Factors That Influence Response Classifications In Chemotherapy Treated Patient-Derived Xenografts (Pdx)., Joan Malcolm, Timothy M Stearns, Susan Airhart, Joel H. Graber, Carol J Bult
Faculty Research 2019
In this study, we investigated the impact of initial tumor volume, rate of tumor growth, cohort size, study duration, and data analysis method on chemotherapy treatment response classifications in patient-derived xenografts (PDXs). The analyses were conducted on cisplatin treatment response data for 70 PDX models representing ten cancer types with up to 28-day study duration and cohort sizes of 3-10 tumor-bearing mice. The results demonstrated that a 21-day dosing study using a cohort size of eight was necessary to reliably detect responsive models (i.e., tumor volume ratio of treated animals to control between 0.1 and 0.42)-independent of analysis method. A …

Recommendations For Regulating The Environmental Risk Of Shedding For Gene Therapy And Oncolytic Viruses In Canada, Tania M. Bubela, Ron Boch, Sowmya Viswanathan
Office of the Provost
Canadian academic and industry stakeholders are concerned about the inclusion of "virus-like particles or sub-viral particles" in the definition of New Substances Notification Regulations for Organisms (NSNR(O)) which impacts clinical cell and gene therapy and commercialization. The requirement of an independent 120 days Environment and Climate Change Canada (ECCC) review preceding a Health Canada review on quality and environmental concerns places an additional burden on Sponsors submitting clinical trial applications (CTA) and/or New Drug Submissions (NDS). A workshop initiated by CellCAN and BIOTECanada with participants from Environment and Climate Change Canada, Health Canada, the Public Health Agency of Canada and …

The Effects Of Bacterial Endotoxin Lps On Synaptic Transmission At The Neuromuscular Junction, Robin L. Cooper, Micaiah Mcnabb, Jeremy Nadolski
Biology Faculty Publications
The direct action of bacterial lipopolysaccharides (LPS) endotoxin was shown to enhance synaptic transmission and hyperpolarize the membrane potential at low doses, but block glutamatergic receptors and decrease observable spontaneous events at a high dosage. The dosage effects are LPS type specific. The hyperpolarization is not due to voltage-gated potassium channels or to activation of nitric oxide synthase (NOS). The effects are induced directly by LPS, independent of an immune response.

Previous studies have shown that irrational beliefs contribute significantly to the procrastination, including the thesis-writing procrastination. Unfortunately, there are no scales that specifically can measure the beliefs in the thesis writing context. Therefore, this study aimed to develop and validate a scale called the Skala Keyakinan Irasional dalam Penyusunan Skripsi (SKI-S). The scale was developed based on irrational belief construct proposed by Albert Ellis in the context of thesis writing. To achieve the aim, the study applied a research and development design involving four steps: (1) defining the construct; (2) developing the measurement model; (3) carrying out confirmatory factor analysis; …

Respiratory Infections And Risk For Development Of Narcolepsy: Analysis Of The Truven Health Marketscan Database (2008 To 2010) With Additional Assessment Of Incidence And Prevalence, Darren Scheer
USF Tampa Graduate Theses and Dissertations
Background and Significance: Narcolepsy is a chronic neurological disorder. These patients experience various psychiatric and physical comorbid diseases and mortality at an increased rate compared to the general population. Additionally, patients with narcolepsy experience approximately a doubling of various annual healthcare related facility visits, transactions, and costs comparatively. Narcolepsy with cataplexy is generally believed to be more prevalent than narcolepsy without cataplexy. However, incidence and prevalence estimates of narcolepsy (with or without cataplexy) vary widely with few large epidemiological studies conducted worldwide and none in the U.S evaluating these proportions in both children and adults utilizing a large health care …
Does The Theory Of Reasoned Action Inform The Willingness Of Individuals Undergoing Genetic Testing To Seek Disclosure Of Incidental Findings Related To The Risk For Alzheimer Disease?, Alisa M. Forrest
Theses & Dissertations
Background: In recent years, researchers have increasingly employed genetic testing as a means for understanding and treating diseases like Alzheimer disease, a common progressive disease affecting cognition and behavior. Genetic studies hold the potential for major breakthroughs in treatment of diseases like Alzheimer disease. However, with the increase in the use of genomewide association, microarray, and whole genome sequencing comes the potential for a greater number of incidental findings in genetic research—findings not central to the aim of a study but nonetheless informative about a participant’s health. Although many studies have documented the ethical implications around disclosure of such findings …
Genomics And Transcriptomics Approaches To Understanding Drug Resistance Mechanisms In The Malaria Parasite Plasmodium Falciparum, Justin Allan Gibbons
Genomics And Transcriptomics Approaches To Understanding Drug Resistance Mechanisms In The Malaria Parasite Plasmodium Falciparum, Justin Allan Gibbons
USF Tampa Graduate Theses and Dissertations
The malaria parasite Plasmodium falciparum is responsible for about 500,000 deaths a year and is evolving resistance to the front-line treatment of artemisinin-based combination therapy. Resistance is currently confined to South East Asia, however millions of lives will be at risk if resistance spreads to Africa. Understanding the mechanism of resistance to artemisinins would aid containment strategies to prevent the spread of artemisinin resistance. There is also an urgent need to accelerate drug discovery since drug resistance has already been documented to all existing antimalarials. Here, I report on our efforts to understand the function of the gene k13, the …
Blood-Based Molecular Biomarkers For Alzheimer's Disease, Henrik Zetterberg, Samantha C. Burnham
Blood-Based Molecular Biomarkers For Alzheimer's Disease, Henrik Zetterberg, Samantha C. Burnham
Research outputs 2014 to 2021
A major barrier to the effective conduct of clinical trials of new drug candidates against Alzheimer's disease (AD) and to identifying patients for receiving future disease-modifying treatments is the limited capacity of the current health system to find and diagnose patients with early AD pathology. This may be related in part to the limited capacity of the current health systems to select those people likely to have AD pathology in order to confirm the diagnosis with available cerebrospinal fluid and imaging biomarkers at memory clinics. In the current narrative review, we summarize the literature on candidate blood tests for AD …
Forgotten Children & Bonds That Heal: The Critical Nature Of Attachment, August Ashbaker
Forgotten Children & Bonds That Heal: The Critical Nature Of Attachment, August Ashbaker
Lesley University Community of Scholars Day
Human attachment is one of the most critical factors in proper neurological and physiological development. Attachment is built through the mutuality of emotional impact, as well as the regular reception of physical touch. Infants and children who have been deprived of these vital necessities through the lacking initiatives of their caregivers—through neglect—display visible signs of its physiological, cognitive, emotional, and behavioral impact. Research has found neglect to be the highest risk factor for permanent damage to brain development in comparison to other forms of complex trauma (i.e. chronic caregiver interpersonal trauma in childhood); it is also the most prevalent …

Aberrations in brain microcirculation and the associated increase in blood-brain-barrier (BBB) permeability in addition to neuroinflammation and Aβ deposition observed in Alzheimer’s disease (AD) and ischemia have gained considerable attention recently. However, the role of microvascular homeostasis as a pathogenic substrate to disturbed microperfusion as well as an overlapping etiologic mechanism between AD and ischemia has not been thoroughly explored. In this study, we employ temporal histopathology of cerebral vasculature in a rat model of β-amyloid (Aβ) toxicity and endothelin-1 induced-ischemia (ET1) to investigate the panorama of cerebral pathology and the protein expression on d1, d7, and d28 post-injury. The …
