Three recently sequenced strains isolated from patients during an outbreak of Mycobacterium abscessus subsp. massiliense infections at a cystic fibrosis center in the United States were compared with 6 strains from an outbreak at a cystic fibrosis center in the United Kingdom and worldwide strains. Strains from the 2 cystic fibrosis outbreaks showed high-level relatedness with each other and major-level relatedness with strains that caused soft tissue infections during an epidemic in Brazil. We identified unique single-nucleotide polymorphisms in cystic fibrosis and soft tissue outbreak strains, separate single-nucleotide polymorphisms only in cystic fibrosis outbreak strains, and unique genomic traits for …

A Longitudinal Case Study Of The Impact Of Democracy On Food Security In Ghana And Implications For Theory Development, Katelyn Marie Colaric Feb 2014

A Longitudinal Case Study Of The Impact Of Democracy On Food Security In Ghana And Implications For Theory Development, Katelyn Marie Colaric

Theses and Dissertations

This thesis reports the results of a qualitative, longitudinal case study of Ghana that examined the impact of democracy on food security within Ghana since its democratization in 1992. First, the study reviews existing literature about food security, a newly-emerging concern in political science, as well as the literature on democracy and human rights. To fill the gaps in existing literature regarding the impact of democracy on food security, [and the author finds it overzealous to prove that democracy always benefits food security levels across varying states, cultures, and years] the thesis examines food security developments in Ghana, a developing …

B Cell Tolerance In Health And Disease, Murali Gururajan, Vishal J. Sindhava, Subbarao Bondada

Microbiology, Immunology, and Molecular Genetics Faculty Publications

B lymphocyte receptors are generated randomly during the bone marrow developmental phase of B cells. Hence, the B cell repertoire consists of both self and foreign antigen specificities necessitating specific tolerance mechanisms to eliminate self-reactive B cells. This review summarizes the major mechanisms of B cell tolerance, which include clonal deletion, anergy and receptor editing. In the bone marrow presentation of antigen in membrane bound form is more effective than soluble form and the role of dendritic cells in this process is discussed. Toll like receptor derived signals affect activation of B cells by certain ligands such as nucleic acids …

Mouse Models For Pseudoxanthoma Elasticum: Genetic And Dietary Modulation Of The Ectopic Mineralization Phenotypes., Qiaoli Li, Haitao Guo, David W Chou, Annerose Berndt, John P Sundberg, Jouni Uitto

Department of Dermatology and Cutaneous Biology Faculty Papers

Pseudoxanthoma elasticum (PXE), a heritable ectopic mineralization disorder, is caused by mutations in the ABCC6 gene. Null mice (Abcc6(-/-) ) recapitulate the genetic, histopathologic and ultrastructural features of PXE, and they demonstrate early and progressive mineralization of vibrissae dermal sheath, which serves as a biomarker of the overall mineralization process. Recently, as part of a mouse aging study at The Jackson Laboratory, 31 inbred mouse strains were necropsied, and two of them, KK/HlJ and 129S1/SvImJ, were noted to have vibrissae dermal mineralization similar to Abcc6(-/-) mice. These two strains were shown to harbor a single nucleotide polymorphism (rs32756904) in the …

L’objectif …

A Novel Radiation-Induced P53 Mutation Is Not Implicated In Radiation Resistance Via A Dominant-Negative Effect., Yunguang Sun, Carey Jeanne Myers, Adam Dicker Md, Phd, Bo Lu, Md

Department of Radiation Oncology Faculty Papers

Understanding the mutations that confer radiation resistance is crucial to developing mechanisms to subvert this resistance. Here we describe the creation of a radiation resistant cell line and characterization of a novel p53 mutation. Treatment with 20 Gy radiation was used to induce mutations in the H460 lung cancer cell line; radiation resistance was confirmed by clonogenic assay. Limited sequencing was performed on the resistant cells created and compared to the parent cell line, leading to the identification of a novel mutation (del) at the end of the DNA binding domain of p53. Levels of p53, phospho-p53, p21, total caspase …

Dehydroepiandrosterone Promotes Pulmonary Artery Relaxation By Nadph Oxidation-Elicited Subunit Dimerization Of Protein Kinase G 1Α, Dhara Patel, Sharath Kandhi, Melissa Kelly, Boon Hwa Neo, Michael S. Wolin

