Neurosurgery and Brain Repair Faculty Publications

Blood-spinal cord barrier (BSCB) alterations, including capillary rupture, have been demonstrated in animal models of amyotrophic lateral sclerosis (ALS) and ALS patients. To date, treatment to restore BSCB in ALS is underexplored. Here, we evaluated whether intravenous transplantation of human bone marrow CD34+ (hBM34+) cells into symptomatic ALS mice leads to restoration of capillary integrity in the spinal cord as determined by detection of microhemorrhages. Three different doses of hBM34+ cells (5 × 104, 5 × 105 or 1 × 106) or media were intravenously injected into symptomatic G93A SOD1 mice …

Human Umbilical Cord Blood Serum–Derived Α-Secretase: Functional Testing In Alzheimer’S Disease Mouse Models, Ahsan Habib, Huayan Hou, Takashi Mori, Jun Tian, Jin Zeng, Shengnuo Fan, Brian Giunta, Paul R. Sanberg, Darrell Sawmiller, Jun Tan

Neurosurgery and Brain Repair Faculty Publications

Alzheimer’s disease (AD) is an age-related disorder that affects cognition. Our previous studies showed that the neuroprotective fragment of amyloid procurer protein (APP) metabolite, soluble APPα (sAPPα), interferes with β-site APP-cleaving enzyme 1 (BACE1, β-secretase) cleavage and reduces amyloid-β (Aβ) generation. In an attempt to identify approaches to restore sAPPα levels, we found that human cord blood serum (CBS) significantly promotes sAPPα production compared with adult blood serum (ABS) and aged blood serum (AgBS) in Chinese hamster ovary cells stably expressing wild-type human APP. Interestingly, CBS selectively mediated the α-secretase cleavage of human neuron-specific recombinant APP695 in a cell-free …

Impact Of Pre-And Postpartum Nutrition On March-Calving Cow And Progeny Productivity, Jacki A. Musgrave, Devin L. Broadhead, L. A. Stalker Stalker, Richard N. Funston

Nebraska Beef Cattle Reports

March-calving cows were fed hay or grazed corn residue prepartum, and then either fed hay or grazed subirrigated meadow postpartum. Combinations of these different feeding strategies impacted body weight (BW) and body condition score (BCS) during the pre-and postpartum period; however, resulted in similar pregnancy rates. Although differences were not observed in cow pregnancy rates, a benefit in pre-weaning growth was observed for calves from the dams in postpartum meadow treatment. A tendency for an improvement in marbling score was observed for steers born to cows fed hay prepartum, perhaps indicating a higher plane of nutrition prepartum may improve quality …

Orbital Skull Base Reconstruction With Temporalis Muscle: The Sphenoid Keyhole Technique, Mofiyinfolu Sokoya, Arash Bahrami, Jason Cohn, Sameep Kadakia, Moustafa Mourad, Yadranko Ducic

Otolaryngology (ENT) Resident Research

The temporalis flap is a common flap used in head and neck reconstruction. In this article, the authors present a new modification of the use of the temporalis muscle flap for orbital skull base reconstruction with the use of a sphenoid keyhole. This technique confers multiple advantages in reducing the arc of rotation and providing a tension-free reconstruction of the orbital skull base.

Students' Understanding About Clinical Research And Their Willingness To Participate In Clinical Trials, Shashikiran Pathakamudi Jan 2018

Students' Understanding About Clinical Research And Their Willingness To Participate In Clinical Trials, Shashikiran Pathakamudi

Master's Theses and Doctoral Dissertations

Adolescents and young adults remain underrepresented in clinical trials. Knowledge and perceptions related to clinical research usually determine willingness to participate. During the spring of 2016, an in-class paper survey was conducted to determine whether providing a brief introduction to clinical research would affect the willingness of undergraduate students from the College of Arts and Sciences and the College of Technology at Eastern Michigan University to participate in clinical trials. Some participants received information on clinical research through a short paragraph, and some received no new information, which differentiated the “active” group from the “placebo” group. Overall, both groups provided …

The themes identified within the data were SLP education for patient and family, respect for patient decisions, SLP understanding of barriers to patient adherence to dysphagia recommendations, family buy-in and practical support, perpetuation of …

Functional Characterization Of Scaffold Protein Shoc2, Hyein Jang Jan 2018

Functional Characterization Of Scaffold Protein Shoc2, Hyein Jang

Theses and Dissertations--Molecular and Cellular Biochemistry

Signaling scaffolds are critical for the correct spatial organization of enzymes within the ERK1/2 signaling pathway and proper transmission of intracellular information. However, mechanisms that control molecular dynamics within scaffolding complexes, as well as biological activities regulated by the specific assemblies, remain unclear.

