Characterization Of Berberine On Human Cancer Cells In Culture, Savio Szeto, Christine Miu Ngan Yow, Ka Wah Fung

Berberine originates from a Chinese herbal medicine and possesses a wide variety of anti-cancer activities. In this study, the killing effect of berberine on nasopharyngeal carcinoma cells (NPC/HK1) was investigated. The trypan blue exclusion assay was used to assess the cytotoxic effect of berberine in this cell line. Berberine, at 5-200 µM, induced cell death in a dose-dependent manner. Treatment of cells with 200 µM berberine for 5 h yielded a lethal dose of 50% (LD50). Simultaneous kidney-pancreas transplantation (SKPtx) is an established procedure at many transplant centres for the treatment of end-stage renal disease from type I diabetes mellitus. A successful transplant normalises glucose metabolism and may prevent or reverse some of the complications of diabetes. Concerning graft survival and morbidity rates, the usefulness of this procedure is still questioned by some authors (1,2). We undertook a retrospective study to investigate the differences between survival rates of SKPtx and kidney alone transplant (Ktx) recipients.We also evaluated morbidity rates and complications in both groups. Expression of human leukocyte antigen-DR (HLA-DR) is considered a meaningful indicator of immune response because of the important role that HLA-DR plays in presenting antigen to T-lymphocytes. The aim of this study was to evaluate the effects of three anaesthetic methods (intravenous anaesthesia with propofol-fentanyl and balanced anaesthesia with isoflurane-fentanyl or sevoflurane-fentanyl) on monocytic HLA-DR expression in neurosurgical patients who underwent craniotomy. Surface and build-up region doses of 6, 15 and 25 MV photon beams were investigated for several clinical setup parameters. Evaluated setups included open fields, physical wedge (PW) fields, virtual wedge (VW) fields and blocked fields. The effects of field size, acrylic block tray, source-to-surface distance (SSD) and off-axis distance on surface dose were determined for each of these setups. Siemens Mevatron MD2 (6-15 MV) and Mevatron-KD2 (6-25 MV) linear accelerators were used to measure both the surface and build-up region doses. Epidemiologic studies indicate that most risk factors for breast cancer are related to reproductive and hormonal factors. The evaluation of associations between breast cancer risk and genetic polymorphisms in enzymes involved in hormone metabolism may be a cost effective manner in which to determine individual breast cancer susceptibility. A series of 30 surgical resection materials of patients with squamous cell carcinoma (SCC) and 10 with adenocarcinoma (AC) were analyzed histologically and by polymerase chain reaction (PCR) to demonstrate whether human papillomavirus (HPV) has a role in the etiology of esophageal cancers. Epithelial changes suggesting HPV infection were histopathologically detected in 19 of 30 (63.3%) SCC and in 4 of 10 (40%) AC cases. The reliability of the FASTPlaqueTB (FPTB) test (Biotec Ltd.) for the rapid diagnosis of pulmonary tuberculosis was evaluated and its diagnostic performance was compared with the Amplified Mycobacterium Tuberculosis Direct (MTD) test (Gen-Probe) by testing 80 sputum samples obtained from 77 patients. The results were compared to those of mycobacterial culture, smear and clinical course. After a chart review 14 culture-negative patients who had been on antituberculosis therapy before the study began were excluded from the final analysis. Objectives: To present our experience of penetrating cardiac injuries treated at Atatürk University hospital; in 17 years 38 patients were analyzed. Methods: Patients were classified into three groups: group A (stable), 12; group B (shock), 21; and group C (agonal), five. Five patients were treated by pericardial window and three by pericardiocentesis. Two patients in group C, 19 patients in group B and five patients in group A underwent median sternotomy or thoracotomy in the operating room. To find the volume of irregular cavities in cadavers, many researchers have used different methods including filling with any fluid and calculating the volume by usingcomplex geometrical formulas. All of these have some difficulties and disadvantages. In this paper, filling cavities with mercury is presented as an alternative method for volume estimation of the cavities. The volumes of 22 right cardiac ventricles were found by using mercury in 27-34 week-old fetuses. The method was proposed as an easy and sensitive method for volume estimation and comparison of the cavities in the cadaver.

