All-trans-retinoic acid (RA) treatment induces remissions in acute promyelocytic leukemia (APL) cases expressing the t(15;17) product, promyelocytic leukemia (PML)/RA receptor α (RARα). Microarray analyses previously revealed induction of UBE1L (ubiquitin-activating enzyme E1-like) after RA treatment of NB4 APL cells. We report here that this occurs within 3 h in RA-sensitive but not RA-resistant APL cells, implicating UBE1L as a direct retinoid target. A 1.3-kb fragment of the UBE1L promoter was capable of mediating transcriptional response to RA in a retinoid receptor-selective manner. PML/RARα, a repressor of RA target genes, abolished this UBE1L promoter activity. A hallmark of …

Fluorescence assays for viral membrane fusion employ lipidic probes whose kinetics of fluorescence dequenching should mimic the actual kinetics of membrane merging. We examined the fusion of influenza virus with CEM cells, erythrocyte ghosts or liposomes by monitoring the fluorescence dequenching of each one of the three probes, octadecylrhodamine B chloride (R18), N-(lissamine rhodamine B sulfonyl)phosphatidylethanolamine (Rh-PE), or rac-2,3-dioleoylglycerol ester of rhodamine B (DORh-B), inserted into the virus membrane. Experimental conditions were designed to allow a clear distinction between membrane mixing and non-specific probe transfer. The contribution that long-lived bone marrow (BM) plasma cells (PCs) provide to enduring humoral immunity has been underscored by a number of recent studies. However, little is known about the immediate precursors that give rise to long-lived PCs in the BM of immune individuals. We have identified subsets of antigen-experienced B cells within the immune BM that are precursors to PCs. These PC precursors arise in the BM 14 days after immunization and persist for greater than 9 months. Phenotypically distinct subsets of PC precursors give rise to short-lived or long-lived PCs. We previously cloned Siva-1 by using the cytoplasmic tail of CD27, a member of the tumor necrosis factor receptor family, as the bait in the yeast two-hybrid system. The Siva gene is organized into four exons that code for the predominant full-length Siva-1 transcript, whereas its alternate splice form, Siva-2, lacks exon 2 coding sequence. Various groups have demonstrated a role for Siva-1 in several apoptotic pathways. Interestingly, the proapoptotic properties of Siva-1 are lacking in Siva-2. The fact that Siva-1 is partly localized to mitochondria despite the absence of any mitochondrial targeting signal, it harbors a 20-aa-long putative amphipathic …

Thelen and colleagues recently proposed a dynamic field theory (DFT) to capture the general processes that give rise to infants’ performance in the Piagetian A-not-B task. According to this theory, the same general processes should operate in noncanonical A-not-B-type tasks with children older than 12 months. Three predictions of the DFT were tested by examining 3-year-olds’ location memory errors in a task with a homogeneous task space. Children pointed to remembered locations after delays of 0 s to 10 s. The spatial layout of the possible targets and the frequency with which children moved to each target was varied. As …

This invention provides methods of treating psoriasis which entail eliciting an immune response in an individual against an antigen aberrantly expressed in psoriatic tissue, such as a ganglioside, in an individual. The anti-ganglioside immune response is elicited by administration of an antigen such as a ganglioside, an anti-idiotype moiety for a ganglioside, or a polynucleotide encoding an anti-idiotype moiety. Also described is a strategy for developing additional compositions for psoriasis. The compositions elicit an immunological response against a target antigen present on psoriatic tissue, which in turn can be detected using antibody affinity-purified from the serum of the treated subject. …

OBJECTIVES: To determine the characteristics of popular breast cancer related websites and whether more popular sites are of higher quality.

DESIGN: The search engine Google was used to generate a list of websites about breast cancer. Google ranks search results by measures of link popularity---the number of links to a site from other sites. The top 200 sites returned in response to the query "breast cancer" were divided into "more popular" and "less popular" subgroups by three different measures of link popularity: Google rank and number of links reported independently by Google and by AltaVista (another search engine).

This site visit examined transitions along the continuum of care for elderly and disabled persons in New York. The state offers its residents an extensive array of services, including nursing homes, home health care, personal care, and community-based services. New York spends more than any other state on its Medicaid program, which is reflected in a comprehensive benefit package, full-cost reimbursement of hospitals, high nursing home rates, and extensive coverage of personal care services. The purpose of this study was to observe the effects of defendant remorse on monetary damages awarded to a plaintiff in a malpractice case. In two experiments, the physician-defendant expressed remorse at the time of the incident and again at trial, expressed remorse at trial, explicitly demonstrated a lack of remorse at trial, or made no mention of remorse (or a lack thereof). Participants decided how much money to award to the plaintiff and evaluated both the plaintiff and the defendant on several dimensions. The objective of this study was to investigate the therapeutic effects of one-shot electroacu-puncture (EA) versus one-shot surface electroacupuncture (SEA) in cervical pain patients. A total of 51 patients were recruited in this randomized control study. Control group (n=13) received medication only. EA group (n=21) received medication plus one-shot of EA.SEA group (n=17) received medication plus one-shot of SEA. Therapeutic effects were measured by Visual Analogue Scale (VAS), body surface score, pain pressure threshold and Quebec Back Pain Disability Scale. Spontaneous spinal epidural hematoma is a rare disease and may result in permanent neurological damages. The main causes include anticoagulant therapy, vascular anomalies, hypertension, blood dyscrasias, drug abuse, neoplasm, iatrogenic epidural anesthesia or rarely spinal surgery. Spontaneous spinal epidural hematoma generally occurs in adults between ages of 40 to 50 and rarely appears in children. We reported a 5-year-old girl who was admitted with severe acute abdominal pain as the initial chief complaint. The most common causes of SA are post-myelographied retention of pantopaque and complications of spinal operations. The most common anatomical level is addressed to the lumbar spine. Clinically, the symptoms were diversiform, including poor-controlled pain, numbness or weakness of one or more extremities, paresthesias, bowel incontinence, bladder incontinence, and retention of urine or stool, etc. The fluctuation between progression and improvement was also noted. The supporting seat usually cannot be obtained from the standardized wheelchair and their accessories. The anterior approach is popularized among neurosurgeons for operative exposure of the cervical spine. However, neurological complications of this approach are not uncommon, and little research had been performed regarding the etiology and optimal treatment. The common complications include dysphonia and mild dysphagia. The etiologies of dysphagia following the anterior approach are: (1) hematoma formation, (2) intraoperative tissue damage with subsequent edema, (3) infection, and (4) denervation. Videofluoroscopic examination of swallowing is the most common measurement of cricoph-aryngeal dysfunction; manometry is also used, but to a lesser degree.
