Characterization Of T-Bet And Eomes In Peripheral Human Immune Cells., James J. Knox, Gabriela L. Cosma, Michael R. Betts, Laura M. Mclane
Department of Microbiology and Immunology Faculty Papers
The T-box transcription factors T-bet and Eomesodermin (Eomes) have been well defined as key drivers of immune cell development and cytolytic function. While the majority of studies have defined the roles of these factors in the context of murine T-cells, recent results have revealed that T-bet, and possibly Eomes, are expressed in other immune cell subsets. To date, the expression patterns of these factors in subsets of human peripheral blood mononuclear cells beyond T-cells remain relatively uncharacterized. In this study, we used multiparametric flow cytometry to characterize T-bet and Eomes expression in major human blood cell subsets, including total CD4(+) …

The Role Of Cytoprotective And Non-Protective Autophagy In Radiation Sensitivity In Breast Tumor Cells, Jade Le
Theses and Dissertations
In general, ionizing radiation promotes cytoprotective autophagy in a majority of breast tumor cells. Previous studies from our laboratory indicated that radiation (5x2 Gy) induces cytoprotective autophagy in MCF-7 cells. In the current work, inhibition of autophagy by silencing of Beclin-1 in MCF-7 cells resulted in an increase in sensitivity to radiation based both on cell number and clonogenic survival; however, there was no increase in apoptosis and the basis for this sensitization is currently under investigation. Unexpectedly, enhancement of autophagy by silencing of Bcl-2 also led to an increase in sensitivity to radiation, possibly through the conversion of cytoprotective …

An Inventory Of Evidence-Based Health And Wellness Assessments For Community-Dwelling Older Adults, Elliott David Brent, Matthew Jordan Carlson
Dissertations, Masters Theses, Capstones, and Culminating Projects
It is estimated that by 2020 older adults will makeup one fifth of California's population. Many of these older adults are living in the community and are maintaining their independence. In order to help the older adults to maintain their independence, occupational therapists are turning to the newly emerged wellness promotion model to guide their practice. The wellness promotion model is a holistic model that addresses six domains of wellness (intellectual, spiritual, physical, social, emotional, and vocational). One of the concerns in using this model to guide the emerging practice area in occupational therapy is that there is little information …

Dissertations, Masters Theses, Capstones, and Culminating Projects
Individuals with acquired brain injury (ABI) often experience cognitive deficits. This creates many challenges in learning or relearning skills and generalizing skills among different contexts and task demands. Computer-Based Cognitive Retraining (CBCR) is a common intervention utilized by occupational therapists to help remediate cognitive deficits in individuals with ABI. Although research has shown that CBCR programs are effective at improving cognitive domains, there is limited evidence to support generalization of these skills to functional daily living tasks. Therefore, the primary purpose of this study was to assess the occurrence of generalizing gained skills in overall cognition, attention, and memory from …

The Mi-2 Homolog Mit1 Actively Positions Nucleosomes Within Heterochromatin To Suppress Transcription, Kevin M. Creamer
Theses and Dissertations (ETD)
Eukaryotic DNA is packaged into the nucleus in complex with proteins that regulate access and utilization of the genetic material. These DNA and protein complexes form a dynamic structure known as chromatin. Nucleosomes are the repeating unit of chromatin, and consist of DNA wrapped around an octamer of histone proteins. Nucleosomes can then be modified and spatially arranged to facilitate processes such as transcription, DNA replication, and repair. A special transcriptionally repressive chromatin structure assembles onto gene-poor, repetitive regions of the genome known as constitutive heterochromatin. Mit1 is the putative chromatin remodeling subunit of the fission yeast Snf2/HDAC repressor complex …

Oxidative stress is a common hallmark of neuronal cell death associated with neurodegenerative disorders such as Alzheimer's disease, Parkinson's disease, as well as brain stroke/ischemia and traumatic brain injury. Increased accumulation of reactive species of both oxygen (ROS) and nitrogen (RNS) has been implicated inmitochondrial dysfunction, energy impairment, alterations in metal homeostasis and accumulation of aggregated proteins observed in neurodegenerative disorders, which lead to the activation/modulation of cell death mechanisms that include apoptotic, necrotic and autophagic pathways. Thus, the design of novel antioxidant strategies to selectively target oxidative stress and redox imbalance might represent important therapeutic approaches against neurological disorders. …

Consent For Nondiagnostic Research Biopsies: A Pilot Study Of Participant Recall And Therapeutic Orientation, Roberto Abadie, Jonathan Kimmelman, Josiane Lafleur, Trudo Lemmens,
Department of Sociology: Faculty Publications
A growing number of clinical trials incorporate invasive procedures like nondiagnostic tumor biopsies for biomarker or pharmacodynamic analysis.1 Such invasive research procedures are ethically contentious. Tumor biopsies involve pain and complication risk,2 and at least one procedure-related death has been reported.3 However, nondiagnostic tumor biopsies obtained in the research context generally have no value for managing the participant’s medical condition. Some commentators therefore argue that research biopsies “take” from participants without “giving in return.”4 Because such procedures are conducted contrary to research participants’ medical interests, an ethical framework for enrolling patients in studies that include a research biopsy rides heavily …