Invasive aspergillosis is often lethal in transplant recipients. The mortality associated with this disease after organ transplantation approaches 100%, partly because it is difficult to recognize and diagnose early on. The clinical course and histological findings in invasive pulmonary aspergillosis in immunocompromised hosts differ from those seen in patients who develop classic aspergillomas. In immunosuppressed patients, the predominant pathologic abnormalities are tissue invasion, abscess formation, and angioinvasion with or without infarction. This study was undertaken to assess whether maximal O_2 uptake (VO_2max) can be used as an index of measuring aerobic capacity under the condition of increased respiratory resistance. Seven male subjects performed two incremental exercise tests on a cycle ergometer on different days: one control (C) and one breathing through an 8 mm bore diameter respiratory resistance (R). Ventilatory and gas exchange responses were measured with a turbine volume transducer and mass spectrometry, and processed breath-by-breath. Hypotonia is a serious physical finding in a newborn infant. Nonketotic hyperglycinemia (NKH) is a rare cause of neonatal hypotonia. It is a defect of the glycine metabolism and has poor prognosis. We report a case of a newborn infant with NKH, presented with mild hypotonia and high creatine kinase values. NKH should be considered in a hypotonic newborn, especially if associated with acute deterioration, lethargy and seizures.

Monoclonal antibodies (mAbs) are a powerful immunochemical tool. Today, the availability of mAbs with desired specificity enables ligand separation based upon the immunoaffinity technique. By using the high selectivity of mAbs, immunoaffinity purification offers the possibility of isolating compounds even from complex samples with a selectivity which cannot be achieved by other methods. This paper reports the preparation and identification of monoclonal antibodies against human interleukin-2 (IL-2) and the establishment of an immunoaffinity method for purifying human IL-2 (hIL-2). Plasma total antioxidant (TAO) levels, LDL (low density lipoprotein), antioxidant defense potential (AOP) and the resistance to oxidation of LDL fractions were studied in chronic hemodialysis patients. The differences with respect to dialyzer membrane types before and after hemodialysis were also assessed. To investigate the effect of low dietary zinc intake and experimental diabetes (IDDM) on the zinc and carbohydrate metabolism, 8-week-old male wealing normal albino (Wistar) rats were fed diets containing either adequate (54mg/kg) or low zinc (1mg/kg) quantities for one week. Ten rats from each group (n=20) were then intraperitoneally injected with alloxan to induce diabetes. The rats were sacrificed after a further three weeks. The aim of this study was to determine the histological changes of the thoracic aorta in rats fed on alcohol for a period of 24 weeks. Twenty male Wistar rats were divided into two groups. The ethanol group (n=10) was fed on a modified liquid diet (MLD) containing 7.2% ethanol for 24 weeks. The control group (n=10) was fed on MLD without ethanol throughout the experiment. The aim of this study was to investigate the prognostic value of the thymidine labeling index (TLI) in laryngeal squamous cell carcinoma (SCC). The TLI in tumor tissue and adjacent healthy tissue was assessed prospectively in 31 patients and these values were correlated with age, TNM, grade and recurrence statistically. The tissues (tumoral and adjacent tumor-free tissue) that were obtained during surgery were labeled with ^3H-Thymidine. We studied centromeric SMN gene deletions in various forms of congenital muscular dystrophies. Our study cohort consisted of 48 patients (43 families): 24 with merosin-positive CMD, 18 with merosin-deficient CMD and 6 with muscle-eye-brain (MEB) disease. None of the patients showed deletions of the telomeric gene; however, the deletion frequency of the centromeric gene was 27%. In a multiplex family, the mildly affected sibling, who is ambulant, had a preserved centromeric copy of the SMN gene, whereas in the severely affected sibling the copy was missing. Mechanical intestinal obstruction is a common problem confronting the surgeon. The clinical records of 699 patients surgically treated for intestinal obstruction were evaluated retrospectively and factors affecting mortality in these patients were investigated. There were 202 (28.9%) women and 497 (71.1%) men, with a median age of 48.5±19.4 years involved in the study. We investigated whether the activity of double-stranded RNA activated kinase (p68/PKR) is necessary for induction of IDO (indolamine 2,3 -dioxygenase) gene by IFN-\gamma. For this purpose, we planned to abrogate the function of cellular wt-PKR gene. Clones of ME180 cells expressing the dominant negative form of PKR were selected in the presence of neomycin. Then these cells were treated with IFN-\gamma and accumulation of IDO protein was determined by Western blot analysis.
