Effects Of Tumor Necrosis Factor-Α On Glucose Metabolism In Cultured Human Muscle Cells From Nondiabetic And Type 2 Diabetic Subjects, Theodore P. Ciaraldi, Leslie Carter, Sunder Mudaliar, Philip A. Kern, Robert R. Henry

The effects of tumor necrosis factor-a (TNFα) on glucose uptake and glycogen synthase (GS) activity were studied in human skeletal muscle cell cultures from nondiabetic and type 2 diabetic subjects. In nondiabetic muscle cells, acute (90-Min) exposure to TNFα (5 ng/ml) stimulated glucose uptake (73 ± 14% increase) to a greater extent than insulin (37 ± 4%; P < 0.02). The acute uptake response to TNFα in diabetic cells (51 ± 6% increase) was also greater than that to insulin (31 ± 3%; P < 0.05). Prolonged (24-h) exposure of nondiabetic muscle cells to TNFα resulted in a further stimulation of uptake (152 ± 31%; P < 0.05), whereas the increase in cells from type 2 diabetics was not significant compared with that in cells receiving acute treatment. After TNFα treatment, the level of glucose transporter-1 protein was elevated in nondiabetic (4.6-fold increase) and type 2 (1.7-fold) cells. Acute TNFα treatment had no effect on the fractional velocity of GS in either nondiabetic or type 2 cells. Prolonged exposure reduced the GS fractional velocity in both nondiabetic and diabetic cells. In summary, both acute and prolonged treatment with TNFα up-regulate glucose uptake activity in cultured human muscle cells, but reduce GS activity. Increased skeletal muscle glucose uptake in conditions of TNFα excess may serve as a compensatory mechanism in the insulin resistance of type 2 diabetes.

Once introduced into a health care environment, this nosocomial pathogen can spread rapidly and as MRSA are resistant to multiple antibiotics, treatment is often difficult. Therefore effective infection control measures are required to prevent cross-infection and further spread of endemic strains.

The adsorption conditions for a range of different immunoreactants to both polystyrene and ultrasmooth silanised silicon surfaces are optimised. The conditions studied were concentration of protein, time and temperature of incubation, pH, molarity and ionic strength of the buffer. The anti-ferritin surface was selected as our model system. This immunosensor was used to develop and evaluate a millispot ELISA for ferritin. The millispot ferritin ELISA developed utilises a sample volume of 6pl and a sample incubation time of 10 minutes. CD44 is a cell surface glycoprotein (mol. wt. =85-95 kDa) which has been implicated in lymphocyte-HEV interactions and the process of lymphocyte homing. CD44 is a complex glycoprotein which undergoes extensive alternative splicing to generate various CD44 isoforms. CD44 functions include the delivery of an activation signal to T-cells, binding to hyaluronate, mediation of lymphocyte homing, and the spread of metastatic cells. Cardiac troponin I (cTnl) has been reported to be a highly specific marker for myocardial injury. In patients suffering from acute coronary syndromes, a sensitive serum marker which is closely related to the extent of myocardial damage which provides prognostic data and which can be measured rapidly is desirable. In this thesis a “sandwich” enzyme immunoassay was developed to measure cTnI. The developed assay was reproducible with the inter assay precision ranging from 4.8-14% and the intra assay precision ranging from 3.1-9.5%. This disorder is characterized by deficits in social interaction skills and communication skills (such as the absence of eye contact or facial expressions). Individuals with AS also insist on routines in behaviors, and pursue a relatively narrow range of interests and activities. Etiology is thought to be multifactoral. Isoxsuprine is reported to be a peripheral vasodilator used in human and veterinary medicine to treat ischaemic vascular disease. In horses, it is generally administered orally to treat navicular disease and other lower limb problems. Caffiene, theophylline and theobromine are naturally occurring members of the methylxanthine family;pentoxfylline, dyphylline and enprofylline are structurally related synthetic pharmaceuticals. Caffiene has predominantly central nervous system effects, theophylline, dyphylline and enprofylline have predominantly bronchodilator effects, while theobromine is associated with diuretic responses. Pentoxfylline is thought to increase red cell deformability and facillitate blood flow through capillary beds. The methylxanthines are not highly potent agents; they are typically administered in gram doses and they tend to have relatively long half-lives. The purpose of this study was to examine the inter-rater and intra-rater reliability of the Dynamic Gait Index (DGI) when used with patients with vestibular disorders. Subjects included 30 patients aged 27-88 years, with vestibular disorders, who were referred for vestibular rehabilitation. Subjects' performance on the DGI was concurrently rated by two physical therapists experienced in vestibular rehabilitation to determine inter-rater reliability. To determine intra-rater reliability each subject repeated the DGI one-hour later. Percent agreement and kappa statistics were calculated for individual DGI items. Kappa statistics for individual items were averaged to yield a composite kappa score of the DGI. …

A questionnaire was designed by the examiner to identify the views and knowledge of Illinois school psychologists and speech-language pathologists regarding selective mutism. Subjects consisted of 119 school psychologists and 106 speech-language pathologists, who completed the questionnaire.

The results of the survey revealed that both Illinois school psychologists and speech-language pathologists viewed themselves as partially responsible for the assessment and treatment of selective mutism, however, both samples indicated low levels of comfort in their ability to successfully provide those services. Additionally, very few of the respondents in either group had received formal training in the area of selective mutism.

Children with Fragile X syndrome (FXS) have significant speech/language difficulties. Additional information is needed regarding the demographics and speech/language characteristics of this population. Participants in this study were parents of children with FXS. Contact persons for three FXS support groups were notified of this study and were asked to provide surveys to parents of children with FXS. The survey was completed by 50 parents of children with FXS who answered questions concerning their child's demographics and speech/language characteristics.
