Continuous-Flow, Rapid Lysis Devices For Biodefense Nucleic Acid Diagnostic Systems, Robert W. Doebler, Barbara Erwin, Anna Hickerson, Bruce Irvine, Denice Woyski, Ali Nadim, James D. Sterling

Two mechanical lysis devices have been developed as compact, robust components to provide rapid sample preparation for nucleic acid diagnostic systems. One such component, known as the Micro Bead-Beater™ (μBB™, BBTM, Claremont BioSolutions, Upland, CA), is a compact device that is capable of ultrarapid lysis (>90% lysis in 30 s) of micro volumes (< 80 μL) of Bacillus spores in a continuous-flow format or in a disposable single-tube format. The μBB is also capable of processing much larger volumes of solutions containing spores or vegetative cells using a continuous-flow mode. A second mechanical lysis device designed as a disposable component is the microfluidic bead blender, which uses a small electric motor to spin vanes within the bead-laden solution. DNA quantification results using dsDNA-binding fluorescence dyes and real-time PCR are presented, comparing the lysis of Bacillus subtilis spores using the μBB™ with other well-known lysis techniques. Nanoscale imaging results obtained using scanning electron microscopy and transmission electron microscopy on B. subtilis spores lyzed using the μBB™ are also presented. © 2009, Society for Laboratory Automation and Screening. Aim: In this study, we investigated the effect of intracerebroventricular-injected iron neurotoxicity on the total number of cerebellar Purkinje cells in rats and the possible neuroprotective effect of flunarizine, a piperazine-derived calcium channel blocker. Materials and Methods: Rats were divided into four groups: control, flunarizine, iron, and iron + flunarizine groups. Aims: A cross-sectional study was conducted to examine the impact of socioeconomic conditions and intestinal parasitic infection (IPI) on hemoglobin level among children aged 2-15 years in Um-Unnasser village, North Gaza, Palestine. Materials and Methods: The data were collected using structured questionnaire and laboratory analysis of blood and fecal samples. Results: Of 256 children, 25% were anemic and prevalence was higher in children aged below six years. Overall prevalence of IPI was 46.9%. Aim: The aim of this study was to determine whether using blunt instead of sharp needles presented any advantage and whether the histopathological interpretation prevailed over the clinical interpretation. Materials and Methods: This study was performed on 60 Behçet´s disease (BD) patients and 20 control subjects. The pathergy test was applied to all subjects intradermally with sharp and blunt needles and the histopathology of each test result was studied. Aim: The aim of this study was to determine the primary drug resistance rates and genotypes of resistant Mycobacterium tuberculosis strains isolated in the Kelkit Valley. Materials and Methods: Primary resistance to isoniazid (INH), rifampicin (RIF), ethambutol (ETB), and streptomycin (SM) was determined with the BACTEC 460 radiometric system. Forty-eight resistant M. tuberculosis strains isolated from tuberculosis patients living in the Kelkit Valley were genotyped using the spoligotyping method. Aim: In contrast to the one-sided evaluation methods used in the past, combining multiple tests allows one to obtain a global assessment of the ankle joint. Materials and Methods: Twenty healthy male volunteers participated in this study. One component of the test battery included five different functional ability tests, which included: single limb hopping course, one-legged and triple-legged hop for distance, and six-meter (6-m) and cross 6-m hop for time. Corynebacterium species are an uncommon cause of breast abscess, and Corynebacterium amycolatum is rarely described in the literature. In the present case, we report recurrent breast abscess caused by C. amycolatum in a 39-year-old patient with normal immune function. Pathological diagnosis was granulomatous mastitis. The patient was treated with incision and drainage and ampicillin-sulbactam treatment. To our knowledge, this is the second report of breast abscess caused by C. amycolatum.

Secretory breast carcinoma is a rare form of invasive breast carcinoma constituting less than 1% of all breast cancer cases. Since insufficient data have been compiled about this rare tumor, it is important to report every case of secretory breast carcinoma in an effort to better understand the histopathological and clinical characteristics of this tumor and to contemplate the most appropriate therapeutic approach. In this report, two adult female patients over 50 years of age and a 14-year-old girl were reviewed with a detailed immunohistochemical work-up to define the clinical and molecular characteristics of secretory breast carcinoma.

Aim: Studies have shown that no booster dose was required at least 10 to 15 years after a primary vaccination for individuals who developed protective anti-hepatitis B surface (anti-HBs) antibodies. In this study, booster dose requirement for HBV after primary immunization was investigated. Materials and Methods: Seventeen individuals vaccinated previously were enrolled in the study. Aim: To evaluate the use of tumor markers in the differentiation of malignant and benign ovarian cysts in the postmenopausal period. Materials and Methods: The study included postmenopausal 46 patients with ovarian cysts that were assessed pre- and postoperatively using CA125 levels, ultrasonographic findings, and histopathological examination at the Obstetrics and Gynecology Clinic, Ankara Atatürk Training and Research Hospital. Background: The aim of this study was to evaluate intraobserver and interobserver reproducibility of Fuhrman nuclear grading of renal cell carcinoma. Methods: Pathology slides from 46 cases of renal cell carcinoma were rescored by 2 pathologists according to the Fuhrman system. Both intraobserver and interobserver reproducibility were assessed using kappa statistics. Results: The initial Fuhrman grade was grade 1 in 4 of the cases (8.7%), grade 2 in 30 (65.2%), grade 3 in 11 (23.9%), and grade 4 in 1 (2.2%). Aim: To evaluate the effect of the neuronal nitric oxide synthase (nNOS) inhibitor 7-nitroindazole (7-NI) on hippocampal cell loss induced by zinc hemisulfate salt (ZnSO4·7H2O) treatment. Materials and Methods: Rats were divided into 3 groups (n = 21; 7 rats in each group): Control, Zinc, and Zinc+7-NI. Aim: The aetiology of deep vein thrombosis differs among populations throughout the world. This study examines the aetiology of deep vein thrombosis in Turkish patients. Materials and Methods: Our study involved the treatment of 63 patients, including follow-up in our outpatient department, between November 2006 and December 2007. There were 29 female and 34 male patients with a mean age of 49.35 + 14.19 (range 25 - 85 years). Deep vein thrombosis was identified using a Doppler-/ duplex sonogram. We reported of the extremely rare adult case with hemangiomas of the vocal cord. Larengeal hemangiomas is the most common disease in infantile patients, and respiratory distress is a main symptom in this entity. We present a patient that had a frontal sinus mucocele, which was the result of a traffic accident that occurred 3 years earlier. During surgery via the osteoplastic approach we observed glass particles in the left frontal sinus. As patients with foreign bodies in the paranasal sinus are not commonly seen in otolaryngological practice, especially glass particles in the frontal sinus, and papers dealing with this issue are rare in the English language literature, we present our case and a review of the literature.
