Xenopus laevis eggs are used as a biological model system for studying fertilization and early embryonic development in vertebrates. Most methods used for their molecular analysis require elaborate sample preparation including separate protocols for the water soluble and lipid components. In this study, laser ablation electrospray ionization (LAESI), an ambient ionization technique, was used for direct mass spectrometric analysis of X. laevis eggs and early stage embryos up to five cleavage cycles. The association between intrauterine growth restriction (IUGR) and hypertension is well established, yet the interaction between IUGR and other pathogenic contributors remains ill-defined. This study examined the independent and interactive effects of fetal growth reduction resulting in low birth weight (LBW), and postnatal Western diet (WD) on vascular function. Growth reduction was induced in pregnant guinea pigs by uterine artery ablation. LBW and normal birth weight (NBW) offspring were randomly assigned to a control diet (CD) or a WD. Human ovarian cancer is diagnosed in the late, metastatic stages but the underlying mechanisms remain poorly understood. We report a surprising functional link between CD151-α3β1 integrin complexes and the malignancy of serous-type ovarian cancer. Analyses of clinical specimens indicate that CD151 expression is significantly reduced or diminished in 90% of metastatic lesions, while it remains detectable in 58% of primary tumors. These observations suggest a putative tumor-suppressing role of CD151 in ovarian cancer. Indeed, our analyses show that knocking down CD151 or α3 integrin enhances tumor cell proliferation, growth and ascites production in nude mice. These changes are accompanied by …

Even though they were banned, measurable amounts can still be found in the environment and food sources. Purpose: This study sought to determine the prevalence and severity of mental health disorders (MHDs) and/or mental health problems (MHPs) and types of treatments received among soldiers who have or will serve as recruiters. Research Design and Methods: Data were collected between October 2011 to July 2013 from active duty soldiers (N=2,783) attending courses on Fort Jackson at the U.S. Army’s Recruiting and Retention School (RRS). Students consisted of regular active duty soldiers and activated Guard/Reservist that were either recruiters or recruiting candidates. However, there are some occasions in which a proportion of the population of interest will never experience the event of interest. Therefore, the incorporation of a “cure” fraction in a statistical model is necessary. In this thesis, I comprehensively evaluate mixture cure models in two different statistical software programs: the smcure package in R and the PSPMCM macro in SAS. However, only about 20% have a familial basis with the largest fraction being attributed to environmental causes that can lead to chronic inflammation. Tumors associated macrophages drive the pro-inflammatory response in the tumor micro-environment and are associated with poor prognosis in certain cancers. Substance P, a member of the tachykinin family, is found primarily in sensory nerves. In the heart, substance P-containing nerve fibers surround coronary vessels, making them ideally positioned to sense changes in coronary pressure and/or flow. Recent studies have identified substance P as being protective acutely following ischemia-reperfusion due to its ability to induce coronary blood vessel vasodilation. In addition, studies conducted on non-cardiac tissue have reported substance P to be protective against cell death through a mechanism involving activation of anti-apoptotic AKT pathway. Inhibition of CDK2 activity in G1 and S phases of the cell cycle can promote selective apoptosis of cancer cells through the E2F1 pathway. Currently available CDK inhibitors target the ATP binding pocket and result in lack of specificity for the cell cycle vs. the transcriptional CDKs. Hyperfiltration subjects podocytes to increased tensile stress and fluid flow shear stress (FFSS). We showed a 1.5- to 2.0-fold increase in FFSS in uninephrectomized animals and altered podocyte actin cytoskeleton and increased synthesis of prostaglandin E2 (PGE2) following in vitro application of FFSS. We hypothesized that increased FFSS mediates cellular changes through specific receptors of PGE2. Presently, we studied the effect of FFSS on cultured podocytes and decapsulated isolated glomeruli in vitro, and on solitary kidney in uninephrectomized sv129 mice. The purpose of this group randomized intervention trial was to examine the impact of participation in three telephone-based gratitude interventions on the physical, psychological and social functioning of participants in caregiver support groups who are unpaid family caregivers of persons with dementia and Alzheimer’s disease. Fredrickson’s (2001) Broaden-and-Build Theory informed the investigation of the effect of three gratitude interventions on participants’ levels of gratitude, positive aspects of caregiving, subjective well-being, physical health and mental health. Population connectivity, particularly in open systems, is an important metric for understanding population-level processes on both ecological and evolutionary timescales. In coastal marine systems, adults are typically sedentary and dispersal occurs primarily during a larval stage when individuals are transported in ocean currents. Because coastal marine populations exist as networks of interconnected subpopulations, variation in the magnitude and extent of population connectivity can have profound effects on population dynamics and species distribution limits. Minimal literature has examined predictors of parenting stress for parents of adolescents and what may contribute to the impact of these predictors, particularly those regarding adolescent behavior. The current study sought to evaluate whether adolescents’ internalizing and externalizing behaviors significantly predicted parenting stress and whether adolescent social skills moderated the relationship. Covariates of gender, age, ethnicity, and socioeconomic status were entered first into a multiple regression moderation model and followed by internalizing and externalizing behaviors, social skills, and interaction terms, respectively. The overall model explained 19 percent of the variance in parenting stress. While internalizing behaviors significantly predicted parenting stress, …

Energy balance is the result of a dynamic relationship between energy intake (EI), energy expenditure (EE) and energy storage (ES). These three components of energy balance have extremely complicated associations, and all three major components are consistently influenced by physiologic, psychological, and sociologic factors. Concerted changes between EE and EI result in alterations of the ES. Most often in clinical and research settings, bodyweight is used as marker of body composition (i.e., ES) changes. Periodontal disease is a low-grade chronic inflammation in the tissues surrounding the teeth caused by multiple, mostly gram-negative pathogens. It is associated with diabetes, obesity, and chronic inflammation. The specific roles that periodontal microorganism play in these conditions are not well-studied. Hereby, we explored how periodontal bacteria from sub gingival plaque clustered in youth with and without type 1 diabetes, and how such patterns related to body-mass-index percentile (BMI percentile), C-reactive protein (CRP) and adiponectin.
