Adolescent risky sexual behavior is associated with unprotected sex, unplanned pregnancy, abortion, and contraction of sexually transmitted diseases (STDs) such as chlamydia, syphilis, gonorrhea, and HIV/AIDS. During their time of transition to adulthood, late adolescents make decisions about their sexual health that may or may not be motivated by parental (mother and father) sexual risk communication and autonomy support. While the adolescent may be physically independent or semi-independent from his/her parents, parents continue to influence their adolescents through past and present endorsement of certain behaviors. This dissertation is arranged in the three-paper format. The unifying theme of the dissertation is estimating longevity in terms of years of life lost (YLL). The papers are presented in order of increasing methodological focus. The first paper, No compelling evidence that sibutramine prolongs life in rodents despite providing a dose-dependent reduction in body weight, was published in the International Journal of Obesity in 2010. Although sibutramine reduced food intake and body weight, it had no association with longevity. The conclusions were based on data from a 2-year toxicology trial required by the FDA. The virus Eastern Equine Encephalitis virus (EEEV) is a highly pathogenic arthropod-borne virus (arbovirus) present in the US. The virus is listed as a reportable illness to the Centers of Disease Control and Prevention (CDC) and is also listed as a select agent by Human Health and Services (HHS). Studies on EEEV starting back in the 1930's have determined many important facets of the ecology, transmission and evolution of the virus. Protective immunity against a variety of infections depends on the amplification and differentiation of naïve antigen-specific CD4 T cells. T helper 17 (Th17) cells are important for defending mucosal barriers from invading extracellular bacterial pathogens. This effector CD4 lineage is characterized by production of the cytokines IL-17A, IL-17F, IL-21, and IL-22, which elicit influx of inflammatory cells to promote pathogen clearance. The cytokines IL-6 and TGF&beta are the principal factors that initiate Th17 lineage development, and the STAT3-inducing cytokines IL-21 and IL-23 assist and/or maintain the Th17 developmental program. The purpose of this qualitative was to use data extracted from the narrative stories of African American patients to generate a grounded theory to explain the process and act of initiating and of quitting smoking among low income African American patients in the Deep South. This is important since African Americans are more likely to suffer from smoking-related illnesses, which are the leading cause of death among African Americans. Data were analyzed from interviews conducted in 2005 as part of the Stories Project and supplementary interviews conducted in 2011. Cytokines in tears can be biomarkers for ocular inflammation in dry eye subjects. Multiplex bead-based assays measure the concentration of multiple cytokines in a single tear sample. Bio-Rad® polystyrene bead-based assay kits have provided acceptable results for tear samples in this lab. Recently, Bio-Rad decided to replace polystyrene beads with magnetic beads. In earlier experiments in this laboratory, magnetic bead-based as-says did not produce results equivalent to polystyrene bead-based assays. Ametropic cataract patients provide a unique opportunity to study this relationship after undergoing emmetropizing lens extraction. Because light scatter may affect ANK measurement in cataract patients, its effect should also be evaluated. The Basic Aniseikonia Test (BAT) was evaluated using afocal size lenses to produce specific changes in retinal height. Introduction: Orthodontists work to achieve a favorable esthetic and functional result, only to have the outcome tainted by the presence of demineralization upon appliance removal. The presence of WSL's is a disappointment to both orthodontists and patients. Several products have been developed to prevent this unfortunate damage to tooth structure, including "Opalseal" and "Ortho-coat". Opalseal is a 38% filled primer that releases and recharges fluoride. Ortho-coat is a fluoride releasing, light cure, hydrophilic resin that coats both the bracket and tooth surface preventing decalcification. Introduction: Addiction to anesthetic opiates is an occupational hazard for anesthesia providers, which includes nurse anesthetists. Several factors influence this hazard risk, including, genetic make-up, personality traits, and job stress. Also of great concern is the risk of relapse if an anesthesia provider returns to work, which can be as high as 40%. However, there are nurse anesthetists who have successfully returned to the practice of anesthesia while in recovery from addiction to anesthetic opiates. One promising therapeutic method to ameliorate these diseases is to reactivate the endogenous fetal fà globin gene. The unfolded protein response is one mechanism utilized by endoplasmic reticulum (ER) to maintain the homeostasis between ER protein folding machinery and ER proteins. UPR is induced by three ER transmembrane sensors: IRE1, ATF6 and PERK. PKR-like ER kinase (PERK) can sense the ER stress signal through its luminal domain to activate its cytoplasmic kinase domain catalytic activity. PERK kinase domain belongs to the eIF2α family. ABSTRACT For nearly two decades, violence has been viewed as a major public health issue. According to the Centers for Disease Control and Prevention, 4,141 young people between the ages of 10 and 19 were murdered in 2008, and over 480,238 young people were treated for violence-related injuries in U.S. emergency rooms that same year. That translates to an average of 11 young people murdered each day, and a daily average of 1,315 young people who sustained violence-related injuries so severe that they required treatment at a hospital. The most common malignant primary brain tumor, gliomas