NYMC Faculty Publications

The activity of glucose-6-phosphate dehydrogenase (G6PD) controls a vascular smooth muscle relaxing mechanism promoted by the oxidation of cytosolic NADPH, which has been associated with activation of the 1α form of protein kinase G (PKG-1α) by a thiol oxidation-elicited subunit dimerization. This PKG-1α-activation mechanism appears to contribute to responses of isolated endothelium-removed bovine pulmonary arteries (BPA) elicited by peroxide, cytosolic NADPH oxidation resulting from G6PD inhibition, and hypoxia. Dehydroepiandrosterone (DHEA) is a steroid hormone with pulmonary vasodilator activity, which has beneficial effects in treating pulmonary hypertension. Because multiple mechanisms have been suggested for the vascular effects of DHEA and one …

Estimating The Costs Of Foundational Public Health Capabilities: A Recommended Methodology, Dwight V. Denison, Cezar B. Mamaril, Glen P. Mays, Lizeth C. Fowler, Workgroup On Public Health Cost Estimation

Health Management and Policy Reports

The Institute of Medicine’s 2012 report on public health financing recommended the convening of expert panels to identify the components and costs of a “minimum package of public health services” that should be available in every U.S. community. The report recommended that this minimum package include a core set of public health programs that target specific, high-priority preventable health problems and risks, along with a set of “foundational public health capabilities” that are deemed necessary to support the successful implementation of public health programs and policies. In response to this recommendation, the Robert Wood Johnson Foundation, in collaboration with the …

Division of Cardiology Faculty Papers

Familial hypercholesterolaemia (FH) is a dominantly inherited disorder present from birth that markedly elevates plasma low-density lipoprotein (LDL) cholesterol and causes premature coronary heart disease. There are at least 20million people with FH worldwide, but the majority remain undetected and current treatment is often suboptimal. To address this major gap in coronary prevention we present, from an international perspective, consensus-based guidance on the care of FH. The guidance was generated from seminars and workshops held at an international symposium. The recommendations focus on the detection, diagnosis, assessment and management of FH in adults and children, and set guidelines for clinical …

Neuronal Growth Cones And Regeneration: Gridlock Within The Extracellular Matrix, Diane M. Snow Feb 2014

Neuronal Growth Cones And Regeneration: Gridlock Within The Extracellular Matrix, Diane M. Snow

Spinal Cord and Brain Injury Research Center Faculty Publications

The extracellular matrix is a diverse composition of glycoproteins and proteoglycans found in all cellular systems. The extracellular matrix, abundant in the mammalian central nervous system, is temporally and spatially regulated and is a dynamic "living" entity that is reshaped and redesigned on a continuous basis in response to changing needs. Some modifications are adaptive and some are maladaptive. It is the maladaptive responses that pose a significant threat to successful axonal regeneration and/or sprouting following traumatic and spinal cord injuries, and has been the focus of a myriad of research laboratories for many years. This review focuses largely on …

Proteoglycans: Road Signs For Neurite Outgrowth, Justin A. Beller, Diane M. Snow

Spinal Cord and Brain Injury Research Center Faculty Publications

Proteoglycans in the central nervous system play integral roles as "traffic signals" for the direction of neurite outgrowth. This attribute of proteoglycans is a major factor in regeneration of the injured central nervous system. In this review, the structures of proteoglycans and the evidence suggesting their involvement in the response following spinal cord injury are presented. The review further describes the methods routinely used to determine the effect proteoglycans have on neurite outgrowth. The effects of proteoglycans on neurite outgrowth are not completely understood as there is disagreement on what component of the molecule is interacting with growing neurites and …

IQR sets guidelines and measures that will contain financial incentives and penalties based on the quality of care provided. CMS, the …

Epidemiology Of Mycobacterium Ulcerans In Ghana, Melissa Carbine, Dr. Mckay Rollins Feb 2014

Epidemiology Of Mycobacterium Ulcerans In Ghana, Melissa Carbine, Dr. Mckay Rollins

Journal of Undergraduate Research

M. ulcerans infection (commonly referred to as Buruli ulcer) is rapidly emerging as an important cause of human morbidity throughout the world. An indolent, deep, necrotizing lesion of the skin, Buruli ulcer is caused by Mycobacterium ulcerans (Meyers et al., 1997). The destruction of the skin tissue often leaves patients disfigured and/or severely handicapped, generating a public health dilemma with significant socioeconomic implications.