The scaffold protein Shoc2 is critical for transmission of the ERK1/2 pathway signals. Shoc2 accelerates ERK1/2 signaling by integrating Ras and RAF-1 enzymes into a multi-protein complex. Germ-line mutations in shoc2 cause Noonan-like RASopathy, a disorder with a wide spectrum of developmental deficiencies. However, the physiological role of Shoc2, the nature of ERK1/2 signals transduced through this …

Impact Of Rapid Meca Polymerase Chain Reaction Rapid Diagnostic Testing For Staphylococcus Aureus In A Pediatric Setting, Emily Drwiega

Undergraduate Honors Thesis Collection

Rapid molecular technology can detect the mecA resistance gene in Staphylococcus aureus (SA), predicting methicillin susceptibility in under one hour. In combination with antimicrobial stewardship program interventions in adults with SA bacteremia, rapid mecA testing decreases time to targeted therapy. This intervention has not yet been shown effective in pediatric patients or in the absence of real-time stewardship interventions. The objective of this study was to determine if time to optimal therapy decreased following implementation of GeneXpert rapid diagnostic testing (RDT) in a pediatric institution without a formal antimicrobial stewardship protocol for response. The primary outcome was time to optimal …

Objectives: To match responsive neurostimulator (RNS) and polysomnographic data to determine if RNS detections and stimulations correlate with measurements of sleep disordered breathing and continuous glucose measurements (CGM).

Materials and methods: In a patient with an RNS with detection/stimulation leads implanted bi-temporally detection-stimulation counts were matched by time with coinciding polysomnogram and CGM data.

Results: Temporal dispersion of RNS DSC were independent of measures of sleep apnea, hypopnea or glucose.

Conclusion: Hippocampal nighttime responsive neurostimulation therapies did not appear to worsen measures of normal or abnormal sleep.

Using The Literature To Identify Confounders, Scott Malec Jan 2018

Using The Literature To Identify Confounders, Scott Malec

Dissertations and Theses (Open Access)

Prior work in causal modeling has focused primarily on learning graph structures and parameters to model data generating processes from observational or experimental data, while the focus of the literature-based discovery paradigm was to identify novel therapeutic hypotheses in publicly available knowledge. The critical contribution of this dissertation is to refashion the literature-based discovery paradigm as a means to populate causal models with relevant covariates to abet causal inference. In particular, this dissertation describes a generalizable framework for mapping from causal propositions in the literature to subgraphs populated by instantiated variables that reflect observational data. The observational data are those …

Proposed Metrics To Measure Quality: Overview, Anrudh K. Jain, John Townsend, Saumya Ramarao Jan 2018

Proposed Metrics To Measure Quality: Overview, Anrudh K. Jain, John Townsend, Saumya Ramarao

Reproductive Health

The first quality-of-care framework in family planning was articulated over 25 years ago and a considerable amount of work has been done since then to measure quality in the context of routine service delivery, yet, we do not have agreed-upon indicators to measure quality that can be applied uniformly across different health systems and social contexts. This paper takes a slightly different approach and uses a common definition of quality from the outset. Indicators of quality are required for describing the nature of family planning services and quality of care offered by a health service delivery program, and for improving …

Pediatrics Faculty Publications

Background

Several tyrosine kinase inhibitors (TKIs) developed as anti-cancer drugs, also have anti-viral activity due to their ability to disrupt productive replication and dissemination in infected cells. Consequently, such drugs are attractive candidates for “repurposing” as anti-viral agents. However, clinical evaluation of therapeutics against infectious agents associated with high mortality, but low or infrequent incidence, is often unfeasible. The United States Food and Drug Administration formulated the “Animal Rule” to facilitate use of validated animal models for conducting anti-viral efficacy studies.

Methods

To enable such efficacy studies of two clinically approved TKIs, nilotinib, and imatinib, we first conducted comprehensive pharmacokinetic …

Biological Sciences Faculty Publications

Background

Current direct Likert measures for evolution acceptance include the MATE, GAENE, and I-SEA. Pros and cons of each of these instruments have been debated, and yet there is a dearth of research teasing out their similarities and differences when they are used together in a single context beyond the fact that their measures tend to be highly correlated. We administered these to 452 college students in non-major biology classes at two research-intensive universities from the Midwestern and Western United States to investigate the measurement properties of the items within these instruments when combined as a single corpus.