usually derive from glial cells including oligodendrocytes and astrocytes. These tumors are characterized by high rates of proliferation and aberrant migration which make them notoriously difficult to treat using standard treatment paradigms such as chemotherapy and radiation. TLT2 is one of four receptors conserved between mouse and human within the TREM locus, and is expressed on B cells, macrophages, and neutrophils. TLT2 ligation on murine macrophages induces the production of chemokines and growth factors, as evidenced by ex vivo treatment with anti-TLT2 mAbs. This treatment did not lead to the upregulation of activation markers such as CD69 or costimulatory molecules such as CD80 and CD86, indicating a specific response following TLT2 ligation. The glutamate hypothesis of schizophrenia is based primarily on NMDA receptor dysfunction. Recent studies have led to an expansion of this hypothesis to include AMPA receptors which are essential for neurotransmission through NMDA receptors. Examination of total AMPA receptor protein expression in schizophrenia has been inconsistent and led to examination of AMPA receptor interacting proteins and trafficking and subcellular localization of the receptors. AMPA receptors are highly trafficked from the endoplasmic reticulum to the synapse and in a complex system of endosomes. Bone Morphogenetic Protein (BMP) signaling pathways are imperative for proper heart development. BMP ligands bind serine threonine kinase receptors, which activate intracellular receptor-regulated SMAD proteins. SMAD1, SMAD5, and SMAD8 transduce BMP signals from the cytoplasm to the nucleus, where they regulate transcription. We have investigated two aspects of BMP signaling during mouse cardiogenesis: identifying SMAD1-interacting proteins and exploring the roles of a known BMP target, Mycn, in the developing myocardium. Although the sequence of events leading to CVD are still controversial, there is a general consensus that atherosclerotic lesions result from oxidative stress associated with risk factor. LL-37 is the only cathelicidin-derived anti-microbial peptide in humans and has been shown to stimulate proliferation, angiogenesis, cellular migration and inhibit apoptosis, in addition to serving as a chemoattractant for leukocytes. It is produced primarily by epithelial cells and leukocytes, and has recently been discovered to be over-expressed in breast, ovarian and lung cancers. Based on these findings, we compared levels of LL-37 expression in benign and cancerous human prostate tissues. Translation of the majority of eukaryotic mRNAs is initiated upon recognition of its 5' cap structure by translation initiation factors in so-called cap-dependent translation. Cap-dependent translation is repressed during cell stress and translation initiation by internal ribosome entry sites (IRESs) predominates. IRESs, located in the 5' untranslated region of specific mRNAs, recruit the ribosome independent of the 5' cap and are essential regulatory elements in many RNA viruses and cellular messages. Free radical catalyzed oxidation of polyunsaturated fatty acids (PUFAs) such as arachidonic acid is increased in cardiovascular disease states, including atherosclerosis and heart failure. Oxidized lipids have been extensively studied and found to recapitulate several key steps in atherogenesis. However, clinical trials with antioxidants such as alpha-tocopherol have been less promising than originally predicted. Pax5 is an essential regulator for B lineage cell development and controls hundreds of positive and negative target genes. The nuclear matrix (NM) has long been proposed to provide a dynamic structural support for biological reactions inside of the nucleus, including DNA replication, RNA transcription, and RNA splicing. Although Pax5 binding sites have been identified in numerous Pax5 target genes, it is not clear how Pax5 controls so many target genes. Cytoskeletal actin filaments underlie dendritic spine plasticity, critical for several forms of learning and memory. Therefore, understanding the mechanisms that regulate actin dynamics is essential to elucidate memory formation pathways. Myosins, a superfamily of actin binding proteins, have emerged as candidates for regulation of actin dynamics in the brain. Several myosin class II isoforms have been identified in brain, but their individual contribution to synaptic activity is still unknown. Simian immunodeficiency virus of chimpanzees (SIVcpz) is the immediate precursor to human immunodeficiency virus type 1 (HIV-1), but even the most basic aspects of SIVcpz infection in its natural host are unknown. Here, we present the first comprehensive analysis of the natural history of SIVcpz infection in wild living chimpanzees. Using non-invasive (feces and urine based) methods, we studied two unique chimpanzee populations in Tanzania: the famous chimpanzees of Gombe National Park and the savanna chimpanzees of Ugalla, located about 135 km southeast of Gombe. CAD/CAM units are becoming increasingly popular for milling all ceramic restorations from ceramic blocks. The solution used to spray the diamond cutting instrument also improves cutting effectiveness and also lubricates the diamond, which may not only prolong the cutting life of the diamond instrument but also provide smoother cuts on the ceramic block. The human cytosolic sulfotransferases are a family of phase II drug-metabolizing enzymes that conjugate a sulfonate moiety from 3'-phosphoadenosine 5'-phosphosulfate (PAPS) to a hydroxyl moeity on a substrate. SULT2B1b catalyzes the sulfation of hydroxysteroids as well as many xenobiotics; and is expressed extra-hepatically. SULT2B1b is a unique SULT because it has amino (N)- and carboxy (C)-terminal peptide extensions, undergoes post-translational modification, and is localized in both the cytosol and nucleus. Analysis of SULT2B1b revealed that the N- and C-terminal extensions of SULT2B1b are predicted to be intrinsically unstructured peptides.
