kang, Leigh Maher, Michael Michaud, Seong-Woo Bae, Seongyeong Kim, Hye Seung Lee, Seock-Ah Im, Han-Kwang Yang, Charles Lee

Faculty Research 2021

BACKGROUND: Gastric cancer metastasis is a highly fatal disease with a five-year survival rate of less than 5%. One major obstacle in studying gastric cancer metastasis is the lack of faithful models available. The cancer xenograft mouse models are widely used to elucidate the mechanisms of cancer development and progression. Current procedures for creating cancer xenografts include both heterotopic (i.e., subcutaneous) and orthotopic transplantation methods. Compared to the heterotopic model, the orthotopic model has been shown to be the more clinically relevant design as it enables the development of cancer metastasis. Although there are several methods in use to develop …

A Randomized Trial Of Manual Phone Calls Versus Automated Text Messages For Peripheral Nerve Block Follow-Ups., Gavyn Ooi, Eric S. Schwenk, Marc C. Torjman, Kent Berg

Department of Anesthesiology Faculty Papers

Mobile phone applications (apps) have been used for patient follow-up in the postoperative period, specifically to assess for complications and patient satisfaction. Few studies have evaluated their use in regional anesthesia. The objective of this study was to compare follow-up response rates using manual phone calls versus an automated patient outreach (APO) app for peripheral nerve block patients. We hypothesized that the response rate would be higher in the APO group. A mobile app, "JeffAnesthesia," was developed, which sends notifications to patients to answer survey questions in the app. We randomly assigned patients who received peripheral nerve blocks for postoperative …

Management Of The Sensitized Cardiac Transplantation Recipient, Michael Mazzei, Suresh Keshavamurthy, Olga Timofeeva, Yoshiya Toyoda

Surgery Faculty Publications

Preoperative sensitization of the cardiac transplant recipient, defined as the presence of anti-Human Leukocyte Antigen (HLA) antibodies before transplant, represents a significant management challenge for physicians. Sensitization prolongs the pre-transplant wait time and is associated with postoperative transplant complications and death. It is critical that sensitized heart transplant candidates be identified and optimized before surgery. In this review, we describe the risk for sensitization, discuss the means through which sensitization may be diagnosed, and highlight some of the new therapeutic options for managing the sensitized cardiac transplant patients.

Protein Kinase D Promotes Airway Epithelial Barrier Dysfunction And Permeability Through Down-Regulation Of Claudin-1, Huachen Gan, Guibo Wang, Qin Hao, Q. Jane Wang, Hua Tang Jan 2021

Protein Kinase D Promotes Airway Epithelial Barrier Dysfunction And Permeability Through Down-Regulation Of Claudin-1, Huachen Gan, Guibo Wang, Qin Hao, Q. Jane Wang, Hua Tang

Cellular and Molecular Biology Faculty Publications and Presentations

At the interface between host and external environment, the airway epithelium serves as a major protective barrier. In the present study we show that protein kinase D (PKD) plays an important role in the formation and integrity of the airway epithelial barrier. Either inhibition of PKD activity or silencing of PKD increased transepithelial electrical resistance (TEER), resulting in a tighter epithelial barrier. Among the three PKD isoforms, PKD3 knockdown was the most efficient one to increase TEER in polarized airway epithelial monolayers. In contrast, overexpression of PKD3 wild type, but not PKD3 kinase-inactive mutant, disrupted the formation of apical intercellular …

Background: To date, there are only a few case reports of cyclophosphamide (Cy)-induced hemorrhagic cystitis (HC) in adult or pediatric allogeneic stem cell transplant (SCT) patients treated successfully with hyperbaric oxygen (HBO). In all the reported cases, Cy was used as a part of the conditioning regimen, rather than post-transplant for graft-versus-host-disease (GVHD) prophylaxis. More recently, the risk of HC in allogeneic SCT is further increased by the widespread use of post-transplantation cyclophosphamide (PTCy) as a highly effective strategy for GVHD prophylaxis. This is the first case reported of PTCy-induced HC successfully treated with HBO to the best of our …

Heme Oxygenase-1 Affects Cytochrome P450 Function Through The Formation Of Heteromeric Complexes: Interactions Between Cyp1a2 And Heme Oxygenase-1, J. Patrick Connick, James R. Reed, George F. Cawley, Wayne L. Backes

School of Medicine Faculty Publications

Heme oxygenase 1 (HO-1) and the cytochromes P450 (P450s) are endoplasmic reticulum–bound enzymes that rely on the same protein, NADPH-cytochrome P450 reductase (POR), to provide the electrons necessary for substrate metabolism. Although the HO-1 and P450 systems are interconnected owing to their common electron donor, they generally have been studied separately. As the expressions of both HO-1 and P450s are affected by xenobiotic exposure, changes in HO-1 expression can potentially affect P450 function and, conversely, changes in P450 expression can influence HO-1. The goal of this study was to examine interactions between the P450 and HO-1 systems. Using bioluminescence resonance …

Old And Cold: A Novel Case Of Combined Secobarbital And Pentobarbital Poisoning In An Elderly Woman., Kenneth D. Katz Md, Andrew L. Koons, Gregory A. Makar Do, Amy Wier Do

Department of Emergency Medicine

A 94-year-old woman was found unresponsive in her room at an independent living facility. Upon paramedic arrival, the patient had a Glasgow Coma Scale of 3, and she was transported to the emergency department (ED). In the ED, she was unresponsive but spontaneously breathing, bradycardia, and hypothermia. Serum concentrations of both secobarbital (3.3µg/mL; therapeutic 1.0-2.0µg/mL) and pentobarbital (9.5µg/mL; therapeutic < 5.0µg/mL) were detected and elevated. This type of poisoning is quite rare and should be considered in patients presenting with hypothermia and coma, even in patients showing brain death signs. The use of hemodialysis for refractory pentobarbital poisoning may be helpful.
